Development of one-tube multiplex polymerase chain reaction (PCR) for detecting Mycobacterium bovis.

A multiplex PCR (m-PCR) with primers targeting the 16S rRNA, Rv3873 and a 12.7-kb fragment in the genomes of a Mycobacterium tuberculosis complex was designed for the differential diagnosis of M. tuberculosis, M. bovis, M. bovis BCG and non-tuberculosis Mycobacterium (NTM). The specificity of this assay was 100%, and the detection limit was 15 pg of genomic DNA. Of the 206 blinded clinical samples, the detection rate of M. bovis infection by m-PCR was lower than that of the interferon gamma (IFN-γ) release assay; however, the false-positive rate by the tuberculin skin test and false-negative s

Multiplex polymerase chain reaction assay developed to diagnose adult bacterial meningitis in Taiwan.

Acute bacterial meningitis causes high morbidity and mortality; the associated clinical symptoms often are insensitive or non-specific; and the pathogenic bacteria are geographically diverse. Clinical diagnosis requires a rapid and accurate methodology. This study aimed to develop a new multiplex polymerase chain reaction (mPCR) assay to detect simultaneously six major bacteria that cause adult bacterial meningitis in Taiwan: Klebsiella pneumoniae, Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus aureus, Escherichia coli, and Acinetobacter baumannii. Species-specific primers fo…

Factors influencing polymerase chain reaction outcomes in patients with clinically suspected ocular tuberculosis.

BACKGROUND: Polymerase chain reaction (PCR) assay can be a useful method for definitive diagnosis in paucibacillary infections such as ocular tuberculosis (TB). In this study, we have evaluated factors affecting PCR outcomes in patients with clinically suspected ocular TB. Patients with clinically suspected ocular TB were investigated by PCR of aqueous or vitreous samples. Three control groups were also tested: group 1 included culture-proven non-tuberculous endophthalmitis, group 2 culture-negative non-tuberculous endophthalmitis, and group 3 patients undergoing surgery for uncomplicated cata…

Polymerase chain reaction-based screening for the ceftriaxone-resistant Neisseria gonorrhoeae F89 strain.

Emergence and spread of Neisseria gonorrhoeae resistant to extended spectrum cephalosporins is a major problem threatening treatment of gonorrhoea and is further highlighted by the recent report of a second ceftriaxone-resistant N. gonorrhoeae strain (F89) in Europe, initially observed in France and subsequently identified in Spain. N. gonorrhoeae antimicrobial resistance (AMR) surveillance has acquired new importance and molecular tools have the potential to enhance bacterial culture-based methods. In this study, we established a polymerase chain reaction (PCR) protocol for direct detection o…

Q fever cholecystitis in an unvaccinated butcher diagnosed by gallbladder polymerase chain reaction.

Acalculous cholecystitis has been described in association with Q fever. We describe the first known case of Q fever cholecystitis diagnosed by polymerase chain reaction on gallbladder tissue. The presence of Coxiella burnetii genome in the diseased gallbladder tissue argues for direct involvement of the organism in the pathogenesis of Q fever cholecystitis.

Real-time polymerase chain reaction for detection of Isospora belli in stool samples.

A real-time polymerase chain reaction (PCR) assay targeting the internal transcribed spacer 2 region of the ribosomal RNA gene was developed for the detection of Isospora belli DNA in fecal samples, including an internal control to detect inhibition during the amplification process. The assay was performed on species-specific DNA controls (n = 27) and a range of positive (n = 21) and negative (n = 120) stool samples, and achieved 100% specificity and sensitivity. The simple fecal sample collection procedure, the high-throughput potential, and the possibility of quantification makes the I. bell…

[Polymerase chain reaction evaluation for canine visceral leishmaniasis diagnosis in dog blood samples].

Canine and human visceral leishmaniasis is endemic in several States of Brazil, and it is associated with infected dogs and the presence of the vector. Aiming at using polymerase chain reaction as a diagnostic tool in dogs, we amplified a 120bp fragment from kDNA of Leishmania spp. by PCR in blood samples. The lower detection limit observed was 0.1 parasites per 500microL of blood, which is a highly satisfactory result. On the other hand, PCR evaluation in 166 blood samples of dogs from Poxoréo, MS, Brazil, resulted in 55% sensitivity and 66.3% specificity, considering indirect imunnofluoresce…

BK virus DNA detection by real-time polymerase chain reaction in clinical specimens.

The BK polyomavirus (BKV) is widespread in the general population. In transplant recipients, the patients' weakened immune response may encourage reactivation of latent infection, leading to BKV-related diseases. Rapid and quantitative detection might help to delineate viral reactivation patterns and could thus play an important role in their clinical management. In our study we developed an "in-house" quantitative real-time PCR to detect BKV DNA. The effectiveness of this assay was evaluated by a retrospective analysis of 118 plasma specimens from 22 bone marrow transplant (BMT) recipients an

Clinical utility of polymerase chain reaction for diagnosis of smear-negative pleural tuberculosis.

BACKGROUND: Polymerase chain reaction (PCR) is a molecular biology technique which can detect Mycobacterium tuberculosis (TB) genome in pleural fluid; however, the results are variable. METHODS: Two hundred and twelve pleural fluid specimens suspected to be possibly associated with tuberculosis with negative acid-fast smears were sent to our laboratory to test for the presence of M. tuberculosis DNA using nested PCR, the target for the amplification being a segment of IS6110 in the genome of M. tuberculosis. The final diagnosis of TB pleurisy was based on combining clinical judgment with radi…

Evaluation of polymerase chain reaction for rapid diagnosis of clinically suspected tuberculous pleurisy.

Pleural effusion is one of the commonest presentations of tuberculosis, the clinical manifestations being typically abrupt resembling bacterial pneumonia. Since delayed hypersensitivity is the underlying immune response, bacterial load is very low. Owing to these facts, tuberculous pleurisy as an extra-pulmonary disease poses a diagnostic dilemma. The conventional bacteriological methods rarely detect Mycobacterium tuberculosis in pleural fluid and are of limited use in diagnosis of tuberculous pleurisy. We evaluated the efficacy of polymerase chain reaction (PCR) in the diagnosis of tuberculo…

Identification of Pentatrichomonas hominis in feline fecal samples by polymerase chain reaction assay.

Pentatrichomonas hominis is considered to be a commensal protozoan of the vertebrate digestive tract. On the basis of light microscopic examination of feces, some investigators presumptively identified P. hominis as a causative agent of feline diarrhea. However, molecular identification of P. hominis infection in the cat has not been reported. Another trichomonad, Tritrichomonas foetus, is recognized as an intestinal pathogen in cats and often presumptively diagnosed on the basis of the presence of trichomonads in diarrheic feces. It is of importance to determine if cats are natural hosts for
