Aims: Oxidative stress and mitochondrial dysfunction play a role in the process of skin photoaging via activation of matrix metalloproteases (MMPs) and the subsequent degradation of collagen. The activation of nuclear factor E2-related factor 2 (Nrf2), a transcription factor controlling antioxidant and cytoprotective defense systems, might offer a pharmacological approach to prevent skin photoaging. We therefore investigated a pharmacological approach to prevent skin photoaging, and also investigated a protective effect of the novel mitochondria-targeted hydrogen sulfide (H2S) delivery molecul

The syndrome of resistance to thyroid hormone is characterized by elevated serum free thyroid hormones, failure to suppress pituitary thyrotropin secretion, and variable peripheral refractoriness to hormone action. Here we describe a novel leucine to valine mutation in codon 454 (L454V) of the thyroid hormone beta receptor (TR beta) in this disorder, resulting in a mutant receptor with unusual functional properties. Although the mutant protein binds ligand comparably to wild-type receptor and forms homo- and heterodimers on direct repeat, everted repeat, or palindromic thyroid response element

Hypoxia is a key contributor to chemoresistance in glioblastoma (GBM), partly through the activation of nuclear factor erythroid 2-related factor 2 (Nrf2)-dependent cellular stress responses and mitochondrial adaptive programs. The present study investigated Nrf2 signaling in hypoxia-associated resistance to temozolomide (TMZ) in U87MG cells, a glioblastoma cell line, and evaluated whether pharmacological suppression of Nrf2 pathway output with brusatol enhances TMZ sensitivity under hypoxic conditions. U87MG cells were cultured under normoxic or hypoxic conditions (1% O2) and treated with TMZ

Up to 40% of patients with estrogen receptor (ER)-positive breast cancer will develop resistance against the majority of current ER-directed therapies. Resistance can arise through various mechanisms such as increased expression levels of coregulators, and key mutations acquired in the receptor's ligand binding domain rendering it constitutively active. To overcome these resistance mechanisms, we explored targeting the ER Activation Function 2 (AF2) site, which is essential for coactivator binding and activation. Using artificial intelligence and the deep docking methodology, we virtually scre

OBJECTIVES: Cardiac microvascular dysfunction contributes to the pathogenesis of cardiac hypertrophy (CH) and potentially progresses to cardiac failure. Dendrophenol (Den), a naturally derived compound isolated from Dendrobium moschatum, exhibits diverse biological properties, including anti-inflammatory, antioxidant, anti-angiogenic, anti-proliferative, and anti-metastatic effects. This investigation aimed to examine ferroptosis involvement in microvascular dysfunction and assess Den's capacity to suppress ferroptosis while ameliorating pressure overload-induced endothelial damage.

INTRODUCTION: Senescence of activated hepatic stellate cells (HSC) reduces extracellular matrix expression to reverse liver fibrosis. Ferroptosis is closely related to cellular senescence, but its regulatory mechanisms need to be further investigated. The iron ions weakly bound to ferritin in the cell are called labile iron pool (LIP), and together with ferritin, they maintain cellular iron homeostasis and regulate the cell's sensitivity to ferroptosis.
METHODS: We used lipopolysaccharide (LPS) to construct a pathological model group and divided the hepatic stellate cells into a blank group,

High dose and long-term steroid treatment can alter antioxidative ability and decrease the viability and function of osteoblasts, leading to osteoporosis and osteonecrosis. Ferroptosis, a new type of cell death characterized by excessive lipid peroxidation due to the downregulation of GPX4 and system Xc- , is involved in glucocorticoid-induced osteoporosis. Endothelial cell-secreted exosomes (EC-Exos) are important mediators of cell-to-cell communication and are involved in many physiological and pathological processes. However, the effect of EC-Exos on osteoblasts exposed to glucocorticoids h

AIMS: This study investigated whether BACH2 affects bronchopulmonary dysplasia (BPD) through ferritinophagy and elucidated its molecular mechanisms.
RESULTS: JUN was identified as a key gene linking BPD and ferritinophagy, showing elevated expression in preterm neonates with BPD and in hyperoxia-exposed alveolar epithelial cells (MLE12). In the cellular BPD model, knockdown of either JUN or its upstream regulator, BACH2, inhibited hyperoxia-induced nuclear receptor coactivator 4 (NCOA4)-dependent ferritinophagy, accompanied by restored cell viability and mitigated oxidative stress, autophagy

Ferroptosis, an iron-dependent form of regulated cell death, is crucial for skin homeostasis and pathology. The H3K36-specific dimethyltransferase NSD2 is a key epigenetic regulator involved in a spectrum of physiological processes and contributes to the pathogenesis of various human cancers. However, the epigenetic mechanism governing ferroptosis sensitivity in keratinocytes remains poorly understood. This study revealed the role of NSD2 in regulating ferroptosis in HaCaT keratinocytes. We demonstrated that NSD2 deletion significantly attenuated Erastin-induced redox imbalance and reduced the

BACKGROUND: Gastric cancer (GC) remains a major global health challenge, underscoring the urgent need for effective therapeutic strategies. Ferritinophagy-driven ferroptosis has recently emerged as a promising anticancer approach; however, potent pharmacological inducers and their modes of action remain largely unexplored.
PURPOSE: This study aimed to investigate whether Tanshinone I (Tan I), a bioactive diterpenoid quinone, exerts antitumor effects against GC via ferritinophagy-dependent ferroptosis and to elucidate its underlying molecular mechanism.
METHODS: Ferroptosis induction was eval

The massive global output and extensive application of silica nanoparticles (SiNPs) raise great human health concerns, especially in the lungs since the respiratory tract is a major portal for NPs entry. Emerging evidence has revealed that SiNPs cause pulmonary impairments, but the underlying mechanism remains elucidated. In this study, we clarify the role of ferroptosis in SiNPs-induced pulmonary toxicity and elucidate its molecular mechanism. First, the Wistar rats were administered to SiNPs (10 mg/kg·bw) with or without ferrostatin-1 (a well-known inhibitor of ferroptosis, shorted as Fer-1,

Spindle cell rhabdomyosarcomas (SCRMS), recognized by the 2020 World Health Organization Classification of Tumors of Soft Tissue and Bone as a distinct entity, comprise a family of malignant skeletal muscle tumors sharing spindle cell morphology. To date, members of this family include (1) MyoD1-mutated SCRMS/sclerosing rhabdomyosarcomas (RMS), (2) intraosseous SCRMS with FET::TFCP2 or MEIS1::NCOA2 fusions, and (3) infantile/congenital SCRMS harboring NCOA1/2 or VGLL3 rearrangements. A rare, emerging subtype of SCRMS has been reported to harbor recurrent ZFP64::NCOA3 fusions. We studied 14 cas

Alzheimer's disease (AD) is characterized by cognitive decline and the accumulation of amyloid-beta plaques and hyperphosphorylated tau protein. The role of tau lactylation at the K677 site in AD progression is not well understood. This study explores how tau K677 lactylation affects ferritinophagy, ferroptosis, and their functions in an AD mouse model. Results show that mutating the K677 site to R reduces tau lactylation and inhibits ferroptosis by regulating iron metabolism factors like NCOA4 and FTH1.Tau-mutant mice showed improved memory and learning skills compared to wild-type mice. MEIS1-NCOA1/2 fusions are recently described gene rearrangements found in rare sarcomas, mainly involving the genitourinary and gynecologic tracts, with 3 cases reported in the uterine corpus. Although local recurrence was very common, no death has been reported, and some investigators consider these sarcomas low grade. Amplification of genes located at the 12q13-15 locus, especially MDM2 , is the hallmark genetic abnormality in well-differentiated and dedifferentiated liposarcoma of the soft tissue. Some uterine tumors have also been reported to harbor MDM2 amplification, including a proporti

Autosomal dominant polycystic kidney disease (ADPKD) is the most common inherited human renal disorder. Progressive enlargement of the kidneys is due to aberrant proliferation of the cyst epithelial cells, together with accumulation of fluid within the cyst cavities due to transepithelial fluid secretion. Multiple studies have suggested that fluid secretion across ADPKD cyst-lining cells is driven by the transepithelial secretion of chloride, mediated by the apical cystic fibrosis transmembrane conductance regulator chloride channel (CFTR) and specific basolateral transporters. Increased level

In cells infected with HIV type 1 (HIV-1), the integrated viral promoter is present in a chromatin-bound conformation and is transcriptionally silent in the absence of stimulation. The HIV-1 Tat protein binds to a stem-loop structure at the 5' end of viral mRNA and relieves this inhibition by inducing a remodeling of the nucleosome arrangement downstream of the transcription-initiation site. Here we show that Tat performs this activity by recruiting to the viral long terminal repeat (LTR) the transcriptional coactivator p300 and the closely related CREB-binding protein (CBP), having histone ac

INTRODUCTION: Muscle atrophy is a critical health challenge during spaceflight. This study investigated the mechanism of muscle atrophy induced by 21-d hindlimb unloading (HLU) and metabolomic changes in rat gastrocnemius muscle (GM) and plasma.
METHODS: The rat tail-suspended model was used to simulate microgravity. The effects of HLU on GM atrophy were assessed by measuring morphological changes, levels of oxidative stress factors and proinflammatory cytokines, and the expression of key proteins in the insulin-like growth factor 1 receptor/phosphatidylinositol 3-kinase/Ak strain transformin

Obesity arises from impaired energy balance, which is centrally coordinated by leptin through activation of the long form of leptin receptor (Leprb). Obesity causes central leptin resistance. However, whether enhanced peripheral leptin sensitivity could overcome central leptin resistance remains obscure. A peripheral metabolic organ targeted by leptin is the liver, with low Leprb expression. We here show that mice fed a high-fat diet (HFD) and obese patients with hepatosteatosis exhibit increased expression of hepatic helicase with zinc finger 2, a transcriptional coactivator (Helz2), which fu

Bisphenol F (BPF) has rapidly emerged as a primary substitute for bisphenol A (BPA) and a ubiquitous environmental contaminant, yet its reproductive toxicity remains poorly understood, particularly at low doses. Here, we demonstrate that oral exposure to BPF (as low as 250 μg/kg) induces ovarian hypertrophy, disrupted follicular architecture and cumulus-oocyte communication, and reduced serum estradiol and progesterone in mice by triggering ferroptosis. Integrating multiomics approaches, we identified NCOA4-mediated ferritinophagy as the central cellular driver of this process. In granulosa ce
