Nonheme iron in sickle erythrocyte membranes: association with phospholipids and potential role in lipid peroxidation.

Previous studies documented the abnormal association of heme and heme proteins with the sickle RBC membrane. We have now examined RBC ghosts … D, which suggests that it represents an abnormal association of iron with polar head groups of aminophospholipids. The biologic feasibility of such a chelate was demonstrated by using an admixture of iron with model liposomes. Even in the presence of tenfold excess adenosine diphosphate, iron partitions readily into phosphatidylserine liposomes; there is no det …

Alteration of membrane phospholipids in hypertrophied rat salivary glands induced by chronic administration of isoproterenol.

Hypertrophy of parotid and submandibular glands, and elevated phospholipid content per gland in both were induced. However, phospholipid content per tissue weight increased only in the parotid gland. Sublingual gland phospholipids were unaffected with regard to the gland weight and phospholipid content; phospholipid class compositions were altered in each of the three glands, typically an increase of phosphatidylcholine and decreases of sphingomyelin and phosphatidic acid. Concerning fatty-acyl groups in phospholipid, prominent increase of octadecadienoic acid was observed in total and individ…

Quantitative loss of individual eicosapentaenoyl-relative to arachidonoyl-containing phospholipids in thrombin-stimulated human platelets.

The thrombin-dependent losses of eicosapentaenoate (EPA) from the various phospholipids of platelets derived from human subjects ingesting a fish lipid concentrate (MaxEPA) were quantitatively assessed and studied in relation to arachidonate (AA). The net loss of AA and EPA from the total phospholipid, phosphatidylcholine (PC) + phosphatidylethanolamine (PE) + phosphatidylserine (PS) + phosphatidylinositol (PI) (loss from phosphatidylinositol minus accumulated phosphatidate), amounted to 44.4 and 7.3 nmol/2 x 10(9) platelets (mean values, n = 4 subjects), respectively, in response to thrombin …

Cone-shaped lipids increase the susceptibility of phospholipids in bilayers to the action of phospholipases.

Addition of cardiolipin or diacylglycerol to dispersions of phosphatidylcholine greatly increased hydrolysis by snake venom or pancreatic phospholipase A2, as well as by a microbial phospholipase. Monogalactosyl diglyceride which, like cardiolipin and diacylglycerol, will form nonbilayer hexagonal II structures also caused an increase in the breakdown of phosphatidylcholine. Addition of digalactosyl diglyceride, a bilayer lipid from the same source, had a much smaller effect on the three phospholipases, indicating that stimulation by the nonbilayer lipids was not due to their fatty acid compos

Recent Advancements in Phospholipid Metabolic Labeling Using Clickable Probes.

Herein, we present a perspective on recent advancements in the field of metabolic labeling as a method for interrogating phospholipids in terms of their biosynthesis, trafficking, and localization. Phospholipids are crucial for establishing cell morphology and signaling processes mediated by interactions between nearly all relevant cellular substrates. Bioorthogonal chemistry has provided a convenient platform in which clickable probes that mimic biologically active precursors are deployed to generate labeled versions of phospholipids in cells, enabling the study of downstream lipid activities…

Phospholipid bicelles revisited - Examining their properties for topical drug delivery.

While phospholipid bicelles have long been known as membrane models, the current work focuses on their potential application as topical drug delivery systems. For this, a systematic investigation of bicelles was performed to define the conditions required to maintain the small size and disk-shaped morphology that are favorable for skin penetrating properties. Bicelles consisting of 1,2- dimyristoyl-sn-glycero-3-phosphocholine (DMPC, C14) and 1,2-dihexanoyl-sn-glycero-3-PC (C6) were used as starting point. PCs bearing acyl chains with either 6 or 7 carbon atoms (C6, C7) could both be mixed with

The mTOR-Dop1a-Agpat2 axis regulates nuclear phospholipid homeostasis.

The molecular mechanisms regulating the phospholipid (PL) metabolism in the nucleus remain to be elucidated. Here, we describe the role of Dop1a in controlling PL abundance in nuclear membranes (NMs) under the control of mTOR signaling. A shortage of lysophosphatidic acid (LPA) triggers the rapid localization of Dop1a to the nuclear pore complexes (NPCs), where Dop1a suppresses PL synthesis by binding to AGPAT2 (1-acylglycerol-3-phosphate O-acyltransferase 2), which is also localized at the NPCs. Loss of Dop1a results in elevated PL production, which leads to the formation of nuclear lipid dro

Bis(monoacylglycero)phosphate: an endolysosomal phospholipid with antioxidant and antiatherogenic properties.

Bis(monoacylglycero)phosphate (BMP) is a unique anionic glycerophospholipid almost exclusively localized to late endosomes/lysosomes (LE/LY), where it accounts for up to 70 % of the total phospholipid content of intraluminal vesicle membranes. It displays distinctive structural features, including an unusual sn-1:sn-1' stereochemistry and enrichment in long-chain unsaturated fatty acids such as oleic acid and docosahexaenoic acid (DHA). These properties underlie its roles in vesicular trafficking, lysosomal hydrolase activation, and cholesterol transport. Beyond these functions, BMP may act as

Characterization and response to exercise training of HDL-specific phospholipid efflux.

The HDL-specific phospholipid efflux (HDL-SPE) assay is a novel cell-free measure of HDL function that is inversely associated with coronary artery disease. However, the effect of exercise training on HDL-SPE is unknown. The purpose of this study was to examine the effect of exercise training on HDL-SPE in a large, diverse cohort free of overt disease. Clinical and functional measures of HDL were taken before and after 20 weeks of endurance exercise training in 508 participants from the HERITAGE Family Study. Associations of HDL-SPE with HDL-related traits were examined using Pearson's correla

From Liposomes to Virosomes: Evolution of Phospholipid Nanocarriers in Drug Delivery.

Phospholipid-based nanocarriers are an adaptable and chemically tunable class of drug delivery systems that self-assemble into bilayered vesicles due to the amphiphilic nature of phospholipids. Phospholipid-based nanocarriers can encapsulate both hydrophilic and hydrophobic drugs through non-covalent interactions and modulation of lipid phase behavior. This review explores the molecular and supramolecular principles governing the formation, stability, and function of key phospholipid-derived nanocarriers, including liposomes, transferosomes, ethosomes, invasomes, phytosomes, pharmacosomes, and…

An evolutionary analysis of lysophospholipid acyltransferases generating membrane phospholipid diversity.

All living organisms are enclosed by biological membranes composed primarily of phospholipids. The diversification of membrane lipids has shaped key membrane functions, including signaling, membrane protein regulation, and immune responses, and may also have driven organismal evolution. In humans, this diversity is mainly generated by lysophospholipid acyltransferases (LPLATs), yet the evolutionary distribution and diversity of LPLATs across organisms remain poorly understood, particularly in pathogens. Here, we systematically identified candidate LPLAT proteins belonging to two major families…

Fabrication of phospholipid polymer-modified alginate hydrogels for bioartificial pancreas.

The bioartificial pancreas, composed of a semi-permeable hydrogel encapsulating insulin-secreting cells, has attracted attention as a treatment for type 1 diabetes. In this study, we developed phospholipid polymer-modified alginate hydrogel beads that encapsulated spheroids of the pancreatic beta cell line MIN6. The hydrogel beads were composed of methacrylated alginic acid, which enabled both ionic and covalent cross-linking, resulting in a hydrogel that was more stable than conventional alginate hydrogels. Furthermore, modification of biocompatible 2-methacryloyloxyethyl phosphorylcholine (M

Cell membranes sustain phospholipid imbalance via cholesterol asymmetry.

Membranes are molecular interfaces that compartmentalize cells to control the flow of nutrients and information. These functions are facilitated by diverse collections of lipids, nearly all of which are distributed asymmetrically between the two bilayer leaflets. Most models of biomembrane structure and function include the implicit assumption that these leaflets have similar abundances of phospholipids. Here, we show that this assumption is generally invalid and investigate the consequences of lipid abundance imbalances in mammalian plasma membranes (PMs). Using lipidomics, we report that cyt…

Phospholipid flippase ATP11A brokers uterine epithelial integrity and function.

Uterine adaptations driven by the steroid hormones estrogen and progesterone are pivotal for embryo implantation and, ultimately, for a successful pregnancy. Here, we show in mice that genetic ablation of the membrane lipid flippase Atp11a causes severe deficits in this hormonal response and profound defects in the morphological organization and transcriptional profile of the uterine epithelial compartment where Atp11a is expressed. Atp11a-null uterine epithelial cells lack tight junctions, and the luminal epithelium exhibits profound disruptions to cellular morphology. Interestingly, the spec

PNPLA6 regulates retinal homeostasis by choline through phospholipid turnover.

Although mutations in human patatin-like phospholipase PNPLA6 are associated with hereditary retinal degenerative diseases, its mechanistic action in the retina is poorly understood. Here, we uncover the molecular mechanism by which PNPLA6 dysfunction disturbs retinal homeostasis and visual function. PNPLA6, by acting as a phospholipase B, regulates choline mobilization from phosphatidylcholine and subsequent choline turnover for phosphatidylcholine regeneration in retinal pigment epithelial cells. PNPLA6-driven choline is supplied from retinal pigment epithelial cells to adjacent photorecepto

Phospholipid scramblase 1: a frontline defense against viral infections.

Phospholipid scramblase 1 (PLSCR1) is the most studied member of the phospholipid scramblase protein family. Its main function is to catalyze calcium (Ca2+)-dependent, ATP-independent, bidirectional and non-specific translocation of phospholipids between inner and outer leaflets of plasma membrane. Additionally, PLSCR1 is identified as an interferon-stimulated gene (ISG) with antiviral activities, and its expression can be highly induced by all types of interferons in various viral infections. Indeed, numerous studies have reported the direct antiviral activities of PLSCR1 through interrupting…

Damage of the Phospholipid Bilayer by Aβ42 at Physiologically Relevant Peptide Concentrations.

Amyloid β (Aβ) aggregates are implicated in the pathology of several neurodegenerative diseases such as Alzheimer's disease, Huntington's disease, and Parkinson's disease, and damage to membranes is considered one of the pathology-related effects of Aβ. Experiments in vitro indicate that Aβ can damage these membranes; however, such experiments were performed at Aβ concentrations in the micromolar range, several orders above the physiologically relevant conditions. Our studies with Aβ42 in the low nanomolar concentrations did not reveal any damage to the supported lipid bilayer, questioning thi
