Isavuconazole is the only US FDA-approved antifungal for treating invasive mucormycosis. We evaluated isavuconazole activity against a globa … avuconazole activity varied by species and this agent inhibited at ≤4 mg/L 85.2%, 72.7%, and 25% of Rhizopus spp. (n = 27; MIC50/90, 1/>8 mg/L), Lichtheimia spp. (n = 11; MIC50/90, 4/8 mg/L), and Mucor spp. (n = 8; MIC50, >8 mg/L) isolates, respectively. Posaconazole MIC50/90 values against Rhizopus, Lichtheimia, and Mucor species were 0.5/8 mg/L, 0.5/1 mg …

Rhino-orbital-cerebral mucormycosis (ROCM) is an invasive mycosis caused by fungi of the order Mucorales. It mainly affects patients with uncontrolled diabetes. This case report concerns a 27-year-old woman with diabetic ketoacidosis who presented with a large erythematous plaque localised in the right orbitofrontal region with exophthalmos and patches of necrosis that rapidly led to death within 3 days before the diagnosis of ROCM due to Rhizopus oryzae, the causal agent. So, without rapid diagnosis and swift management, ROCM results in a potential fatality.

Rhinocerebral mucormycosis is the most common form of mucormycosis occurring commonly in patients of diabetic ketoacidosis. Fungi of the order Mucorales belong to six families, among whom Rhizopus is the most common, while Mucor is a rare cause. We report a 45-year-old female with uncontrolled diabetes mellitus diagnosed to have rhinocerebrocutaneous mucormycosis caused by Mucor species. The diagnosis was confirmed on histology and culture. A high-index of suspicion is required for early diagnosis and timely initiation of therapy to optimize the outcome. Our patient succumbed to her infection.

Rhinocerebral mucormycosis is a serious infectious disease with a high mortality rate that requires early medical treatment. Liposomal amphotericin B is the medical treatment of choice because of its low renal toxicity and good tissue diffusion. Surgical treatment is necessary, even if all affected areas cannot be reached. The surgical indication should be based on individual characteristics and disease evolution. The case of a 56-year-old man with rhinocerebral mucormycosis caused by Rhizopus and treated with liposomal amphotericin B and extensive rhino-orbital surgical treatment is commented…

PURPOSE: To report rapidly progressive rhino-orbitocerebral mucormycosis in a 47-year-old woman with both diabetes mellitus and the acquired immunodeficiency syndrome (AIDS).
METHOD: Case report including postmortem examination of the eyes, orbit, and brain of a patient with rhino-orbitocerebral mucormycosis, diabetes mellitus, and AIDS.
RESULTS: Invasion by hyphae of the Rhizopus species produced an occlusive vasculitis and massive infarction of the optic chiasm. Fungal organisms were discovered in multiple granulomata of the optic nerve.
CONCLUSION: Infarction at the optic chiasm probably…

We report on a case of a 26-year-old intravenous narcotic abuser with a primary cerebral mucormycotic abscess caused by Rhizopus oryzae. He was treated with a combination of intravenous and intraventricular amphotericin B and surgical drainage with a successful outcome. There was no evidence that his infection was acquired by the rhinocerebral route, it seems likely that he injected himself with a contaminated batch of narcotic or amphetamine. Mucormycosis presenting in this way has been described previously but this is only the second such case to survive. Early diagnosis and treatment is ess…

A fatal rhinocerebral zygomycotic infection caused by Saksenaea vasiformis in a 71-year-old man was diagnosed based on the presence of broad, infrequently septate, branched, hyaline hyphae in tissue obtained from the right and left base of the skull, soft tissue, both maxillary sinuses and the sphenoid sinus; isolation of S. vasiformis from the tissue; and demonstration of mucoraceous antibodies by enzyme-linked immunosorbent assay. Fluorescent antibody studies carried out with a Rhizopus arrhizus conjugate, stained hyphal fragments in tissue with a 2+ intensity.

In this paper we describe a new method for the assay of calcium-dependent phospholipases A. In this method released fatty acids are quantitated by an enzymatic colorimetric reagent kit which is commercially available. We have tested the applicability of this assay with enzymes from porcine pancreas (phospholipase A2), snake venom (phospholipase A2), and Rhizopus arrhizus (a lipase with phospholipase A1-like activity) as well as with a phospholipase A2 activity of bovine seminal vesicle fluid. We conclude that the kit procedure is a convenient, rapid and sensitive endpoint assay for calcium-dep…

Mucormycosis is considered a rare but highly lethal fungal infection, often occurring in patients with poorly controlled diabetes or immunosuppression. Pulmonary mucormycosis progresses rapidly and is often associated with pulmonary infarction and hemoptysis. In this case report, we presented a young, immunocompetent female patient with newly diagnosed diabetes who was diagnosed early with Rhizopus delemar infection through metagenomic next-generation sequencing. Despite early diagnosis, the infection progressed rapidly, invading the tracheal cartilage and upper mediastinal soft tissue, ultima…

A 39-year-old female with insulin-dependent diabetes mellitus developed Rhizopus infection of the maxillary sinus. Subsequent to successful treatment with amphotericin B and surgical debridement, she developed purulent meningitis due to a mucoid strain of Pseudomonas aeruginosa. Analysis of cerebrospinal fluid documented the presence of a uronic acid polymer at a concentration of 40 micrograms/ml. In spite of parenteral and intrathecal antibiotic therapy, the patient died. This case illustrates that mucoid strains of P. aeruginosa may result in fatal infection and that alginate capsule is prod…

Results using homogenate antigens of Rhizomucor pusillus against immunized rabbit sera, mouse ascites fluid and sera from mucormycotic-infected mice, detected by immunodiffusion and complement fixation assays are presented. Using immunodiffusion techniques, common antigens and unique antigens from Rh. pusillus, Rhizopus oryzae and Absidia corymbifera were observed. Complement fixation titers of greater than 256 and 128 were obtained in immunized rabbit sera and mouse ascites fluid, respectively. However, no appreciable titers of antibody were measured from mucormycotic-infected animals by eith…

Endosymbioses have profoundly impacted the evolution of life and continue to shape the ecology of a wide range of species. They give rise to new combinations of biochemical capabilities that promote innovation and diversification1,2. Despite the many examples of known endosymbioses across the tree of life, their de novo emergence is rare and challenging to uncover in retrospect3-5. Here we implant bacteria into the filamentous fungus Rhizopus microsporus to follow the fate of artificially induced endosymbioses. Whereas Escherichia coli implanted into the cytosol induced septum formation, effec…

Defining suitable enzymes for reaction steps in novel synthetic pathways is crucial for developing microbial cell factories for non-natural products. Here, we developed a computational workflow to identify C12 alcohol-active UDP-glycosyltransferases. The workflow involved three steps: (1) assembling initial candidates of putative UDP-glycosyltransferases, (2) refining selection by examining conserved regions, and (3) 3D structure prediction and molecular docking. Genomic sequences from Candida, Pichia, Rhizopus, and Thermotoga, known for lauryl glucoside synthesis via whole-cell biocatalysis, …

INTRODUCTION AND IMPORTANCE: Rhino-orbital-cerebral mucormycosis is an opportunistic infection caused by fungus species Rhizopus and Mucor. Early recognition and aggressive management is crucial for favorable outcomes. A delay in diagnosis and treatment is fatal.
