Thermal and Calorimetric Evaluations of Some Chemically Modified Carbohydrate-Based Substrates with Phosphorus-Containing Groups.

In the present article, we report on the chemical modifications of some carbohydrate-based substrates, such as potato starch, dextran, β-cyclodextrin, agar agar and tamarind, by reacting with diethylchlorophosphate (DECP), in dispersions in dichloromethane (DCM), in the presence of triethylamine (TEA) as the base. The modified substrates, after recovery and purification, were analyzed for their chemical constitutions, thermal stabilities and calorimetric properties using a variety of analytical techniques. These included: solid-state 31P NMR, inductively coupled plasma-optical emission spectro

Raw starch microparticles as BCG adjuvant: Their efficacy depends on the virulence of the infection strains.

The persistence of tuberculosis (TB) as one of the top 10 causes of death worldwide, the growing incidence of multidrug-resistant tuberculosis and the controversial efficacy of the Bacille Calmette-Guérin (BCG) vaccine drives the development of new generation multistage vaccines against this disease that can boost BCG-primed immunity. The use of polymeric microparticles for this purpose increases due to their advantages, especially their good safety levels and intrinsic immunostimulant properties. We recently explored and demonstrated the reinforcing and adjuvant potential of starch microparti

Wheat Bran-Derived Carbohydrates as Functional Food Ingredients: Extraction and Evaluation of Prebiotic Potential.

Wheat bran (WB) is a rich source of non-starch polysaccharides, particularly arabinoxylans, which can be valorized into functional food ingredients with potential prebiotic properties. This study investigated the extraction of carbohydrates from WB using ultrasonic and hydrothermal treatments and evaluated their technological and functional characteristics and in vitro prebiotic potential. The extraction yields reached 43.47% and 72.36% for ultrasonic and hydrothermal methods, respectively. The extracted carbohydrates exhibited low in vitro digestibility (0.86%-2.04%), indicating resistance to

A central circadian oscillator confers defense heterosis in hybrids without growth vigor costs.

Plant immunity frequently incurs growth penalties, which known as the trade-off between immunity and growth. Heterosis, the phenotypic superiority of a hybrid over its parents, has been demonstrated for many traits but rarely for disease resistance. Here, we report that the central circadian oscillator, CCA1, confers heterosis for bacterial defense in hybrids without growth vigor costs, and it even significantly enhances the growth heterosis of hybrids under pathogen infection. The genetic perturbation of CCA1 abrogated heterosis for both defense and growth in hybrids. Upon pathogen attack, th

Obesity and reversed growth retardation in a child with type Ia glycogen storage disease.

Type Ia Glycogen storage disease is an autosomal recessive hepatic metabolic disease due to a lack of glucose-6-phosphatase (G-6-Pase) activity presenting with growth retardation, lactic acidosis, fasting hypoglycemia with hypoinsulinemia, hyperuricemia, hepatomegaly, and hepatic adenoma with a risk of malignancy. The gene that encodes G-6-Pase was mapped to 17q21. There are some genotype-phenotype correlations. We report a case with delF327 mutation which is devoid of G-6-Pase activity; however clinical presentation in this case differs somewhat. Although correction of hypoglycemia and lactic

Haemodilution tolerance in patients with mitral regurgitation.

Haemodynamic parameters and oxygen consumption were determined in 20 patients with mitral regurgitation before and after a 12 ml.kg-1 isovolaemic exchange of blood for 6% hydroxyethyl starch. During haemodilution, mean (SEM) haemoglobin concentration decreased from 13.0 (0.4) to 10.3 (0.4) g.dl-1 (p = 0.001). With cardiac filling pressures maintained at predilution levels, cardiac index increased from 1.84 (0.08) to 1.94 (0.08) l.min-1.m-2 (p = 0.025) while systemic vascular resistance decreased from 1556 (86) to 1425 (83) dyne.s.cm-5 (p = 0.002) and oxygen extraction increased from 31.7 (1.1)

[Isolation and characterization of two strains of Streptomyces able to metabolize natural polysaccharides including mannan (author's transl)].

Two strains of aerobic and mesophilic microorganisms were isolated from palm-tree plantation sand. They grew on insoluble polysaccharides: mannan, cellulose, chitin as only source of carbon. This lytic activity was used for the purification of the two strains. The morphology of the organisms and the presence of LL-diaminopimetic acid in their cell-wall are characteristic of the genus Streptomyces. Investigations lead to: 1) the characterization of their specific polysaccharidase activity toward insoluble and natural beta-and alpha-glycans (mannan, cellulose, chitin, pectine and starch) and the

Nanoscale Hydroxyapatites Reprogram Phyllosphere Microbiomes and Disease Resistance-Related Phytohormone Biosynthesis in Magnaporthe Oryzae-Infected Rice.

Phyllosphere-microbe-plant interactions underlying nanoscale hydroxyapatite (nHAP)-mediated disease control remain poorly understood. Here, foliar application of nHAP (100-400 mg/L) was evaluated for controlling rice blast caused by Magnaporthe oryzae under greenhouse conditions. The optimal treatment (200 mg/L) reduced disease severity by 42.6% and outperformed ionic Ca and P controls, as well as a commercial fungicide. Mechanistically, nHAP exhibited superior foliar retention (6.88%), enhancing local Ca and P bioavailability, and reshaping the phyllosphere microbiome by enriching beneficial

Comparative transcriptome and metabolome profiling uncovers tissue-specific mechanisms in potato flesh and skin.

Potato (Solanum tuberosum L.), a globally cultivated tuber crop, is crucial for food security. The metabolic specialization between tuber skin and flesh directly influences tuber stress resistance and quality. Transcriptome-metabolome analysis of two cultivars (IAW30 and IAY31) revealed tissue-specific metabolic profiles and molecular mechanisms. Flesh accumulates primary metabolites such as saccharides, amino acids and derivatives, and vitamins, whereas skin is enriched in secondary metabolites including glycoalkaloids and flavonoids. Marker metabolite analysis identified four flesh-specific

Self-layered fruit preservation coating based on chitosan and curcumin.

Long-term fruit storage requires preservatives to reduce spoilage and maintain freshness. For food safety, additives must adhere stably to fruit surfaces without penetrating the skin. This study developed a coating preservation solution (FS) with highly water-soluble aldehyde starch-curcumin-gelatin composite particles (ASCG) and chitosan, offering high adhesion and low permeability for maintaining the quality of fruits such as cherry tomatoes and apples during storage. The coating formed a 1 μm layer on waxy fruit skins, with ASCG creating a bottom antioxidant/antimicrobial layer and surface-

Construction of butyrylated starch film-coated microspheres for colon-targeted delivery of Akkermansia muciniphila postbiotics.

Emerging evidence suggests that Akkermansia muciniphila postbiotics show promising potential in promoting metabolic health. Nevertheless, their bioactivity and colonic availability can be compromised during upper gastrointestinal transit. To address this limitation, a colon-targeted delivery system was developed, based on butyrylated starch (BS) film-coated microspheres (BSFCMs). BS with a tunable degree of substitution was synthesized. Films fabricated from BS exhibited enhanced hydrophobicity and digestion resistance, with the optimal performance observed at DS = 1.79. Next generation industrial biotechnology based on extremophilic bacteria.

Industrial biotechnology aims to produce bulk chemicals including polymeric materials and biofuels based on bioprocessing sustainable agriculture products such as starch, fatty acids and/or cellulose. However, traditional bioprocesses require bioreactors made of stainless steel, complicated sterilization, difficult and expensive separation procedures as well as well-trained engineers that are able to conduct bioprocessing under sterile conditions, reducing the competitiveness of the bio-products. Amid the continuous low petroleum price, next generation industrial biotechnology (NGIB) allows bi

Integrative physiological and transcriptomic analyses reveal key metabolic pathway responses in wax gourd (Benincasa hispida) leaves under potassium deficiency and excess.

Potassium (K) is a key nutrient for increasing yield, improving quality, and enhancing stress resistance in wax gourd (Benincasa hispida). However, the phenotypic, physiological, ecological, and molecular adaptation mechanisms of wax gourd under different K levels remain poorly understood. In this study, a field experiment was conducted with three treatments, i.e., K deficiency (K0), moderate K (300 kg K2O hm-2, K300), and excessive K (500 kg K2O hm-2, K500), to evaluate the effects of K fertilization on the morphology, physiology, and transcriptomics of wax gourd. Results showed that compared

AlphaFold-driven structural proteomics reveals extensive cellulosome machinery in human ruminococcal symbionts.

Cellulosomes are large, surface-displayed enzyme complexes that enable anaerobic bacteria to degrade recalcitrant plant polysaccharides, yet … enriched in amylases and related starch-binding proteins, which may enable this microbe to degrade resistant starches that evade digestion in the upper gastrointestinal tract. Together, these findings reveal that ruminococcal cellulosomes are far more prevalent and diverse than previously appreciated and demonstrate the power of structural proteomics to uncover d …

Meningococcal disease in Malawi: studies on the genetic relatedness of the bacteria.

Seventy-seven meningococci, isolated from patients and carriers during a large epidemic of meningococcal meningitis in Malawi, were characterized in terms of antimicrobial susceptibility, plasmid content and multi-locus enzyme electrophoresis (MLEE). All the isolates were sensitive to chloramphenicol but six had high enough minimum inhibitory concentrations of penicillin (> or = 2 mg/litre) to render them clinically resistant. Only one isolate was sensitive to sulphonamides but all the isolates were sensitive to rifampicin and ciprofloxacin, two drugs that would be suitable alternatives in pro

Energy consumption and intestinal microbiome disorders of yellow catfish (Pelteobagrus fulvidraco) under cold stress.

The yellow catfish (P. fulvidraco), as one of the economically-relevant freshwater fish found in China, cannot tolerate cold stress. Understanding the physiological and biochemical mechanisms under cold stress may provide insights for improving yellow catfish management in the cold. Therefore, we investigated the metabolic and intestinal microbiota changes in cold stress in response to induced cold stress. We found that cold stress in yellow catfish lead to a significant increase in the consumption of glucose and triglycerides, as well as increased use of cholesterol as an alternate energy sou
