Predicting HLA class II antigen presentation through integrated deep learning.

Accurate prediction of antigen presentation by human leukocyte antigen (HLA) class II molecules would be valuable for vaccine development and cancer immunotherapies. Current computational methods trained on in vitro binding data are limited by insufficient training data and algorithmic constraints. Here we describe MARIA (major histocompatibility complex analysis with recurrent integrated architecture; https://maria.stanford.edu/ ), a multimodal recurrent neural network for predicting the likelihood of antigen presentation from a gene of interest in the context of specific HLA class II alleles

FetNet: a recurrent convolutional network for occlusion identification in fetoscopic videos.

PURPOSE: Fetoscopic laser photocoagulation is a minimally invasive surgery for the treatment of twin-to-twin transfusion syndrome (TTTS). By using a lens/fibre-optic scope, inserted into the amniotic cavity, the abnormal placental vascular anastomoses are identified and ablated to regulate blood flow to both fetuses. Limited field-of-view, occlusions due to fetus presence and low visibility make it difficult to identify all vascular anastomoses. Automatic computer-assisted techniques may provide better understanding of the anatomical structure during surgery for risk-free laser photocoagulatio

Artificial intelligence-assisted FTIR spectroscopy for hormone receptor subtyping in formalin-fixed breast Cancer tissues.

BACKGROUND: Determination of estrogen receptor (ER) and progesterone receptor (PR) status is critical for breast cancer subtyping and guiding endocrine therapy. Although immunohistochemistry (IHC) remains the diagnostic gold standard, it is costly, labor-intensive, and prone to interobserver variability. These limitations are particularly restrictive in low-resource settings where access to standardized receptor testing is limited. OBJECTIVE: This study presents a proof-of-concept evaluation of attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy combined with artif

Artificial intelligence in drug discovery: from algorithmic foundations to clinical translation.

Artificial intelligence is quietly reconfiguring the landscape of drug discovery. It sharpens the search for new targets, refines lead compounds, and helps tailor clinical trial designs. Behind these advances sits a family of methods-convolutional, recurrent, and graph neural networks, generative adversarial networks, variational autoencoders, diffusion models, and Transformers-that has found its way into every stage of the pipeline. Tasks that once demanded years of trial and error, from pulling meaningful features out of molecules to predicting drug-target affinity and crystal structures wit

The genetic architecture of epilepsy across molecular mechanisms and clinical heterogeneity.

Epilepsy comprises a highly heterogeneous group of neurological disorders unified by a persistent predisposition to recurrent seizures, yet driven by remarkably diverse genetic, molecular, and network-level mechanisms. Advances in genomic technologies have revealed that epilepsy arises from a multilayered genetic architecture encompassing rare high-penetrance monogenic variants, common polygenic risk factors, brain-restricted somatic mosaicism, and extensive gene-environment interactions. These genetic substrates converge on core biological pathways regulating neuronal excitability, synaptic t

Finger drawing on smartphone screens enables early Parkinson's disease detection through hybrid 1D-CNN and BiGRU deep learning architecture.

BACKGROUND: Parkinson's disease (PD), a progressive neurodegenerative disorder prevalent in aging populations, manifests clinically through characteristic motor impairments including bradykinesia, rigidity, and resting tremor. Early detection and timely intervention may delay disease progression. Spiral drawing tasks have been established as effective auxiliary diagnostic tools. This study developed a hybrid deep learning model to analyze motion data from finger drawings of spiral and wave lines on smartphone screens, aiming to detect early Parkinson's disease. METHODS: We recruited 58 age-ma

Optimal Low-Cost MEMS INS/GNSS Integrated Georeferencing Solution for LiDAR Mobile Mapping Applications.

Mobile mapping systems using LiDAR technology are becoming a reliable surveying technique to generate accurate point clouds. Mobile mapping systems integrate several advanced surveying technologies. This research investigated the development of a low-cost, accurate Microelectromechanical System (MEMS)-based INS/GNSS georeferencing system for LiDAR mobile mapping applications, enabling the generation of accurate point clouds. The challenge of using the MEMS IMU is that it is contaminated by high levels of noise and bias instability. To overcome this issue, new denoising and filtering methods we

Impact of Noise on Deep Learning-Based Pseudo-Online Gesture Recognition with High-Density EMG.

Deep neural network (DNN)-based approaches have demonstrated high accuracy in surface electromyography (sEMG)-driven gesture recognition under controlled conditions. However, estimation performance is known to degrade substantially when exposed to perturbations commonly encountered in the real-world. Here, we investigate the impact of typical noises on an autoencoder augmented recurrent neural network gesture estimator driven by engineered features and feature sets. High-density sEMG (HD-sEMG) signals offering rich information particularly suited to DNN-based algorithms were collected from thi

Optimization assisted framework for thyroid detection and classification: A new ensemble technique.

Ultrasound (US) is an inexpensive and non-invasive technique for capturing the image of the thyroid gland and nearby tissue. The classification and detection of thyroid disorders is still in its infant stage. This study aims to present a new thyroid diagnosis approach, which consists of three phases like "(i) feature extraction, (ii) feature dimensionality reduction, and (iii) classification". Initially, the thyroid images as well as its related data are given as input. From the input image, the features such as" Grey Level Co-occurrence Matrix(GLCM), Grey level Run Length Matrix(GLRM), propos

Multimodal Ensemble Deep Learning to Predict Disruptive Behavior Disorders in Children.

Oppositional defiant disorder and conduct disorder, collectively referred to as disruptive behavior disorders (DBDs), are prevalent psychiatric disorders in children. Early diagnosis of DBDs is crucial because they can increase the risks of other mental health and substance use disorders without appropriate psychosocial interventions and treatment. However, diagnosing DBDs is challenging as they are often comorbid with other disorders, such as attention-deficit/hyperactivity disorder, anxiety, and depression. In this study, a multimodal ensemble three-dimensional convolutional neural network (

End-to-End Deep Learning Model for Predicting Treatment Requirements in Neovascular AMD From Longitudinal Retinal OCT Imaging.

Neovascular age-related macular degeneration (nAMD) is nowadays successfully treated with anti-VEGF substances, but inter-individual treatment requirements are vastly heterogeneous and currently poorly plannable resulting in suboptimal treatment frequency. Optical coherence tomography (OCT) with its 3D high-resolution imaging serves as a companion diagnostic to anti-VEGF therapy. This creates a need for building predictive models using automated image analysis of OCT scans acquired during the treatment initiation phase. We propose such a model based on deep learning (DL) architecture, comprise

Readmission prediction via deep contextual embedding of clinical concepts.

OBJECTIVE: Hospital readmission costs a lot of money every year. Many hospital readmissions are avoidable, and excessive hospital readmissions could also be harmful to the patients. Accurate prediction of hospital readmission can effectively help reduce the readmission risk. However, the complex relationship between readmission and potential risk factors makes readmission prediction a difficult task. The main goal of this paper is to explore deep learning models to distill such complex relationships and make accurate predictions. MATERIALS AND METHODS: We propose CONTENT, a deep model that pr

Emotion generation method in online physical education teaching based on data mining of teacher-student interactions.

Different from conventional educational paradigms, online education lacks the direct interplay between instructors and learners, particularly in the sphere of virtual physical education. Regrettably, extant research seldom directs its focus toward the intricacies of emotional arousal within the teacher-student course dynamic. The formulation of an emotion generation model exhibits constraints necessitating refinement tailored to distinct educational cohorts, disciplines, and instructional contexts. This study proffers an emotion generation model rooted in data mining of teacher-student course

Machine learning for intraoperative prediction of viability in ischemic small intestine.

OBJECTIVE: Evaluation of intestinal viability is essential in surgical decision-making in patients with acute intestinal ischemia. There has been no substantial change in the mortality rate (30%-93%) of patients with acute mesenteric ischemia (AMI) since the 1980s. As the accuracy from the first laparotomy alone is 50%, the gold standard is a second-look laparotomy, increasing the accuracy to 87%-89%. This study investigates the use of machine learning to classify intestinal viability and histological grading in pig jejunum, based on multivariate time-series of bioimpedance sensor data. APPRO
