Genetic and inflammatory interplay between IL1B (-511C/T) polymorphism and diabetic nephropathy in type 2 diabetes.

BACKGROUND: Inflammatory cytokines, particularly interleukin-1β (IL-1β), have a main role in diabetic nephropathy (DN) pathogenesis. Genetic variations within the IL1B promoter region may modulate cytokine expression and affect vulnerability to renal injury in type 2 diabetes mellitus (T2DM). We aimed to assess the correlation between the IL1B (-511C/T;rs16944) polymorphism, IL-1β serum levels, and renal function among Egyptian cases with T2DM. METHODS: This study was conducted on 150 subjects, who were divided into 50 T2DM cases with DN, 50 T2DM patients without DN, and 50 healthy control su

Insights into AQP5 Polymorphism and Genetic Associations in Molar Incisor Hypomineralization and Dental Fluorosis.

INTRODUCTION: While fluoride exposure is a well-established factor in the development of dental fluorosis (DF), individual variability in susceptibility suggests that additional factors may also contribute to its manifestation. The etiology of molar incisor hypomineralization (MIH), in turn, is multifactorial and remains not fully understood. The aim of this study was to investigate the association between MIH, DF, caries, and polymorphisms in the AQP5, MMP2, and COMT genes in children exposed to high fluoride levels in drinking water. METHODS: A total of 268 schoolchildren aged 6-12 years fr

Genetic insights into vitamin D deficiency: a case-control study of GC and CYP24A1 gene polymorphism.

Despite the sunny climate, vitamin D deficiency is highly prevalent in several parts of the world. Several risk factors are associated with VD deficiency, including single nucleotide polymorphism and post-translational modifications in its transport protein, known as vitamin D binding protein (DBP) or GC and CYP24A1, a protein associated with its degradation. Our study explores the impact of rs4588 and rs7041 in the GC gene, along with CYP24A1 rs4809960 and rs2585428, on serum vitamin D and the risk of vitamin D insufficiency. This study enrolled 114 healthy controls and 239 vitamin D-deficien

Research on the Genetic Polymorphism and Function of inlA with Premature Stop Codons in Listeria monocytogenes.

Listeria monocytogenes is a Gram-positive bacterial species that causes listeriosis, a major foodborne disease worldwide. The virulence factor inlA facilitates the invasion of L. monocytogenes into intestinal epithelial cells expressing E-cadherin receptors. Naturally occurring premature stop codon (PMSC) mutations in inlA have been shown to result in the production of truncated proteins associated with attenuated virulence. Moreover, different L. monocytogenes strains contain distinct inlA variants. In this study, we first characterized inlA in 546 L. monocytogenes strains isolated from vario

Potential genetic polymorphism of matrix metalloproteinase (MMP)-9 in Iranian migraine patients with Toxoplasma gondii infection.

Toxoplasma gondii is an intracellular protozoan parasite that causes neuroinflammation in the brain and a constant need for peripheral leukocyte migration. Matrix metalloproteinase 9 (MMP-9) can play a major role in this neuroinflammation and be implicated in some neurological disorders, such as migraines. Therefore, the genetic polymorphism evaluation of MMP-9 in migraine patients with T. gondii infection was performed. One hundred fourteen migraine patients and 114 healthy controls were evaluated for the presence of anti-Toxoplasma IgG antibodies. Seventy-two migraine patients and 40 healthy

Genetic Background Strongly Influences the Impact of Carrying the Thr92Ala-DIO2 Polymorphism in the Male Mouse.

About half of the world population carries at least one allele of the Ala92-DIO2, which slows down the activity of the type 2 deiodinase (D2), the enzyme that activates T4 to T3. Carrying the Ala92-DIO2 allele has been associated with increased body mass index and insulin resistance, but this has not been reproduced in all populations. To test if the genetic background affects the impact of this polymorphism, here we studied the genetically distant C57Bl/6J (B6) and FVB/N (FVB) mice carrying the Ala92-Dio2 allele as compared to control mice carrying the Thr92-Dio2 allele. Whereas B6-Ala92-Dio2

Insertion/deletion polymorphism for genetic background and forensic performance exploration of the Sui group from Guizhou.

Insertion/deletion polymorphisms (InDels) as ideal genetic markers for forensic genetics are appreciated by scholars both nationally and internationally because they integrated the favorable features of single nucleotide polymorphisms (SNPs) and short tandem repeats (STRs). Nevertheless, with the limited identification efficiency of InDels, the multiplex amplification systems of InDels might just be applied as the supplementary methods in paternity testing with respect to commonly used STRs. In the current research, we successfully genotyped 105 unrelated individuals from the Guizhou Sui popul

Increase in the genetic polymorphism of varicella-zoster virus after passaging in in vitro cell culture.

Primary infections with the varicella-zoster virus (VZV) result in varicella, while latent reactivation leads to herpes zoster. Both varicella and zoster can be prevented by live attenuated vaccines. There have been reports suggesting that both clinical VZV strains and those in vaccine preparations are genetically polymorphic, containing mixtures of both wild-type and vaccine-type sequences at certain vaccine-specific sites. In this study, the genetic polymorphism of the VZV genome was examined by analyzing the frequencies of minor alleles at each nucleotide position. Next-generation sequencin

Genetic Polymorphism in the RYR1 C6487T Is Associated with Severity of Hypospadias in Chinese Han Children.

OBJECTIVE: Hypospadias is a common congenital malformation of the male external genitalia. Most cases have an unknown etiology, which is probably a mix of monogenic and multifactorial forms, implicating both genetic and environmental factors. Ryanodine receptor 1 (RYR1) mutations are a common cause of congenital diseases associated with both dominant and recessive inheritance in humans. Herein, we evaluated the correlations of RYR1 C6487T polymorphism with the risk and severity of hypospadias. METHODS: 263 congenital hypospadias children and 312 healthy children were recruited.
