Naturally occurring pasteurellosis in laboratory rabbits: chemical and serological studies of whole cells and lipopolysaccharides of Pasteurella multocida.

Whole cells and lipopolysaccharides (LPS) of 10 isolates of Pasteurella multocida from laboratory rabbits were subjected to chemical and serological analysis. LPS of most of these isolates possessed pyrogenic potency comparable to LPS from Salmonella minnesota 9700, although their average ketodeoxyoctonate content was only 18% of that of salmonella. A gel diffusion precipitin test for somatic antigens extracted in a formal-saline solution demonstrated several isolates with three to four somatic antigens, with some variation in the major somatic type from one test to another. Conversely, the us…

Rapid on-site detection of Pasteurella multocida in cattle using an LFS-based RPA-CRISPR/Cas12a assay.

BACKGROUND: Pasteurella multocida is an important bacterial pathogen associated with bovine respiratory disease (BRD), causing substantial economic losses in cattle production worldwide. Rapid field detection of P. multocida remains challenging because conventional diagnostic methods are often time-consuming and require specialized laboratory equipment. This study aimed to establish a rapid visual assay for P. multocida to support field diagnosis and disease management in cattle. METHODS: A recombinase polymerase amplification (RPA)-CRISPR/Cas12a assay targeting the species-specific kmt1 gene…

High prevalence of virulence genes and multi-drug resistance in Pasteurella multocida from goats in Sichuan, China.

Pasteurella multocida is one of the most important pathogens that infect goats, causing serious economic losses in the goat breeding industry. To understand the biological characteristics of P. multocida from goats, a comprehensive characterization of bacteria isolated from 342 nasal swabs and 8 lung tissue samples from goat farms in Sichuan, China, was performed. A total of 34 isolates were assigned to one capsular type, D, and one lipopolysaccharide (LPS) genotype, L3, indicating that the D: L3 was the predominant serotype in goat farms. In the 34 isolates, multiple virulence-related genes w…

Comparative evaluation of PCR and loop-mediated isothermal amplification (LAMP) assays for detecting Pasteurella multocida in poultry.

AIMS: To develop a colourimetric loop-mediated isothermal amplification (LAMP) assay for the detection of Pasteurella multocida in clinical poultry samples and compare the performance of this assay with PCR. A secondary aim was to evaluate a simple DNA extraction method that could enable LAMP-based testing in the field without the need for specialised laboratory equipment. METHODS: Primer sets for both LAMP and PCR were designed to amplify the KMT1 gene of P. multocida. DNA was extracted from 12 P. multocida isolates using a commercial extraction kit, and subjected to analysis using both LAMP…

Experimental pathogenicity and comparative genome analysis of high- and low-virulence strains of rabbit-origin Pasteurella multocida.

Pasteurella multocida, the causative pathogen of rabbit pasteurellosis, causes significant economic losses in the commercial rabbit industry. However, the associated pathogenic mechanism of P. multocida remains unclear. The aim of this study is to compare the genomes and pathogenicity of high- and low-virulence strains of P. multocida to advance the current understanding of rabbit pasteurellosis. The high-virulence strain rapidly proliferates in the lung and spleen of infected mice within approximately 9 h, maintaining a high bacterial load until host death. Meanwhile, the low-virulence strain…

Biofilm formation and avian immune response following experimental acute and chronic avian cholera due to Pasteurella multocida.

Pasteurella multocida is the causative agent of avian cholera, an important economic and ecological disease that can present as a peracute, acute, chronic, or asymptomatic infection. Acute avian cholera is associated with encapsulated P. multocida, while chronic and asymptomatic cases of avian cholera may be associated with capsule-deficient P. multocida isolates. We hypothesize that biofilm formation is also associated with chronic and asymptomatic avian cholera. Experimental infections of chickens with encapsulated, biofilm-deficient P. multocida strain X73, proficient biofilm forming P. mul…

First detection of Pasteurella multocida type B:2 in Hungary associated with systemic pasteurellosis in backyard pigs.

This is the first report of Pasteurella multocida type B in Hungarian pigs. This disease was observed in backyard-raised pigs in three households within a small area. Neither the source of the infection nor the epidemiological connection between any of the premises could be determined. The most consistent lesion was dark red discolouration of the skin of the ventral neck and brisket, with accompanying oedema and haemorrhages. The morbidity was low and lethality relatively high, with three dead (50%) and two euthanised (33%) out of six affected animals. A total of three isolates of P. multocida…

Aggressive Pasteurella multocida rhinosinusitis with orbital extension in a patient with liver cirrhosis: A case report and literature review.

Pasteurella multocida often causes skin and soft tissue infections associated with animal bites and scratches. However, aggressive respiratory tract infections without animal exposure have been reported in immunocompromised hosts. We report the case of a Japanese woman in her early 80 s with liver cirrhosis that developed acute rhinosinusitis caused by P. multocida without animal-related wounds. She presented with swelling and redness of the left eyelid, and subsequently developed decreased vision and impaired eye movement. Imaging studies revealed an ethmoid sinus abscess with a bony defect o…

Comparison of Immunoprotective Efficacy of Six Antigenic Proteins of Pasteurella multocida Serotype a in KM Mice (Mus musculus).

Pasteurella multocida serotype A (P. multocida) is frequently associated with severe respiratory disease in swine (Sus scrofa), highlighting the need for effective preventive strategies. To identify protective antigens suitable for a subunit vaccine targeting porcine P. multocida infection, six recombinant proteins (rAspA, rLolA, rOmpP6, rOppA, rRps6, rSmpA) were expressed in a prokaryotic system, and their efficacy was evaluated in a Mus musculus (Kunming) mouse model. All proteins were purified using His-tag affinity chromatography, and SDS-PAGE analysis confirmed expression with bands at th…

The lipopolysaccharide outer core transferase genes pcgD and hptE contribute differently to the virulence of Pasteurella multocida in ducks.

Fowl cholera caused by Pasteurella multocida exerts a massive economic burden on the poultry industry. Lipopolysaccharide (LPS) is essential for the growth of P. multocida genotype L1 strains in chickens and specific truncations to the full length LPS structure can attenuate bacterial virulence. Here we further dissected the roles of the outer core transferase genes pcgD and hptE in bacterial resistance to duck serum, outer membrane permeability and virulence in ducks. Two P. multocida mutants, ΔpcgD and ΔhptE, were constructed, and silver staining confirmed that they all produced truncated LP…

Antimicrobial resistance of Pasteurella multocida type B isolates associated with acute septicemia in pigs and cattle in Spain.

BACKGROUND: Pasteurella multocida is the etiological agent responsible for several diseases in a wide range of hosts around the world and thus, causes serious economic losses. Acute septicemia associated with capsular type B P. multocida has recently emerged in Europe and continuous outbreaks of these acute processes have been described in Spain since they were first detected in pigs in 2009 and cattle in 2015. The scarcity of studies on the antimicrobial susceptibility of this capsular type of P. multocida and growing concern about the general increase of antimicrobial resistance mean that st…
