Comparative transcriptome analysis reveals higher expression of stress and defense responsive genes in dwarf soybeans obtained from the crossing of G. max and G. soja.

BACKGROUND: Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. Dwarfism in plants prevents lodging and therefore it's a desired trait in crops. OBJECTIVE: To find differentially expressed genes to classify and understand the regulation of genes related to plant growth in mutant dwarf soybeans, which appeared in the F5 generation. METHODS: We obtained a few segregated dwarf soybeans in the populations derived from the crossing of Glycine max var. Peking and Glycine soja var. IT182936 in an F5 RIL population. These dwarf so

Rice OsVAMP714, a membrane-trafficking protein localized to the chloroplast and vacuolar membrane, is involved in resistance to rice blast disease.

Membrane trafficking plays pivotal roles in many cellular processes including plant immunity. Here, we report the characterization of OsVAMP714, an intracellular SNARE protein, focusing on its role in resistance to rice blast disease caused by the fungal pathogen Magnaporthe oryzae. Disease resistance tests using OsVAMP714 knockdown and overexpressing rice plants demonstrated the involvement of OsVAMP714 in blast resistance. The overexpression of OsVAMP7111, whose product is highly homologous to OsVAMP714, did not enhance blast resistance to rice, implying a potential specificity of OsVAMP714 …

Disruption of the primary salicylic acid hydroxylases in rice enhances broad-spectrum resistance against pathogens.

Salicylic acid (SA) is a crucial hormone involved in plant immunity. Rice (Oryza sativa) maintains high SA levels that are not induced by pathogens. However, the roles of SA in rice immunity and yield remain largely unknown. Here, we identified SA 5-hydroxylases 1 (OsS5H1) and 2 (OsS5H2) as the primary enzymes engaged in catalysing SA to 2,5-dihydroxybenzoic acid (2,5-DHBA) in rice. SA levels were significantly increased in the oss5h mutants, while they were dramatically decreased in the OsS5H1 and OsS5H2 overexpression lines. The mutants were resistant, whereas the overexpression lines were s…

Enzymatic regulation of methionine/homocysteine levels by the folate pathway dictates broad-spectrum defense in Arabidopsis.

Folate-dependent one‑carbon (1C) metabolism is vital for plant development and stress responses, but its specific role in immunity remains unclear. Arabidopsis Methylenetetrahydrofolate reductase 2 (MTHFR2), a prominent enzyme in 1C metabolism, is crucial in regulating methionine biosynthesis and controlling broad-spectrum immunity. Here, we show by HRMS profiling that loss-of-function mutation in MTHFR2 disturbs methionine and homocysteine concentrations, which lead to differential immune responses against a broad-spectrum of pathogens, including Magnaporthe oryzae, Rhizoctonia solani, and Ps

Effects of Atmospheric CO2 Levels on the Susceptibility of Maize to Diverse Pathogens.

Rising atmospheric CO2 has significant implications for crop productivity and food security. Based on studies in C3 plants, elevated CO2 (eCO2) can shape plant-pathogen interactions, although the outcomes are often variable. The question of how eCO2 influences immunity and disease development in C4 plants, such as the globally important cereal crop maize (Zea mays L.), has not been systematically examined. We challenged maize plants grown under ambient CO2 (aCO2, 420 ppm) and eCO2 (550 ppm) with bacterial, viral, fungal, and oomycete pathogens. Plants grown in eCO2 were more susceptible to sug

Plant-based vaccines against viral hepatitis: A panoptic review.

The traditional vaccines against hepatitis have been instrumental in reducing the incidence of some types of viral hepatitis; however, the need for cost-effective, easily distributable, and needle-free vaccine alternatives has led to the exploration of plant-based vaccines. Plant-based techniques offer a promising avenue for producing viral hepatitis vaccines due to their low-cost cultivation, scalability, and the potential for oral administration. This review highlights the successful expression of hepatitis B surface antigens in plants and the subsequent formation of virus-like particles, wh

Discovery of Transfer Factors in Plant-Derived Proteins and an In Vitro Assessment of Their Immunological Activities.

Repeated exposure to pathogens leads to evolutionary selection of adaptive traits. Many species transfer immunological memory to their offspring to counteract future immune challenges. Transfer factors such as those found in the colostrum are among the many mechanisms where transfer of immunologic memory from one generation to the next can be achieved for an enhanced immune response. Here, a library of 100 plants with high protein contents was screened to find plant-based proteins that behave like a transfer factor moiety to boost human immunity. Aqueous extracts from candidate plants were tes

Plant-derived, stress-responsive microRNA mimics reduce powdery mildew infections of barley.

Barley powdery mildew, caused by Blumeria hordei, leads to economic losses worldwide. Here, in silico analyses and proof-of-concept experiments were conducted to determine if specific plant-derived synthetic microRNAs (miRNAs) may be considered as a new direction for the control of barley powdery mildew. Previous studies have demonstrated that barley miRNAs regulate plant immunity against B. hordei. Based on the predicted targets of stress-responsive miRNAs in barley, four miRNAs that are upregulated during biotic stresses were selected and applied through Spray-Induced Gene Silencing (SIGS). …

Plant oxalate oxidases: key enzymes in redox and stress regulation.

Oxalate oxidases (OXOs) are integral to plant physiology, facilitating the degradation of oxalate (oxalic acid) into hydrogen peroxide (H2O2) and carbon dioxide. OXOs are considered a key source of H2O2 in plant cells, especially under oxidative stress, which activates diverse physiological roles, including cell wall fortification, redox signaling, and modulation of ion homeostasis. As a result, OXOs significantly influence embryo viability, cell wall stiffening, apoplastic redox activity, and defense, especially under abiotic stresses such as drought, salinity, and temperature. Additionally,

Overexpressing the N-terminus of CATALASE2 enhances plant jasmonic acid biosynthesis and resistance to necrotrophic pathogen Botrytis cinerea B05.10.

Salicylic acid (SA) acts antagonistically to jasmonic acid (JA) in plant immunity. We previously reported that CATALASE2 (CAT2) promotes JA-biosynthetic acyl-CoA oxidase (ACX) activity to enhance plant resistance to necrotrophic Botrytis cinerea, and SA represses JA biosynthesis through inhibiting CAT2 activity, while the underlying mechanism remains to be further elucidated. Here, we report that the truncated CAT2 N-terminus (CAT2-N) interacts with and promotes ACX2/3, and CAT2-N-overexpressing plants have increased JA accumulation and enhanced resistance to B. cinerea B05.10, but compromised…

Immunogenic properties of plant-derived recombinant smallpox vaccine candidate pB5.

The extracellular virion membrane protein B5 is a potent inducer of immune responses capable of protecting mice and primates against poxvirus infections. Here, we examined the antibody response induced in mice immunized intramuscularly (i.m.) or intranasally (i.n.) with plant-derived B5 (pB5) accompanied or not with plant total soluble protein (TSP) at various concentrations. Increasing amounts of TSP inhibited the pB5-specific response in both i.m.- and i.n.-immunized mice, with more dramatic effects in the latter. pB5 administered to mucosal surfaces induced specific IgG and IgA responses, w

Fungal endophytes relieve the growth-defence trade-off of their plant hosts through the production of bioactive alkaloids.

A central paradigm in plant biology is that there is a trade-off between growth and defence. We propose that Epichloë fungal endophytes reli … xpression of photosynthesis-related genes (e.g., RBCS1), and had mixed effects on the expression of plant immunity-related genes (e.g., PR-1). GA treatment increased concentrations of ABA, salicylic acid, and jasmonic acid, and did not affect auxin concentrations. Additionally, soil derived from GA-treated plants showed increased abundance of putative bacterial …

Rice stripe virus NS2 and NS3 proteins jointly stabilize OsIAA25 to inhibit auxin-mediated immunity in rice.

Rice stripe virus (RSV) causes substantial rice (Oryza sativa) yield losses and poses a serious threat to agricultural production. Auxin is a pivotal hormone in orchestrating the antagonism between plant growth and defense. Conversely, pathogen manipulation of auxin signaling promotes disease and inhibits plant growth, but how this hormone influences immunity is not fully understood. Here, we report that RSV infection results in dwarfism and short crown roots in rice and reduces the sensitivity of rice plants to NAA. The RSV-encoded nonstructural protein 3 (NS3) interacts with Aux/IAA represso

Studying Plant MIF/D-DT-Like Genes and Proteins (MDLs).

Human macrophage migration inhibitory factor (MIF) is an inflammatory cytokine with chemokine-like characteristics and an upstream regulator of host innate immunity. It is a critical mediator of a variety of human diseases, such as acute and chronic inflammatory diseases, autoimmunity, atherosclerosis, and cancer. MIF is an atypical chemokine that not only signals through its cognate receptor CD74, but also interacts with the classical chemokine receptors CXCR2 and CXCR4. MIF and its homolog D-dopachrome tautomerase (D-DT)/MIF-2 are structurally unique proteins that are conserved across kingdo

Phylogeny and evolution of plant macrophage migration inhibitory factor/D-dopachrome tautomerase-like proteins.

BACKGROUND: The human (Homo sapiens) chemokine-like protein macrophage migration inhibitory factor (HsMIF) is a pivotal mediator of inflammatory, infectious and immune diseases including septic shock, colitis, malaria, rheumatoid arthritis, and atherosclerosis, as well as tumorigenesis. HsMIF has been found to exhibit several sequential and three-dimensional sequence motifs that in addition to its receptor binding sites include catalytic sites for oxidoreductase and tautomerase activity, which provide this 12.5 kDa protein with a remarkable functional complexity. A human MIF paralog, D-dopachr
