Controlled environment agriculture (CEA) is expanding rapidly as a climate-resilient approach for year-round lettuce production near consumption centers. However, despite its supply chain advantages and high productivity, the economic viability and scalability of CEA-grown lettuce remain constrained by the continued reliance on cultivars developed for variable open-field conditions rather than stable, high-input, and space-limited environments of indoor production systems. This mismatch between existing germplasm and CEA-specific production requirements represents a critical gap that has not b

Thaxtomin A (TA) is a phytotoxin secreted by Streptomyces scabies that causes common scab in potatoes. However, the mechanism of potato proteomic changes in response to TA is barely known. In this study, the proteomic changes in potato leaves treated with TA were determined using the Isobaric Tags for Relative and Absolute Quantitation (iTRAQ) technique. A total of 693 proteins were considered as differentially expressed proteins (DEPs) following a comparison of leaves treated with TA and sterile water (as a control). Among the identified DEPs, 460 and 233 were upregulated and downregulated, r

Plants have evolved various defense mechanisms against biotic stress. The most common mechanism involves the production of metabolites that act as defense compounds. Bacterial angular leaf spot disease (Xanthomonas fragariae) of the strawberry (Fragaria x ananassa) has become increasingly destructive to strawberry leaves and plant production. In this study, we examined metabolic changes associated with the establishment of long-term bacterial disease stress using UPLC-QTOF mass spectrometry. Infected leaves showed decreased levels of gallic acid derivatives and ellagitannins, which are related

The growth-defence trade-offs pose a major challenge to breeding high-yield and disease-resistant crops. Aquaporins are membrane channels that facilitate the transport of water and other small compounds, therefore regulating the growth-defence trade-offs. However, the molecular mechanism that governs the function of aquaporins in the trade-offs remains unclear. Here, we report that Triticum aestivum TaPIP1;6 and TaPIP2;10, aquaporins of the plasma membrane intrinsic protein (PIP) family, function as dual substrate channels that concurrently enhance plant growth and resistance to powdery mildew

Historic breeding program end-use quality data was used to perform genomic selection in two broad frameworks, incorporating fixed effect loci and genotype by environment interactions. Exceptional end-use quality in hexaploid bread wheat is an important factor for selling wheat in the global marketplace. However, many end-use quality traits are time-consuming and expensive to test, meaning their measurement is often delayed until late-stage trials of a breeding program. Genomic selection (GS) is one tool that has been shown to allow for earlier generation improvement of many traits in wheat bre

Genomic selection (GS) is a powerful tool for accelerating genetic gain in potato (Solanum tuberosum L.) breeding, particularly for complex traits. In this study, three practical aspects of GS implementation in a potato breeding program were examined. First, the predictive ability of GS models was evaluated for three key traits (total yield, marketable yield, and specific gravity) using two elite potato populations with shared ancestry, tested across seven location-year environments. Two cross-validation strategies were used to reflect practical breeding scenarios: predicting unphenotyped line

Ascochyta blight (AB), caused by the necrotrophic fungus [Ascochyta rabiei (Pass.) Labr.], is one of the most destructive diseases of chickpea (Cicer arietinum L.), causing yield losses of up to 100% under favorable conditions. The pathogen possesses a heterothallic mating system with two mating-type idiomorphs, MAT1-1 and MAT1-2, which contribute to high genetic diversity and the frequent breakdown of host resistance. This review summarizes current knowledge of AB biology, epidemiology, and management, highlighting recent advances in molecular diagnostics, host-pathogen interactions, and popu

Although upland cotton (Gossypium hirsutism L.) originated in the tropics, this early maturity cotton can be planted as far north as 46°N in China due to the accumulation of numerous phenotypic and physiological adaptations during domestication. However, how the genome of early maturity cotton has been altered by strong human selection remains largely unknown. Herein, we report a cotton genome variation map generated by the resequencing of 436 cotton accessions. Whole-genome scans for sweep regions identified 357 putative selection sweeps covering 4.94% (112 Mb) of the upland cotton genome, in

Altering gene dosage through variation in gene copy number is a powerful approach to addressing questions regarding gene regulation, quantitative trait loci, and heterosis, but one that is not easily applied to sexually transmitted species. Elite poplar (Populus spp) varieties are created through interspecific hybridization, followed by clonal propagation. Altered gene dosage relationships are believed to contribute to hybrid performance. Clonal propagation allows for replication and maintenance of meiotically unstable ploidy or structural variants and provides an alternative approach to inves

BACKGROUND: Faba bean cultivation in northern Europe is challenged by high variability in yield performance due to strong genotype × environment interaction (GEI). Appropriate GEI modeling and interpretation is essential for both cultivar recommendation and breeding for Nordic conditions.
METHODS: Grain yield data from fifteen spring faba bean cultivars evaluated in eleven Norwegian environments during four growing seasons (2022-2025) were analyzed using the Additive Main Effects and Multiplicative Interaction (AMMI) model. Climatic drivers of GEI were investigated using factorial regression,

Genomic data from diverse germplasm used for application in targeted breeding germplasm. Six SNPs identified that can characterize all haplotypes present at SD1 locus in diverse rice. Three alleles of the SD1 gene identified in US rice germplasm including two semidwarf alleles. Two SNPs identified and validated that differentiate the SD1 allele present in US germplasm. KASP assays designed for both SNPs for use in high-throughput breeding applications. Plant height is an important target in US rice (Oryza sativa L.) breeding programs and the large effect of the sd1 semidwarf gene makes it a su

Iron deficiency is endemic in much of the world, and food system-based approaches to eradication may be viable with new plant breeding approaches to increase the micronutrient content in staple crops. It is thought that conventional plant breeding approaches provide varieties of rice that have 400-500% higher iron contents than varieties commonly consumed in much of Asia. The efficacy of consuming high-iron rice was tested during a 9-mo feeding trial with a double-blind dietary intervention in 192 religious sisters living in 10 convents around metro Manila, the Philippines. Subjects were rando…

Adzuki bean (Vigna angularis) is an important medicinal and edible legume with high nutritional and functional value. However, a systematic multi-dimensional quality evaluation system for mainstream varieties remains insufficient, restricting targeted breeding and industrial diversified utilization of adzuki bean. This study comprehensively assessed the agronomic, nutritional, processing and functional traits of 22 dominant Chinese adzuki bean varieties to screen promising candidate germplasms for specific applications. Multiple indices covering agronomic traits, mineral nutrition, seed color,

Genome-wide association studies (GWASs) have become a central tool in plant genomics, providing high-resolution access to the genetic architecture of complex traits in cereal crops. This review observes the application of GWASs to biotic and abiotic stress responses and nutritional quality in rice (Oryza sativa), wheat (Triticum aestivum), maize (Zea mays), and barley (Hordeum vulgare). However, conventional SNP-based GWASs may overlook structural and presence-absence variation, and significant associations do not by themselves identify the causal gene or variant. Recent methodological develop

Horsegram (Macrotyloma uniflorum), a member of the Fabaceae family, is a nutritious and low-cost legume used for both grain and fodder. This study employed a genome-wide association approach to identify loci linked to key agronomic traits in horsegram. Plant height, seed size, and shoot fresh weight were evaluated in a panel of 96 diverse genotypes. GBS was performed using the Illumina HiSeq platform, yielding 20,241 high-quality SNPs after filtering at a 5% minor allele frequency. Population structure analysis classified genotypes into three admixed subgroups. Phenotyping was conducted over t

Salt stress is a major environmental factor limiting crop growth and productivity. We found a long-chain acyl-CoA synthetase (LACS) gene ZmLACS9 that was associated with salt tolerance traits in maize (Zea mays). Mutation of ZmLACS9 impaired plant growth, while overexpression of ZmLACS9 improved plant growth vigour under salt stress conditions. Metabolome and transcriptome analyses indicated that ZmLACS9 may regulate the expression of lipid trafficking-related genes, thereby influencing the concentrations of galactolipids and phospholipid in the photosynthetic membrane under salt stress. Under

Hybrid seed failure arising from wide crosses between plant species is a recurring obstacle in plant breeding, impeding the transfer of desirable traits. This postzygotic reproductive barrier primarily occurs in the endosperm, a tissue that nourishes the embryo and functions similarly to the placenta in mammals. We found that incompatible seeds show a loss of DNA methylation and chromatin condensation in the endosperm, similar to seeds lacking maternal RNA polymerase IV activity. This similarity is linked to a decrease in small interfering RNAs in the endosperm (sirenRNAs), maternal RNA polyme…

Soil is essential for sustaining life on land. Plant roots play a crucial role in stabilising soil and minimising erosion, although these mechanisms are still not completely understood. Consequently, identifying and breeding for plant traits to enhance erosion resistance is challenging. Root hair mutants in Arabidopsis thaliana were studied using three different quantitative methods to isolate their effect on root-soil cohesion. We present compelling evidence that micro-scale interactions of root hairs with surrounding soil increase soil cohesion and reduce erosion. Arabidopsis seedlings with

Cassava (Manihot esculenta) is an important staple crop, especially in the arid tropics. Because roots of commercial cassava cultivars contain a limited amount of provitamin A carotenoids, both conventional breeding and genetic modification are being applied to increase their production and accumulation to fight vitamin A deficiency disorders. We show here that an allelic polymorphism in one of the two expressed phytoene synthase (PSY) genes is capable of enhancing the flux of carbon through carotenogenesis, thus leading to the accumulation of colored provitamin A carotenoids in storage roots.
