Reflections on the early days of the DNA damage response pathway in eukaryotes.

The DNA damage response (DDR) pathway has emerged as the central signaling network that preserves genome stability by using sensors to monitor the genome for aberrant structures and establishing signaling centers that promote organismal homeostasis. This review revisits the formative years of DDR research, focusing on the genetic dissection of the pathway in Saccharomyces cerevisiae and its evolution into a broadly conserved eukaryotic signaling system. Early studies of ribonucleotide reductase regulation, replication stress-induced transcription, and cell cycle control identified key regulato

Integrated Metabolomics and Proteomics Reveal Gender-Dependent Immunotoxicity Mechanisms of Mineral Oil Saturated Hydrocarbons in Fischer 344 Rats.

A 6 month subchronic toxicity study demonstrated that exposure to the mineral oil saturated hydrocarbon (MOSH) subfraction (predominant carbon range C20-30) induced dose- and gender-dependent immunometabolic disruption in Fischer 344 rats. Both low- (1.5 g/kg) and high-dose (15 g/kg) exposure significantly reduced the arterial blood CD4+/CD8+ T-cell ratio in both genders (female: 1.85 ± 0.40 and 1.27 ± 0.13 vs 3.14 ± 0.20 in controls; male: 1.04 ± 0.39 and 0.95 ± 0.26 vs 4.38 ± 0.54 in controls), indicating concomitant immune dysregulation and inflammation. Integrated metabolomic and proteomic

A katE katG double-knockout E. coli strain eliminates the risk of catalase contamination in recombinant proteins.

Recombinant protein expression in E. coli is a key methodology for modern biomedical research. Typically, a polyhistidine-tagged ("His-tagged") protein is purified using immobilized metal affinity chromatography (IMAC), achieving close to apparently homogenous target protein preparations. However, contaminant host proteins may nonetheless be co-purified at trace amounts. This includes bacterial catalase, which can even be found crystallized instead of an intended target protein. Here, we found that less than 0.03% of the original endogenous bacterial catalase remaining in a final recombinant p

Adaptive Regulation of dNTP Homeostasis Confers Osimertinib Resistance in EGFR-Mutant Non-Small Cell Lung Carcinoma.

UNLABELLED: Maintaining sustained deoxyribonucleotide triphosphate (dNTP) pools is essential for DNA replication fidelity and genome stability. In EGFR-mutant non-small cell lung carcinoma (NSCLC), we find that disruption of dNTP homeostasis plays a critical role in determining sensitivity to the EGFR inhibitor osimertinib (Osi) and in shaping mechanisms of acquired resistance. Transcriptomic and biochemical analyses revealed that Osi suppresses RRM2 expression, a key regulator of dNTP synthesis, through downregulation of the transcription factor MYBL2. In response to Osi-mediated replication

Metabolomics-driven insights into the multi-target antibacterial mechanisms of 2-methoxycinnamaldehyde against Bacillus cereus and its application in pork preservation.

Bacillus cereus is a major foodborne pathogen characterized by robust biofilm formation and increasing antimicrobial resistance. This study identified 2-Methoxycinnamaldehyde (MCA) from Toona sinensis as the principal antibacterial compound and evaluated its inhibitory mechanisms and preservation potential. MCA exhibited potent activity against B. cereus ATCC 11778 (MIC = 150 μg/mL; MBC = 200 μg/mL) and resistant strains. In vitro assays demonstrated that MCA induced concentration-dependent membrane disruption, DNA damage, and intracellular protein leakage. Furthermore, treatment at 1 MIC and

Prenatal paracetamol exposure and autism spectrum disorder: A critical review of proposed mechanisms, epidemiological evidence, and causal inference.

Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition characterized by persistent deficits in social communication and restricted, repetitive patterns of behavior. Recent claims linking prenatal exposure to paracetamol (acetaminophen), the most commonly analgesic during pregnancy, to increased risk of ASD. This review aims to critically evaluate the validity of these associations, clarify potential biological mechanisms, and provide a balanced, evidence-based perspective to inform clinical practice. Although several observational cohort studies report statistical association

Metformin inhibits PDGFβ signaling to suppress hyaluronan and cytokine production in thyroid eye disease.

Thyroid eye disease (TED) is characterized by fibroblast-driven inflammation and extracellular matrix expansion, which contribute to orbital congestion and proptosis. Platelet-derived growth factor-β (PDGFβ) promotes hyaluronan (HA) synthesis and cytokine production in orbital fibroblasts (OFs); however, whether metabolic modulation can counteract this pathway is unknown. We tested whether metformin, an indirect activator of AMP-activated protein kinase (AMPK), attenuates PDGFβ signaling in TED OFs. Primary OFs from 14 TED and 4 non-TED donors were treated with PDGFβ with or without metformin

Self-assembled phenylboronic acid nanomedicine targets sialic acid to synergistically activate ferroptosis via RRM1 suppression and GPX4 Inhibition for precision colon cancer therapy.

Despite the promise of ferroptosis in cancer therapy, its insufficient potency and off-target toxicity remain significant challenges. Few studies have explored the therapeutic potential of nanoparticle (NP)-mediated active targeting strategies to induce robust ferroptosis in colon cancer (COC) cells effectively. In this study, we developed sialic acid (SA)-targeted NPs by conjugating artesunate (ART) with alanine (Ala) and phenylboronic acid (PBA) to formulate ART-Ala-PBA NPs (AAP NPs). Compared with nontargeted ART-Ala NPs, AAP NPs accumulated significantly within cancer tissues, inhibited tu

CSN6 Promotes Pancreatic Cancer Progression and Gemcitabine Resistance via Antagonizing DCAF1-Mediated Ubiquitination of NPM1.

Pancreatic ductal adenocarcinoma (PDAC) is a fatal cancer with poor prognosis. COP9 signalosome subunit 6 (CSN6), a key regulator of different E3 ubiquitin ligases, plays oncogenic roles in various cancers. However, its function in PDAC remains elusive. Here, this demonstrates that human PDAC tumors expressing high levels of CSN6 present with poor prognosis and gemcitabine resistance. Conditional knockout (KO) of CSN6 hinders tumor formation in a KPP spontaneous PDAC mouse model. Proteomic analysis indicates that CSN6 promotes ribosome biogenesis by activating rDNA transcription and protein sy

Nitric Oxide Inhibition of Glycyl Radical Enzymes and Their Activases.

Innate immune response cells produce high concentrations of the free radical nitric oxide (NO) in response to pathogen infection. The antimicrobial properties of NO include nonspecific damage to essential biomolecules and specific inactivation of enzymes central to aerobic metabolism. However, the molecular targets of NO in anaerobic metabolism are less understood. Here, we demonstrate that the Escherichia coli glycyl radical enzyme pyruvate formate lyase (PFL), which catalyzes the anaerobic metabolism of pyruvate, is irreversibly inhibited by NO. Using electron paramagnetic resonance and site

Dimethylarsinic acid induces bladder carcinogenesis via the amphiregulin pathway.

Dimethylarsinic acid (DMA) is a major metabolite in the urine of humans and rats exposed to inorganic arsenicals, and is reported to induce rat bladder carcinogenesis. In the present study, we focused on early pathways of carcinogenesis triggered by DMA that were also active in tumors. RNA expression in the bladder urothelium of rats treated with 0 and 200 ppm DMA in the drinking water for 4 weeks and in bladder tumors of rats treated with 200 ppm DMA for 2 years was initially examined using microarray analysis and Ingenuity Pathway Analysis (IPA). Expression of 160 genes was altered in both t

The Human Ocular Surface Microbiome and Its Associations with the Tear Proteome in Dry Eye Disease.

Although dry eye disease (DED) is one of the most common ocular surface diseases worldwide, its pathogenesis is incompletely understood, and treatment options are limited. There is growing evidence that complex interactions between the ocular surface microbiome (OSM) and tear fluid constituents, potentially leading to inflammatory processes, are associated with ocular surface diseases such as DED. In this study, we aimed to find unique compositional and functional features of the OSM associated with human and microbial tear proteins in patients with DED. Applying whole-metagenome shotgun seque

HNRNPK alleviates RNA toxicity by counteracting DNA damage in C9orf72 ALS.

A 'GGGGCC' repeat expansion in the first intron of the C9orf72 gene is the most common genetic cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). The exact mechanism resulting in these neurodegenerative diseases remains elusive, but C9 repeat RNA toxicity has been implicated as a gain-of-function mechanism. Our aim was to use a zebrafish model for C9orf72 RNA toxicity to identify modifiers of the ALS-linked phenotype. We discovered that the RNA-binding protein heterogeneous nuclear ribonucleoprotein K (HNRNPK) reverses the toxicity of both sense and antisense repea

Proteome-Based Investigation Identified Potential Drug Repurposable Small Molecules Against Monkeypox Disease.

Monkeypox Virus (MPXV), the causative agent of Monkeypox (MPX) disease, is an emerging zoonotic pathogen spreading in different endemic and non-endemic nations and creating outbreaks. MPX treatment mainly includes Cidofovir and Tecovirimat but they have several side effects and solely depending on these drugs may promote the emergence of drug-resistant variants. Hence, new drugs are required to control the spread of the disease. In this study, we explored the MPXV proteome to suggest repurposable drugs. DrugBank screening revealed drugs such as Brinzolamide, Dorzolamide, Methazolamide, Zidovud

Double-staining Immunohistochemistry Reveals in Malignant Pleural Mesothelioma the Coexpression of ERCC1 and RRM1 as a Frequent Biological Event Related to Poorer Survival.

Malignant pleural mesothelioma (MPM) is a rare cancer with a poor prognosis. To date, standard MPM therapy is still limited to surgery, radiotherapy, and chemotherapy, including pemetrexed and platinum compounds. The main mechanisms of platinum resistance are associated with DNA repair pathways. Excision repair cross-complementing group 1 (ERCC1) and ribonucleotide reductase subunit M1 (RRM1) are important components of the DNA repair, considered as prognostic and predictive biomarkers in various cancer types. The main goal of the present study was to investigate the ERCC1 and RRM1 expression

Imatinib protects against human beta-cell death via inhibition of mitochondrial respiration and activation of AMPK.

The protein tyrosine kinase inhibitor imatinib is used in the treatment of various malignancies but may also promote beneficial effects in the treatment of diabetes. The aim of the present investigation was to characterize the mechanisms by which imatinib protects insulin producing cells. Treatment of non-obese diabetic (NOD) mice with imatinib resulted in increased beta-cell AMP-activated kinase (AMPK) phosphorylation. Imatinib activated AMPK also in vitro, resulting in decreased ribosomal protein S6 phosphorylation and protection against islet amyloid polypeptide (IAPP)-aggregation, thioredo

AMP-activated protein kinase signaling regulated expression of urea cycle enzymes in response to changes in dietary protein intake.

Abundance of urea cycle enzymes in the liver is regulated by dietary protein intake. Although urea cycle enzyme levels rise in response to a high-protein (HP) diet, signaling networks that sense dietary protein intake and trigger changes in expression of urea cycle genes have not been identified. The aim of this study was to identify signaling pathway(s) that respond to changes in protein intake and regulate expression of urea cycle genes in mice and human hepatocytes. Mice were adapted to either HP or low-protein diets followed by isolation of liver protein and mRNA and integrated analysis of

AMP-activated kinase is a regulator of fibroblast growth factor 23 production.

Fibroblast growth factor 23 (FGF23) is a proteohormone regulating renal phosphate transport and vitamin D metabolism as well as inducing left heart hypertrophy. FGF23-deficient mice suffer from severe tissue calcification, accelerated aging and a myriad of aging-associated diseases. Bone cells produce FGF23 upon store-operated calcium ion entry (SOCE) through the calcium selective ion channel Orai1. AMP-activated kinase (AMPK) is a powerful energy sensor helping cells survive states of energy deficiency, and AMPK down-regulates Orai1. Here we investigated the role of AMPK in FGF23 production.
