Proteomic organ-specific signatures, ageing traits, disease risks and genetic architecture in an East Asian population.

BACKGROUND: Protein-based ageing clocks can be used to help identify individuals at high risk of death or morbidity. There is a lack of evidence in non-European populations on clocks derived from combining different proteomic platforms and their relationship with age-related traits, diseases and genetic architecture. METHODS: The prospective China Kadoorie Biobank assayed proteins via Olink and SomaScan in ∼4000 individuals with a mean age of 58 years. We used organ-enriched plasma proteins identified from the Genotype-Tissue Expression Project and trained Light Gradient Boosting models on ch

The immunobiology of pancreatic cancer metastasis: premetastatic niche formation, organ-specific immune landscapes, and therapeutic opportunities.

Pancreatic ductal adenocarcinoma (PDAC) is associated with poor overall survival and is largely refractory to standard therapies. Most patients present with locally advanced or metastatic disease, and those with early-stage tumors amenable to surgical resection face high rates of metastatic relapse. Despite the many successes of immunotherapy in other solid tumors, these approaches have shown only marginal activity in PDAC. In PDAC, metastasis occurs through a coordinated, multistep process characterized by a gradient loss of immune surveillance. This process begins with establishment of a rec

Bionic construction of iron ion-specific porous adsorbents for mitigating iron toxicity across diverse organisms.

Herein, we prepared iron-specific porous adsorbents with six oxygen-formed deformable octahedral adsorption sites. The resulting solids exhibited remarkable selectivity and high-capacity adsorption of Fe3+ ions in simulated TCM solutions (20 ppm). Notably, the materials selectively enrich Fe3+ ions from various plants with high capacity while maintaining the concentrations of other essential metal ions unchanged.

Disentangling polymer confinement from specific-folding interactions reveals the drivers of E. coli chromosome organization.

The three-dimensional organization of the bacterial chromosome is critical for gene regulation. Chromosome conformation capture (Hi-C) has enabled genome-wide mapping of chromosomal folding, yet ensemble-averaged contact maps entangle biologically specific-folding interactions (SFIs) with nonspecific polymer compaction and mask single-cell heterogeneity. Here, we developed a polymer-based simulation framework in E. coli to address these limitations. We found that a null model of 300,000 random polymer configurations recapitulated the global chromosomal organization observed in Hi-C data, estab

Bridging Nano-Interface Interactions and Organ-Specific Toxicity: A Review of Machine Learning for Nanomaterials Risk Assessment.

Risk assessment of engineered nanomaterials (ENMs) is essential for protecting human health and the environment. Traditional hazard assessments rely primarily on in vivo testing, which faces technical challenges in extrapolation validity, ethical dilemmas, and high costs. Machine learning (ML) models offer alternative approaches that are aligned with the 3R principles (Replacement, Reduction, and Refinement) for reducing animal use. ML methods help address the economic, ethical, and temporal limitations of traditional nanotoxicology while advancing mechanistic understanding. This review presen

Organ-Specific Targeted Phenolic Profiling, Antioxidant, and Enzyme-Inhibitory Activities of the Gypsophilous Endemic Hedysarum pestalozzae.

The present study provides an integrative, targeted phenolic-bioactivity mapping of Hedysarum pestalozzae Boiss., an endemic Fabaceae species from gypsum habitats of Türkiye. Methanolic extracts of flowers, leaves, stems, and roots were comparatively evaluated for their phenolic profiles, antioxidant potentials, and enzyme inhibitory effects. Targeted LC-ESI-MS/MS analysis of selected phenolics revealed organ-dependent chemical profiles, with flowers exhibiting the highest accumulation of flavonols-hyperoside, kaempferol, quercetin-and phenolic acids such as protocatechuic acid, together with

Organ-specific and time-dependent oxidative damage induced by chronic copper exposure: modulation by vitamin E.

Chronic exposure to copper ions, Cu(II), induces organ-specific oxidative damage and mitochondrial dysfunction, with distinct temporal and biochemical profiles across tissues. This study evaluated the protective effects of vitamin E (α-tocopherol) against Cu(II)-induced phospholipid and protein oxidation, redox imbalance, and bioenergetic disruption in rats. Cu(II) treatment led to early lipid peroxidation in the brain, heart, and lung, followed by delayed oxidative damage in the liver and kidney. Vitamin E effectively prevented phospholipid oxidation in all organs, but its protection against

Organ-specific equivalent uniform dose constraints and radiobiological parameters for radiation treatment planning of abdominal tumors.

BACKGROUND: Radiation therapy planning (RTP) for abdominal tumors often requires multiple dose-volume constraints for each organ at risk (OAR), which vary with different fractionation schemes. This variability can complicate biologically optimized treatment planning. PURPOSE: This study aims to extract fractionation-independent dose constraints based on the concept of equivalent uniform dose (EUD), utilizing published clinical data for a range of fractionation regimens. The goal is to derive organ-specific radiobiological parameters as well as the EUD for each OAR that can be used in biologic

Structure-biodistribution relationships of chemically and physically modified albumin: a systematic approach to organ-specific drug delivery.

Human serum albumin (HSA) undergoes post-translational modifications that alter its receptor recognition and biodistribution. While HSA receptors such as gp18, gp30, cubilin, and megalin regulate HSA distribution, the relationship between HSA structural changes and organ-specific targeting remains poorly understood. In this study, we systematically prepared five types of modified HSA (acid-treated, base-treated, oxidized, fatty acid-modified, and heat-treated) and characterized their structural alterations using circular dichroism, fluorescence spectroscopy, and site-specific fluorescent probe

Multi-gene biomarkers reveal spatial organization and subpopulation-specific damage response in biliary epithelial cells.

Biliary epithelial cell (BEC) heterogeneity remains challenging to define. Here, we performed scRNA-seq on Sox9EGFP+ liver epithelium from mice subjected to bile duct ligation (BDL) and sham controls. We identified 4 BEC subpopulations in sham controls, each associated with specific enrichment of a minimal, 1-2 gene biomarker ("metagene"). Metagenes were validated by hybridization chain reaction (HCR) FISH, which also facilitated spatial analysis of intrahepatic bile ducts. BECs belonging to a given subpopulation were most likely to have neighbors of the same identity, forming homogenous cellu

MFRP is a molecular hub that organizes the apical membrane of RPE cells by engaging in interactions with specific proteins and lipids.

Membrane frizzled-related protein (MFRP), present in the retinal pigment epithelium (RPE), is an integral membrane protein essential for ocular development and the normal physiology of the retina. Mutations in MFRP are associated with autosomal recessive nonsyndromic nanophthalmos, leading to severe hyperopia and early-onset retinitis pigmentosa. While several preclinical gene-augmentation and gene-editing trials hold promise for future therapies aimed at stopping degeneration and restoring retinal function, the molecular mechanisms involved in MFRP biology are still not well understood. Here,

Cardiopulmonary bypass with deep hypothermic circulatory arrest results in organ-specific transcriptomic responses in pediatric swine.

The organ-level molecular response to cardiac surgery with cardiopulmonary bypass (CPB) remains inadequately understood and may be heterogeneous. Here, we measured organ-specific gene expression in a piglet model of CPB with deep hypothermic circulatory arrest (DHCA). Infant piglets underwent peripheral CPB with 75 min of DHCA and 6 h of critical care after separation from CPB. Mechanically ventilated animals served as controls. Tissue was obtained from the lung, kidney, liver, heart, and ileum. RNA sequencing was performed using NovaSeq 6000 and evaluated via differentially expressed gene (DE

Prevalence of Various Systemic and Organ-Specific Autoimmune Markers in Addison's Disease Patients Compared to Healthy Controls.

Background: Addison's disease (AD) is a rare disorder that often develops in the context of autoimmune polyglandular syndromes. However, the prevalence of rheumatological autoimmune diseases and corresponding autoimmune markers in AD is poorly investigated. Therefore, the present study aims to explore systemic and organ-specific immune markers in a cohort of AD patients from a single tertiary endocrine center. Material and methods: In total, 43 adult AD patients and 31 controls were included in the study. 21-hydroxylase autoantibodies (21OHAb), glutamic acid decarboxylase autoantibodies (GADAb

Control of spatiotemporal activation of organ-specific fibers in the swine vagus nerve by intermittent interferential current stimulation.

Vagus nerve stimulation (VNS) is emerging as potential treatment for several chronic diseases. However, limited control of fiber activation, e.g., to promote desired effects over side effects, restricts clinical translation. Towards that goal, we describe a VNS method consisting of intermittent, interferential sinusoidal current stimulation (i2CS) through multi-contact epineural cuffs. In experiments in anesthetized swine, i2CS elicits nerve potentials and organ responses, from lungs and laryngeal muscles, that are distinct from equivalent non-interferential sinusoidal stimulation. Resection a

Association of the blood levels of specific volatile organic compounds with nonfatal cardio-cerebrovascular events in US adults.

BACKGROUND: Cardio-cerebrovascular diseases constitute a major global public health burden. Volatile organic compounds (VOCs) exposure has become progressively severe, endangering human health and becoming one of the main concerns in environmental pollution. The associations of VOCs exposure with nonfatal cardio-cerebrovascular events have not been identified in observational study with a large sample size, so we aim to examine the association in US adult population. METHODS: Adults aged > 18 years with complete data regarding selected blood levels of VOCs (including benzene, ethylbenzene, o-

Virus-specific T cell therapy to treat refractory viral infections in solid organ transplant recipients.

Solid organ transplant recipients require ongoing immunosuppression to prevent acute rejection, which puts them at risk of opportunistic infections. Viral infections are particularly challenging to prevent and treat as many establish latency and thus cannot be eliminated, whereas targets for small molecule antiviral medications are limited. Resistance to antivirals and unacceptable toxicity also complicate treatment. Virus-specific T cell therapies aim to restore host-specific immunity to opportunistic viruses that is lacking due to ongoing immunosuppressive therapy. This minireview will provi

Cell-type-specific prediction of 3D chromatin organization enables high-throughput in silico genetic screening.

Investigating how chromatin organization determines cell-type-specific gene expression remains challenging. Experimental methods for measuring three-dimensional chromatin organization, such as Hi-C, are costly and have technical limitations, restricting their broad application particularly in high-throughput genetic perturbations. We present C.Origami, a multimodal deep neural network that performs de novo prediction of cell-type-specific chromatin organization using DNA sequence and two cell-type-specific genomic features-CTCF binding and chromatin accessibility. C.Origami enables in silico e

Chronic alcohol intake regulates expression of SARS-CoV2 infection-relevant genes in an organ-specific manner.

BACKGROUND: Chronic alcohol consumption and alcohol use disorder have a tremendous impact on the patient's psychological and physiological health. There is evidence that chronic alcohol consumption influences SARS-CoV2 infection risk, but so far, the molecular mechanism underlying such an effect is unknown. METHODS: We generated the expression data of SARS-CoV2 infection-relevant genes (Ace2, Tmprss2, and Mas) in different organs in rat models of chronic alcohol exposure and alcohol dependence. Ace2 and Tmprss2 represent the virus entry point, whereas Mas activates the anti-inflammatory respo
