Study of the tools available in biological dosimetry to estimate the dose in cases of accidental complex overexposure to ionizing radiation: the Lilo accident.

PURPOSE: To compare the efficiency of different cytogenetic tools in estimating the doses received by four people involved in the Lilo accident and to monitor the dose estimate over 4.5 years. MATERIALS AND METHODS: Several young Georgian frontier guards handled at least one of the 12 Caesium sources found in a former Russian military camp. Overexposure lasted from July 1996 to May 1997. The Institute for Radiological Protection and Nuclear Safety (IRSN) obtained blood samples taken at several intervals post-exposure from the four most highly-exposed people. Dose estimation was performed usin

Temperature and the formation of radiation-induced chromosome aberrations. II. The temperature dependence of lesion repair and lesion interaction.

Dicentric chromosome aberration yields have been measured after single-exposure and split-dose irradiations of human lymphocytes with 150 kV X-rays. Various temperature programmes between 4 and 37 degrees C were applied before, during and after irradiations and in the radiation-free interval. It was found that chromatin lesion repair was completely suppressed at 21 degrees C and below, whereas lesion formation is reduced only below 17 degrees C. The interaction between repairable lesions which leads to exchange-type aberrations is also suppressed by low temperatures. Hypothermic suppression of

Incorporation of (3H)thymidine stimulated by ultraviolet radiation into human fibroblast cultures.

We studied DNA repair synthesis after ultraviolet irradiation in human fibroblasts cultured in vitro by measuring the ultraviolet-stimulated incorporation of [3H]thymidine into cells in which the semi-conservative DNA replication was inhibited by hydroxyurea. Experiments performed with five fibroblasts lines derived from healthy donors showed a relatively fast initial process ( that is completed within 1 h for 100 erg/mm2 and within 2 h for 500 erg/mm2) and a subsequent slower process, evident between 2 and 6 h after irradiation. The repair capacity of normal cells is expressed by the differen

Lack of synergistic effect between X-ray and UV irradiation on the frequency of chromosome aberrations in PHA-stimulated human lymphocytes in the G1 stage.

PHA-stimulated human lymphocytes in the G1 stage were irradiated with UV radiation and X-rays, and the cells were analyzed for chromosomal aberrations in the first mitotic division. The frequency of dicentric chromosomes after single X-irradiation in the G1 stage was about twice the yield in the G0 stage. No increase in the yield of dicentrics was observed after combined irradiation with UV and X-rays. This is contrary to the finding for G0 lymphocytes, where a 2-fold increase of chromosome aberrations was observed. UV irradiation of G1 lymphocytes induced chromatid-type aberrations whereas no

Radiation-induced Non-targeted Effect and Carcinogenesis; Implications in Clinical Radiotherapy.

Bystander or non-targeted effect is known to be an interesting phenomenon in radiobiology. The genetic consequences of bystander effect on non-irradiated cells have shown that this phenomenon can be considered as one of the most important factors involved in secondary cancer after exposure to ionizing radiation. Every year, millions of people around the world undergo radiotherapy in order to cure different types of cancers. The most crucial aim of radiotherapy is to improve treatment efficiency by reducing early and late effects of exposure to clinical doses of radiation. Secondary cancer indu

ANALYSIS OF A GENE CONTROLLING CELL DIVISION AND SENSITIVITY TO RADIATION IN ESCHERICHIA COLI.

Adler, Howard I. (Oak Ridge National Laboratory, Oak Ridge, Tenn.), and Alice A. Hardigree. Analysis of a genetic locus controlling cell division and sensitivity to radiation in Escherichia coli. J. Bacteriol. 87:720-726. 1964.-Radiation sensitivity in Escherichia coli is under control of genes that are transferred during conjugation between donor and recipient strains. Conjugation experiments establish that one of these genes occupies a locus on the E. coli K-12 linkage map between the genes controlling ability to utilize lactose and galactose. It affects sensitivity to both ionizing and ultr

Type I IFN protects cancer cells from CD8+ T cell-mediated cytotoxicity after radiation.

Treatment of tumors with ionizing radiation stimulates an antitumor immune response partly dependent on induction of IFNs. These IFNs directly enhance dendritic cell and CD8+ T cell activity. Here we show that resistance to an effective antitumor immune response is also a result of IFN signaling in a different cellular compartment of the tumor, the cancer cells themselves. We abolished type I IFN signaling in cancer cells by genetic elimination of its receptor, IFNAR1. Pronounced immune responses were provoked after ionizing radiation of tumors from 4 mouse cancer cell lines with Ifnar1 knocko

Apoptosis and the adaptive response in human lymphocytes.

PURPOSE: To determine whether the sensitivity of human lymphocytes for apoptosis induced by either a membrane oxidizing agent or a DNA damaging agent is modified by an adaptive response. MATERIALS AND METHODS: Peripheral blood lymphocytes from normal human donors were exposed to low doses of the DNA damaging agent gamma-radiation, or the membrane oxidizing agent t-butyl hydroperoxide (t-BuOOH), incubated for various times and then tested for their sensitivity to induction of apoptosis by a subsequent exposure to a high dose of either agent. Apoptosis was measured using a fluorescent assay of

PATTERN OF ACUTE LYMPHOBLASTIC LEUKEMIA IN THE SPECTRUM OF RADIATION-ASSOCIATED HEMATOLOGICAL DISEASES (literature review).

The review presents literature data on the incidence and mechanisms of acute lymphoblastic leukemia (ALL) inadults and children, taking into account the role of genetic changes, chromosomal aberrations, and epigenetic disorders that occur pre and postnatally as a result of exposure to ionizing radiation in a wide range of doses. The possibility of developing a set of preventive measures for the occurrence of radiationassociated ALL is discussed basedon the determination of individual variability of patients and the role of the disease's modifying risk factors.

Comparison of radiation-induced aberration frequencies in chromosomes 1 and 2 of two human donors.

PURPOSE: The purpose of this study was to analyse donor and time dependent variations in the frequencies of radiation-induced aberrations in chromosomes 1 and 2. MATERIALS AND METHODS: Human lymphocytes from two donors were irradiated with 1 and 2 Gy of X-rays. Chromosomal aberrations were scored in chromosomes 1 and 2 painted with different fluorochromes and in Giemsa stained cells. Two time displaced experiments were performed with lymphocytes of each donor. RESULTS: In cells of both donors chromosome 1 was generally more frequently involved in translocations than chromosome 2. This result

Identification of radiation-induced aberrant hypomethylation in colon cancer.

BACKGROUND: Exposure to ionizing radiation (IR) results in the simultaneous activation or downregulation of multiple signaling pathways that play critical roles in cell type-specific control of survival or death. IR is a well-known genotoxic agent and human carcinogen that induces cellular damage through direct and indirect mechanisms. However, its impact on epigenetic mechanisms has not been elucidated, and more specifically, little information is available regarding genome-wide DNA methylation changes in cancer cells after IR exposure. Recently, genome-wide DNA methylation profiling technolo

A comparison of the relative activities of 8 radiosensitizers in the SOS chromotest.

Misonidazole, and RSU 1069 and 6 of its analogues are all reported to show increased cytotoxicity towards hypoxic cells compared to oxic cells. DNA is considered to be the target through which these drugs exert their cytotoxic activity. Therefore we monitored induction of the SOS response in uvrABC excinuclease proficient and deficient strains of E. coli, under oxic and hypoxic conditions, as an indirect method of assessing the activity of these drugs towards DNA in a biological system. This was done using the SOS chromotest which utilizes E. coli strains which possess a sfiA::lacZ fusion allo

[Reparative processes of damaged chromosomes in non-stimulated human peripheral blood lymphocytes by the fractionated irradiation method and temperature variation].

The fractionation experiments were carried out on resting lymphocytes. Non-stimulated lymphocytes were X-irradiated at the total dose of 200 r separated into two equal parts by either 5-hour or 20-hour intervals. The whole blood samples were kept during the intervals between the exposure at the temperature of 20degrees C or 37degrees C. All the cultures were made after the last radiation exposure at 37degrees C. Dicentrics and centric rings were scored. It is shown that a fractionation effect takes place in resting lymlf-dose at 37degrees C and is absent at the temperature of 20degrees C. It i

Modern trends in human genetics.

Every physician is deeply concerned with the tremendous advances in genetics during the last decade. Through 'gene surgery' DNA transplantation on both somatic and germinal tissue is feasible. Thus, genetic determinism is no longer ineluctable. Tissue culture is a tool to avoid cellular degeneration and achieve cellular immortality.Precise chromosome identification allows one to follow transmission from generation to generation, as well as human gene mapping. Through clinical and especially biochemical methods, genetic entities are dissected more accurately. Ecological genetics studies the ris

Basal cell carcinoma arising in outdoor workers versus indoor workers: a retrospective study.

Basal cell carcinoma (BCC) is the most prevalent malignancy in white individuals and continues to be a serious health problem. Individuals who have sustained exposure to UV radiation are at the highest risk for developing BCC. The aim of this study was to compare the features of BCC in outdoor workers (OWs) with a history of occupational exposure to UV radiation versus indoor workers (IWs). We found that OWs are more likely to develop nodular BCC with no increased risk for superficial BCC. The age of onset of BCC was older in OWs than in IWs. Truncal BCC was more common in IWs, which may sugge

Mesothelioma following Wilms' tumor in childhood.

A high percentage of children with Wilms' tumor are cured with multimodal treatment. A small percentage of these children will develop second tumors, perhaps related to a genetic predisposition to neoplasia or possibly secondary to the treatment utilized for Wilms' tumor. Malignant mesothelioma has been associated with contact with asbestos but has also been reported after radiation exposure. Two patients are reported who developed malignant mesothelioma of the pleura after treatment for Wilms' tumor in childhood. Both received orthovoltage radiation; one patient also received triethylenemelam
