Gene-for-gene disease resistance without the hypersensitive response in Arabidopsis dnd1 mutant.

The cell death response known as the hypersensitive response (HR) is a central feature of gene-for-gene plant disease resistance. A mutant line of Arabidopsis thaliana was identified in which effective gene-for-gene resistance occurs despite the virtual absence of HR cell death. Plants mutated at the DND1 locus are defective in HR cell death but retain characteristic responses to avirulent Pseudomonas syringae such as induction of pathogenesis-related gene expression and strong restriction of pathogen growth. Mutant dnd1 plants also exhibit enhanced resistance against a broad spectrum of virul

Simple Whole-Mount Staining Protocol of F-Actin for Studies of the Female Gametophyte in Agavoideae and Other Crassinucellate Ovules.

During plant sexual reproduction, F-actin takes part in the elongation of the pollen tube and the movement of sperm cells along with it. Moreover, F-actin is involved in the transport of sperm cells throughout the embryo sac when double fertilization occurs. Different techniques for analysis of F-actin in plant cells have been developed: from classical actin-immunolocalization in fixed tissues to genetically tagged actin with fluorescent proteins for live imaging of cells. Despite the implementation of live cell imaging tools, fixed plant tissue methods for cytoskeletal studies remain an essen…

Recent insights into air space formation in plant shoots.

An adequate supply of oxygen and carbon dioxide is essential for plant survival. Although plant cell walls are somewhat porous, their hydrated nature hampers gas diffusion. Furthermore, the cuticular wax coating the epidermal layer of aerial tissues strongly inhibits gas exchange. Because plants lack specialized systems that bind and transport gases, gases must be directly delivered to the target cells. This necessitates the establishment of effective gas transport pathways connecting stomata to the target cells. However, our understanding of this process remains fragmented. Recent studies hav

In vitro antiplasmodial activity of selected plants from the Colombian North Coast with low cytotoxicity.

BACKGROUND: Plants are an important option in the treatment of malaria, especially in endemic regions, and are a less expensive and more accessible alternative with a lower risk of toxicity. Colombia has a great diversity of plants, and evaluation of natural extracts could result in the discovery of new compounds for the development of antimalarial drugs. The purpose of this work was to evaluate the in vitro antiplasmodial activity and the cytotoxicity of plant extracts from the Colombian North Coast against Plasmodium falciparum. MATERIALS AND METHODS: The antiplasmodial activity of 12 plant

Pharmacological Strategies for Manipulating Plant Ca2+ Signalling.

Calcium is one of the most pleiotropic second messengers in all living organisms. However, signalling specificity is encoded via spatio-temporally regulated signatures that act with surgical precision to elicit highly specific cellular responses. How this is brought about remains a big challenge in the plant field, in part due to a lack of specific tools to manipulate/interrogate the plant Ca2+ toolkit. In many cases, researchers resort to tools that were optimized in animal cells. However, the obviously large evolutionary distance between plants and animals implies that there is a good chance

DEAP1 encodes a fasciclin-like arabinogalactan protein required for male fertility in rice.

Arabinogalactan proteins (AGPs) are widely distributed in plant cells. Fasciclin-like AGPs (FLAs) belong to a subclass of AGPs that play important roles in plant growth and development. However, little is known about the biological functions of rice FLA. Herein, we report the identification of a male-sterile mutant of DEFECTIVE EXINE AND APERTURE PATTERNING1 (DEAP1) in rice. The deap1 mutant anthers produced aberrant pollen grains with defective exine formation and a flattened aperture annulus and exhibited slightly delayed tapetum degradation. DEAP1 encodes a plasma membrane-associated member…

Plant MIR167e-5p Inhibits Enterocyte Proliferation by Targeting β-Catenin.

MicroRNAs (miRNAs) are important negative regulators of genes involved in physiological and pathological processes in plants and animals. It is worth exploring whether plant miRNAs play a cross-kingdom regulatory role in animals. Herein, we found that plant MIR167e-5p regulates the proliferation of enterocytes in vitro. A porcine jejunum epithelial cell line (IPEC-J2) and a human colon carcinoma cell line (Caco-2) were treated with 0, 10, 20, and 40 pmol of synthetic 2'-O-methylated plant MIR167e-5p, followed by a treatment with 20 pmol of MIR167e-5p for 0, 24, 48, and 72 h. Suppression of microRNA159 impacts multiple agronomic traits in rice (Oryza sativa L.).

BACKGROUND: microRNAs (miRNAs) are important regulators in plant growth and development. miR159 is a conserved miRNA among different plant species and has various functions in plants. Studies on miR159 are mostly done on model plant, Arabidopsis thaliana. In rice, studies on miR159 were either based upon genome-wide expression analyses focused upon responses to different nitrogen forms and abiotic stress or upon phenotypic studies of transgenic plants overexpressing its precursor. STTM (Short Tandem Target Mimic) is an effective tool to block the activity of endogenous mature miRNA activity in

Multiple Subcutaneous Nodules Caused by Plant Fungus Thyridium flavostromatum in an Immunocompromised Patient.

Thyridium flavostromatum is a newly discovered species of plant fungus identified from the host plant Phyllostachys pubescens. It is a type of ascomycetes that has yellowish stromata and gets the name "flavostromatum." Immunocompromised patients are infected with rare and opportunistic fungal infections. We here present a case of multiple subcutaneous tender nodules caused by T. flavostromatum in a man with rheumatoid arthritis on immunosuppressants. Aspirate from the nodules revealed yeast-like cells with chlamydospores that were not identified in the matrix-assisted laser desorption ionizati

Antifungal plant defensins: increased insight in their mode of action as a basis for their use to combat fungal infections.

Plant defensins are small, cationic peptides with a highly conserved 3D structure. They have been studied extensively in the past decades. Various biological activities have been attributed to plant defensins, such as anti-insect and antimicrobial activities, but they are also known to affect ion channels and display antitumor activity. This review focuses on the structure, biological activity and antifungal mode of action of some well-characterized plant defensins, with particular attention to their fungal membrane target(s), their induced cell death mechanisms as well as their antibiofilm ac

Whiskers-mediated maize transformation.

There has been rapid progress in recent years in extending gene transfer capabilities to include plant species that fall outside the normal host range of Agrobacterium. Methods that allow direct DNA delivery into plant cells have contributed significantly to this expanded capability. Whiskers treatment is one means of delivering macromolecules, including DNA, to plant cells. Using relatively simple equipment and inexpensive materials, whiskers-mediated transformation of maize is possible. A critical prerequisite, however, is the establishment and maintenance of embryogenic tissue cultures as a…

Plant-Based Diet and Erectile Dysfunction: A Narrative Review.

Evidence shows that the consumption of plant foods, particularly those in their whole form (fruits, vegetables, whole grains, nuts, seeds, and legumes from which no edible part has been removed), improves cardiometabolic risk factors and is associated with reduced risk of cardiovascular diseases (CVDs), diabetes, cancer, chronic kidney disease and mortality compared with animal (meat, fish, eggs and dairy) and nonwhole plant foods (sugar-sweetened beverages, refined grains, etc.). Erectile dysfunction (ED) is considered a strong predictor of CVD. The underlying defect in arteriogenic ED is end

Morphological plasticity of endophytic Chitinophaga pinensis.

Environmental changes, whether due to climate change or human influences, compromise the resilience of plants to biotic and abiotic stresses, such as pathogens, drought and heat. Plant microbiota are known to promote plant resilience. To be able to harness the power of the plant microbiome we need to identify microbiota with health-promoting properties. Recent studies have demonstrated that the bacterium Chitinophaga pinensis enhances plant health and increases resistance to fungal infections. Here, we show that C. pinensis exhibits an unusually high morphological plasticity, switching between

Chromium toxicity and tolerance mechanisms in plants through cross-talk of secondary messengers: An overview of pathways and mechanisms.

Environmental sources of chromium (Cr) such as solid waste, battery chemicals, industrial /waste, automotive exhaust emissions, mineral mining, fertilizers, and pesticides, have detrimental effects on plants. An excessive amount of Cr exposure can lead to toxic accumulations in human, animal, and plant tissues. In plants, diverse signaling molecules like hydrogen sulfide (H2S) and nitric oxide (NO) play multiple roles during Cr stress. Consequently, the molecular mechanisms of Cr toxicity in plants, such as metal binding, modifying enzyme activity, and damaging cells are examined by several st

Plant virus-derived siRNAs stay a step ahead of viral short-range spread.

Whether mobile virus-derived siRNAs can pre-immunize uninfected plant tissues has been a long-standing assumption in plant biology. In this issue, Albertini et al. demonstrate that they indeed establish an AGO1-dependent immune zone of adjacent uninfected short-range cells, and this mobile pre-immunization is impacted by viral suppressors of RNA silencing.

Co-extraction of Small RNAs and Total RNA from Arbuscular Mycorrhizal Roots Using Laser Microdissection.

Understanding gene expression at the cellular level is essential for dissecting plant-microbe interactions. Laser microdissection (LMD) is a powerful approach for isolating specific plant cell types without the need for molecular markers, enabling the recovery of nucleic acids from defined cellular populations. In plant systems, LMD has been widely applied to study cell-type-specific transcriptomes, including those involved in arbuscular mycorrhizal (AM) symbiosis, where cellular heterogeneity limits bulk tissue analyses. Recent evidence highlights the role of small RNAs (sRNAs) as key regulat
