Safety and tolerability of zonisamide in elderly patients with epilepsy.

OBJECTIVES: To assess the safety/tolerability of zonisamide in elderly patients. MATERIALS & METHODS: A pooled analysis of clinical study data from elderly (≥65 years) patients receiving add-on/monotherapy zonisamide for partial seizures was compared with pooled adult (18-65 years) study data. Assessments included treatment-emergent adverse events (TEAEs), clinical laboratory parameters and weight change. RESULTS: Data were analyzed from 95 elderly and 1389 adult patients. Incidence of total TEAEs was similar (elderly, 78/95 [82%] vs adult, 1165/1389 [84%]); but lower in elderly versus adult

The efficacy and safety of newer anticonvulsants in patients with dementia.

Anticonvulsants are a class of medications that have received considerable interest as possible treatments in patients with behavioural disturbances in dementia. The role of these medications for such a use remains controversial. The current paper reviews the published evidence surrounding the safety and efficacy (i.e. as a behavioural and cognitive treatment) of newer anticonvulsants in patients with dementia. A MEDLINE, International Pharmaceutical Abstracts, PsycINFO and clinicaltrials.gov search through to December 2011 was conducted for anticonvulsants that have received regulatory approv

Efficacy and safety of linezolid in liver transplant patients.

Bacterial infections are the main cause of death within the first year after liver transplantation, and the increased incidence of multidrug-resistant gram-positive pathogens has created a major challenge in the treatment of these patients. Linezolid, the first US Food & Drug Administration-approved oxazolidinone, offers a valuable novel treatment option for serious gram-positive infections. Linezolid is relatively non-toxic but prolonged treatment with linezolid was associated with thrombocytopenia. Here we report on the experience of linezolid treatment in adult liver transplant patients, wh

[Antibacterial therapy of patients with urolithiasis: efficacy and safety].

Retrospective analysis provided data on the efficacy and safety of antimicrobial therapy in the treatment of urinary tract infections in 21 patients with urolithiasis in an urological unit of a municipal multiprofile hospital. For estimation of the antimicrobial therapy efficacy ATC/DDD method was used (The Anatomical Therapeutic Chemical Classification System with Defined Daily Doses). The analysis showed that the isolates (> 50%) were highly resistant to the majority of the antibiotics prescribed by the physicians for the empirical therapy within 90% DU (Drug Utilization). Moreover, adverse

Safety of rasagiline in elderly patients with Parkinson disease.

The authors examined age effects on adverse events from two randomized, controlled trials of rasagiline, comparing younger (younger than 70 years) and older (70 years and older) subjects. Older patients were more prone to serious adverse effects than younger patients, but there was no statistical interaction between age and rasagiline exposure. This absence of an age-rasagiline interaction suggests that rasagiline does not require special safety precautions for elderly subjects with Parkinson disease.

Thoracentesis in patients with hematologic malignancy: yield and safety.

BACKGROUND: Pleural effusions occur in patients with hematologic malignancies, particularly during periods of hospitalization. Thoracentesis is often performed to diagnose infection and to exclude the presence of complicated parapneumonic effusions. The efficacy and safety of thoracentesis in this setting has not been well-studied. DESIGN: Retrospective chart review of hospitalized patients with hematologic malignancies undergoing thoracentesis. The aim of this study was to assess the role of thoracentesis in establishing a diagnosis of infection in this population and to determine the risk o

The safety and tolerability of donepezil in patients with Alzheimer's disease.

Cholinesterase (ChE) inhibitors, which prevent the hydrolysis of acetylcholine, have been approved for the symptomatic treatment of Alzheimer's disease (AD) for over a decade. However, the first ChE inhibitors were associated with a high incidence of side-effects and general tolerability concerns, including hepatotoxicity. Side-effects associated with increased cholinergic activity, particularly in the gastrointestinal (GI) system, can prevent patients from achieving effective doses of drug. In addition, the advanced age and frail nature of patients with AD mean that poor tolerability is a ser

The safety of UVB phototherapy in patients with HIV infection.

BACKGROUND: In patients with psoriasis and human immunodeficiency virus type 1 (HIV-1) infection, therapeutic options may be limited by their potential immunosuppressive effects. UVB radiation can activate HIV-1 gene expression in transgenic mice and in vitro. It is not known whether this viral activation leads to a clinically significant effect or if these findings can be extrapolated to humans. OBJECTIVE: This study was performed to evaluate the safety of UVB light treatment in HIV-infected persons. METHODS: We prospectively studied the effect of UVB phototherapy on five HIV-infected patie

The safety of dental extractions in patients with hematologic malignancies.

Dental disorders have been recognized as major sources of infection in patients with hematologic malignancies (HM). Management of severe dental infections usually includes dental extractions (DE), but the safety of extractions in patients with HM who are at risk for bleeding, sepsis, and poor wound healing has not been well established. In conjunction with an aggressive program of dental care, 142 DE were performed in 26 patients with acute leukemia, myelodysplastic syndromes, and myeloproliferative disorders. Granulocytopenia (less than 1,000 granulocytes/microL) was present during or within

Evaluating the impact of operating room simulations on communication skills and patient safety (EIORS-CPS).

BACKGROUND: The WHO Surgical Safety Checklist is a cornerstone of perioperative safety, yet its effective implementation hinges on nontechnical skills like communication. High-fidelity simulation (HFS) is a promising tool for training these competencies. This study evaluated the impact of HFS on anesthesiology students' perceptions of patient safety, specifically examining the mediating role of improved communication through the checklist. A convergent parallel mixed-methods study was conducted with 137 fourth-year medical students specializing in anesthesiology. Participants completed a SimMa…
