PFKFB3 controls acinar IP3R-mediated Ca2+ overload to regulate acute pancreatitis severity.

Acute pancreatitis (AP) is among the most common hospital gastrointestinal diagnoses; understanding the mechanisms underlying the severity of AP is critical for development of new treatment options for this disease. Here, we evaluate the biological function of phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3) in AP pathogenesis in 2 independent genetically engineered mouse models of AP. PFKFB3 was elevated in AP and severe AP (SAP), and KO of Pfkfb3 abrogated the severity of alcoholic SAP (FAEE-SAP). Using a combination of genetic, pharmacological, and molecular studies, we defined

Polyglucosan bodies in intramuscular nerve branches are a poor predictor of GBE1 mutation and adult polyglucosan body disease.

INTRODUCTION: Adult polyglucosan body disease (APBD) is associated with formation of polyglucosan bodies in peripheral nerve branches. Some muscle biopsies show these inclusions in intramuscular nerve branches. It has not been established whether the presence of multiple polyglucosan bodies in intramuscular peripheral nerve branches could or should suggest testing for APBD. METHODS: Fifteen muscle biopsies from adults between the ages of 36 and 84 years, all showing polyglucosan bodies in intramuscular peripheral nerve twigs, were tested by sequencing of the GBE1 gene. RESULTS: In 4 patients

Metabolic effects of ethanol on primary cell cultures of rat skeletal muscle.

Individuals who have consumed alcohol chronically accumulate glycogen in their skeletal muscles. Changes in the energy balance caused by alcohol consumption might lead to alcoholic myopathy. Experimental models used in the past, such as with skeletal muscle biopsy samples of alcohol-dependent individuals or in animal models, do not distinguish between direct effects and indirect effects (i.e., alterations to the nervous or endocrine system) of alcohol. In the current study, we evaluated the direct effect of ethanol on skeletal muscle glycogen concentrations and related glycolytic pathways. We

Localization of human liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFKFB1) within a YAC contig in Xp11.21.

6-Phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFK-2/FBPase-2) catalyzes the synthesis and degradation of fructose-2,6-bisphosphate, a potent regulator of glycolysis. Previous studies assigned the gene for human liver PFK-2/FBPase-2 (HGMW-approved symbol PFKFB1) to the X chromosome; however, precise localization remained ambiguous, with the gene variously placed between Xcen-q13, Xq27-q28, and Xp11.22-p11.21. We have localized the gene within a YAC contig clustered around ALAS2 (human erythroid delta-aminolevulinate synthase) in Xp11.21 and have identified eight YACs positive for the ge

Skeletal Muscle and Systemic Adaptations to Repeated Sprint Training with Blood Flow Restriction or in Normobaric Hypoxia.

INTRODUCTION: Repeated sprint training in hypoxia (RSH) or with blood flow restriction (RS-BFR) are known to enhance the repeated-sprint ability. However, studies directly comparing their underlying molecular mechanisms are scarce. METHODS: Here we used a comprehensive approach combining pre- and post-intervention testing including sprint performance tests, neuromuscular assessments, skeletal muscle biopsies for protein levels quantification, and blood samples for systemic metabolites analyses to characterize the specific adaptations induced by RSH and RS-BFR. Recreationally active men perfor

A comparative analysis of serum biochemistry, gene expression, and gut microbiota in domestication-susceptible and domestication-resistant mandarin fish (Siniperca chuatsi).

The feeding behavior of mandarin fish (Siniperca chuatsi) is unique in that it exclusively preys on live fish throughout its life. Feed-based mandarin fish production has become central to aquaculture, yet domestication success remains low. This study examines intrinsic factors underlying poor feed acceptance in mandarin fish, alongside traits of domestication-susceptible individuals (group A) through a comprehensive comparison. The results showed that: compared to domestication-resistant mandarin fish (group B): (1) the glucose content and amylase activity in the serum of group A are signific

Comparative gut microbial enzyme functions in human amoebiasis: Implications for host-microbe interactions and veterinary parasitic models.

BACKGROUND: The intestinal microbiome is a critical component of host defense and metabolic regulation in both humans and animals. Parasitic infections such as amebiasis disrupt microbial enzyme activities, potentially influencing host physiology. AIM: To investigate functional differences in gut microbial enzymes between humans infected with Entameba histolytica and healthy controls and to provide insights relevant to comparative parasitic pathobiology. METHODS: Stool samples collected from patients diagnosed with amebiasis, as well as from healthy control individuals, were analyzed using 1

Mitoquinol improves phagocytosis and glycolysis in ethanol-exposed macrophages via HIF-1α-PFKP axis.

Alcohol use disorder increases sepsis mortality. Acute ethanol exposure impairs pathogen clearance in the macrophages via dampened glycolysis and phagocytosis, exaggerates oxidative stress, and regulates the function of the hypoxia-regulating factor 1α (HIF-1α), a master regulator of glycolysis. Decreased expression of the platelet isoform of phosphofructokinase (PFKP), a key glycolytic enzyme, in ethanol-exposed macrophages, is reported. However, transcriptional regulation of PFKP with ethanol exposure is unclear. We hypothesized that acute ethanol exposure-induced oxidative stress dampens ma

TLR7/8 Agonist R848 Regulates X Sperm Energy Metabolism in Dairy Cows via Inhibiting Glycolysis and Fatty Acid β-Oxidation by LKB1/AMPK Pathway.

Sex control is an efficient biotechnology for improving animal reproductive efficiency which can significantly improve the economic benefits of dairy industry. Flow cytometry is a common method for sex control in dairy cows and can effectively sort X/Y sperm, but its equipment is expensive. Recent studies have shown a new inexpensive potential method that utilizing R848 (a TLR7/8 agonist) to effectively stratify mammalian X/Y sperm by activating X sperm specific proteins. However, the molecular mechanism has not been fully elucidated. In this study, R848 altered X sperm motion trail and strati

Exogenous thyroxine increases cardiac GLUT4 translocation in insulin resistant OLETF rats.

During insulin resistance, the heart undergoes a metabolic shift in which fatty acids (FA) account for roughly about 99% of the ATP production. This metabolic shift is indicative of impaired glucose metabolism. A shift in FA metabolism with impaired glucose tolerance can increase reactive oxygen species (ROS), lipotoxicity, and mitochondrial dysfunction, ultimately leading to cardiomyopathy. Thyroid hormones (TH) may improve the glucose intolerance by increasing glucose reabsorption and metabolism in peripheral tissues, but little is known on its effects on cardiac tissue during insulin resist

The Protective Effect of 11-Keto-β-Boswellic Acid against Diabetic Cardiomyopathy in Rats Entails Activation of AMPK.

This study examined the protective effect of 11-keto-β-boswellic acid (AKBA) against streptozotocin (STZ)-induced diabetic cardiomyopathy (DC) in rats and examined the possible mechanisms of action. Male rats were divided into 5 groups (n = 8/each): (1) control, AKBA (10 mg/kg, orally), STZ (65 mg/kg, i.p.), STZ + AKBA (10 mg/kg, orally), and STZ + AKBA + compound C (CC/an AMPK inhibitor, 0.2 mg/kg, i.p.). AKBA improved the structure and the systolic and diastolic functions of the left ventricles (LVs) of STZ rats. It also attenuated the increase in plasma glucose, plasma insulin, and serum an

Liver X Receptor Agonist Therapy Prevents Diffuse Alveolar Hemorrhage in Murine Lupus by Repolarizing Macrophages.

UNLABELLED: The generation of CD138+ phagocytic macrophages with an alternative (M2) phenotype that clear apoptotic cells from tissues is defective in lupus. Liver X receptor-alpha (LXRα) is an oxysterol-regulated transcription factor that promotes reverse cholesterol transport and alternative (M2) macrophage activation. Conversely, hypoxia-inducible factor 1-α (HIF1α) promotes classical (M1) macrophage activation. The objective of this study was to see if lupus can be treated by enhancing the generation of M2-like macrophages using LXR agonists. Peritoneal macrophages from pristane-treated mi

Reproducibility and absolute quantification of muscle glycogen in patients with glycogen storage disease by 13C NMR spectroscopy at 7 Tesla.

Carbon-13 magnetic resonance spectroscopy (13C MRS) offers a noninvasive method to assess glycogen levels in skeletal muscle and to identify excess glycogen accumulation in patients with glycogen storage disease (GSD). Despite the clinical potential of the method, it is currently not widely used for diagnosis or for follow-up of treatment. While it is possible to perform acceptable 13C MRS at lower fields, the low natural abundance of 13C and the inherently low signal-to-noise ratio of 13C MRS makes it desirable to utilize the advantage of increased signal strength offered by ultra-high fields

Effects of prepubertal-onset exercise on body weight changes up to middle age in rats.

The present study was conducted to examine whether prepubertal-onset exercise might help adults maintain long-term body weight (BW) reduction and increased energy metabolism after the cessation of exercise. Furthermore, the effects of the exercise regimen were compared with those of food restriction. Twenty-three male obese-diabetic [Otsuka Long-Evans Tokushima Fatty (OLETF)] rats were randomly assigned to prepubertal-onset exercise (Childhood-Ex), food restriction (Childhood-Diet), and sedentary control (OLETF-Sed) groups. Childhood-Ex rats exercised voluntarily every day using a rotating whe

Sympathetic activation in exercise is not dependent on muscle acidosis. Direct evidence from studies in metabolic myopathies.

Muscle acidosis has been implicated as a major determinant of reflex sympathetic activation during exercise. To test this hypothesis we studied sympathetic exercise responses in metabolic myopathies in which muscle acidosis is impaired or augmented during exercise. As an index of reflex sympathetic activation to muscle, microneurographic measurements of muscle sympathetic nerve activity (MSNA) were obtained from the peroneal nerve. MSNA was measured during static handgrip exercise at 30% of maximal voluntary contraction force to exhaustion in patients in whom exercise-induced muscle acidosis i

Action of the diabetogenic drug streptozotocin on glycolytic and glycogenolytic metabolism in adult rat brain cortex and hippocampus.

In sporadic Alzheimer's disease (AD), a number of metabolic alterations to the brain have been observed soon after the onset of the initial clinical symptoms. In particular, impairments of glucose utilization and related metabolic pathways are prominent and well-established findings in incipient AD, resembling metabolic abnormalities such as have been found in noninsulin-dependent diabetes mellitus. To mimic these abnormalities, we administered an intracerebroventricular (icv) injection of streptozotocin (STZ) to rats and studied the effects of glucose and glycogen metabolism in the cerebral c
