Cannabinoid Receptor Type 2 Agonist JWH-133 Stimulates Antiviral Factors and Decreases Proviral, Inflammatory, and Neurotoxic Proteins in HIV-Infected Macrophage Secretome.

Although antiviral therapy has improved quality of life, around 50% of people with HIV (PWH) experience neurodegeneration and cognitive decline. This is prompted in part by the migration of HIV-infected monocyte-derived macrophages (MDMs) to the brain, leading to neuronal death. Previous studies in our lab have shown that HIV-infected MDMs secrete cathepsin B (CATB), which is a pro-inflammatory neurotoxic enzyme that is reduced by the addition of cannabinoid receptor-2 (CB2R) agonist JWH-133 to cell cultures. In this study, we aimed to identify the proteins secreted (secretome) by HIV-infected

Protective Signature of IFNγ-Stimulated Microglia Relies on miR-124-3p Regulation From the Secretome Released by Mutant APP Swedish Neuronal Cells.

Microglia-associated inflammation and miRNA dysregulation are key players in Alzheimer's disease (AD) pathophysiology. Previously, we showed miR-124 upregulation in APP Swedish SH-SY5Y (SWE) and PSEN1 iPSC-derived neurons and its propagation by the secretome (soluble and exosomal fractions). After modulation with miR-124 mimic/inhibitor, we identified common responsive mechanisms between such models. We also reported miR-124 colocalization with microglia in AD patient hippocampi. Herein, we determined how miR-124 modulation in SWE cells influences microglia polarized subtypes in the context of

Analysis of ovarian cancer cell secretome during epithelial to mesenchymal transition reveals a protein signature associated with advanced stages of ovarian tumors.

Ovarian cancer (OvCA) is the most lethal neoplasia among gynecologic malignancies and faces high rates of new cases particularly in South America. In special, the High Grade Serous Ovarian Carcinoma (HGSC) presents very poor prognosis with deaths caused mainly by metastasis. Among several mechanisms involved in metastasis, the Epithelial to Mesenchymal Transition (EMT) molecular reprogramming represents a model for latest stages of cancer progression. EMT promotes important cellular changes in cellular adhesion and cell-cell communication, which particularly depends on the paracrine signaling

Comparative secretome profile analysis of cultured immortalized human endometrial stromal cells supplemented with implanted versus nonimplanted blastocyst-conditioned medium: A preliminary analysis.

AIM: Human endometrial stromal cells (HESCs) were previously shown to be capable of discriminating embryos with different qualities. Here we aimed to compare the specific response of the HESC secretome to implanted blastocyst-conditioned medium (BCM) versus nonimplanted medium and identify cytokine candidates useful for the assessment of blastocyst implantation. METHODS: Cleavage embryos were individually cultured in one microdrop of medium for blastocyst formation. The BCM was collected after fresh blastocyst transfer on day 5 and used to supplement HESC culture medium. A high-throughput ant

Immunoscreening of the M. tuberculosis F15/LAM4/KZN secretome library against TB patients' sera identifies unique active- and latent-TB specific biomarkers.

Tuberculosis (TB) protein biomarkers are urgently needed for the development of point-of-care diagnostics, new drugs and vaccines. Mycobacterium tuberculosis extracellular and secreted proteins play an important role in host-pathogen interactions. Antibodies produced against M. tuberculosis proteins before the onset of clinical symptoms can be used in proteomic studies to identify their target proteins. In this study, M. tuberculosis F15/LAM4/KZN strain phage secretome library was screened against immobilized polyclonal sera from active TB patients (n = 20), TST positive individuals (n = 15) a

The exosomal secretomes of mesenchymal stem cells extracted via 3D-printed lithium-doped calcium silicate scaffolds promote osteochondral regeneration.

The development of surface modification techniques has brought about a major paradigm shift in the clinical applications of bone tissue regeneration. Biofabrication strategies enable the creation of scaffolds with specific microstructural environments and biological components. Lithium (Li) has been reported to exhibit anti-inflammatory, osteogenic, and chondrogenic properties by promoting several intracellular signaling pathways. Currently, research focuses on fabricating scaffolds with simultaneous dual bioactivities to enhance osteochondral regeneration. In this study, we modified the surfa

Mesenchymal stem cells and their secretome partially restore nerve and urethral function in a dual muscle and nerve injury stress urinary incontinence model.

Childbirth injures muscles and nerves responsible for urinary continence. Mesenchymal stem cells (MSCs) or their secretome given systemically could provide therapeutic benefit for this complex multisite injury. We investigated whether MSCs or their secretome, as collected from cell culture, facilitate recovery from simulated childbirth injury. Age-matched female Sprague-Dawley rats received pudendal nerve crush and vaginal distension (PNC+VD) and a single intravenous (iv) injection of 2 million MSCs or saline. Controls received sham injury and iv saline. Additional rats received PNC+VD and a s

Metabolomic signatures of probiotic secretomes reveal strain-specific biofunctional potential: insights from Lacticaseibacillus rhamnosus R0011 and Bifidobacterium longum R0175.

Probiotic secretomes are cell-free byproducts of bacterial growth, which contain bioactive metabolites. These metabolites have antioxidant and anti-inflammatory effects and mediate the host-microbe and microbe-microbe interactions. However, the metabolomic profiles of probiotic secretomes are strain-specific and have not been fully investigated. Untargeted metabolomic analysis was used to characterise the metabolomic profiles of Lacticaseibacillus rhamnosus R0011 (R11) secretome (LR) and Bifidobacterium longum R0175 (R175) secretome (BL). LR and BL had distinct metabolomic profiles. LR exhibit

Secretome Derived from Umbilical Cord Mesenchymal Stem Cells as a Therapeutic Approach for Osteoarthritis: Insights from In Vitro, In Vivo, and Clinical Evidence.

Osteoarthritis (OA) is a chronic degenerative joint disorder characterized by articular cartilage deterioration, synovial inflammation, and progressive loss of joint function. Current treatments predominantly address symptoms without modifying structural disease progression, highlighting the need for disease-modifying strategies. Mesenchymal stem cells (MSCs), particularly umbilical cord-derived MSCs (UC-MSCs), exert therapeutic effects primarily through paracrine mechanisms rather than direct engraftment. The UC-MSCs secretome, comprising soluble bioactive factors and extracellular vesicles,

Comparative neuroprotective and cognitive effects of human umbilical cord mesenchymal stem cells, secretome, and exosomes in a rat model of pilocarpine-induced epilepsy.

BACKGROUND AND AIM: Epilepsy is a chronic neurological disorder characterized by recurrent seizures, neuronal degeneration, and cognitive impairment. Current antiepileptic therapies mainly control seizures and do not effectively prevent epileptogenesis or restore neurological function. Human umbilical cord mesenchymal stem cells (HuMSCs) and their cell-free derivatives, including secretome and exosomes, have emerged as promising regenerative therapies. This study aimed to compare the neuroprotective and cognitive effects of HuMSCs, secretome, and exosomes in a rat model of pilocarpine-induced

Cytoprotective and Immunomodulatory Properties of Mesenchymal Stem Cell Secretome and Its Effect on Organotypic Hippocampal Cultures in Mouse Model of Temporal Lobe Epilepsy.

Temporal lobe epilepsy (TLE), the most common form of epilepsy, is often resistant to symptomatic treatment and characterized by persistent neuroinflammation, creating an urgent need for therapeutic strategies that can modulate early disease mechanisms. In this study, we examined the ability of the human MSC-derived secretome to influence epileptic hippocampal tissue during the latent phase of epileptogenesis using an ex vivo model. For this purpose, we characterized the MSC-derived secretome using multiplex Luminex profiling, optimized organotypic hippocampal cultures (OHCs) by evaluating cel

Value of a secretomic approach for distinguishing patients with COVID-19 viral pneumonia among patients with respiratory distress admitted to intensive care unit.

In intensive care units, COVID-19 viral pneumonia patients (VPP) present symptoms similar to those of other patients with Nonviral infection (NV-ICU). To better manage VPP, it is therefore interesting to better understand the molecular pathophysiology of viral pneumonia and to search for biomarkers that may clarify the diagnosis. The secretome being a set of proteins secreted by cells in response to stimuli represents an opportunity to discover new biomarkers. The objective of this study is to identify the secretomic signatures of VPP with those of NV-ICU. Plasma samples and clinical data from

Electrical Pulse Stimulation Partially Restores Insulin Signaling in Palmitate-Induced Insulin-Resistant Skeletal Muscle Cells via Mitochondrial Remodeling and Contractile Activity-Dependent Secretome.

Lipid oversupply induces insulin resistance in skeletal muscle (SkM), partly by altering mitochondrial function and dynamics. Although electrical pulse stimulation (EPS) improves insulin signaling in myotubes, the mechanisms contributing to this effect under lipotoxic conditions remain unclear. Thus, we investigated whether EPS restores lipid-induced insulin resistance in association with changes in mitochondrial respiration and dynamics, lipotoxic intermediates and evaluated the contribution of contraction-induced secreted factors, including small extracellular vesicles (sEV). Mesenchymal stem cell-derived secretome may attenuate nephrotoxicity by modulating cyclooxygenase-2 and caspase-3 levels in Wistar rats: an ELISA-based analysis.

AIM: Nephrotoxicity is a major clinical concern because it can impair kidney function through inflammation and apoptosis. This study aimed to evaluate the effects of mesenchymal stem cell (MSC)-derived secretome on cyclooxygenase-2 (COX-2) and caspase-3 levels in a rat model of doxorubicin-induced nephrotoxicity. METHODS: A total of 24 male Wistar rats were randomly allocated into four groups: normal control (NC), nephrotoxicity control (TC), early MSC-derived secretome treatment (P1), and delayed MSC-derived secretome treatment (P2). Nephrotoxicity was induced using a single intraperitoneal

Legionella pneumophila type II secretome reveals a polysaccharide deacetylase that impacts intracellular infection, biofilm formation, and resistance to polymyxin- and serum-mediated killing.

Prior analyses suggested that the type II secretion system (T2SS) of Legionella pneumophila secretes ≥47 proteins beyond its 26 known substrates. Upon examination of mutants of wild-type strain 130b that lack those exoproteins most conserved across the Legionella genus, we discovered that protein "06635" majorly promotes L. pneumophila replication within amoebae. Immunoblotting, proteomics, and whole-cell enzyme-linked immunosorbent assay (ELISA) confirmed that 06635 exists in culture supernatants and on the bacterial outer surface and does so in a T2SS-dependent manner. Bioinformatic analyses

Curcumin prevents high glucose-induced stimulatory effects of renal cell secretome on fibroblast activation via mitigating intracellular free radicals and TGF-β secretion.

Diabetic kidney disease (DKD) is a leading cause of kidney failure. However, the involvement of renal fibroblasts and their communications with renal epithelial cells during DKD remain poorly understood. We investigated the potential role of renal proximal tubular epithelial cells (PTECs) in renal fibroblast activation that might lead to DKD. Additionally, the protective effects of curcumin, a known antioxidant, against renal fibroblast activation induced by high glucose-treated PTECs were investigated. Secretome was collected from HK-2 PTECs under normal glucose, high glucose, high glucose pr

Depletion of Polypyrimidine tract binding protein 1 (ptbp1) activates Müller glia-derived proliferation during zebrafish retina regeneration via modulation of the senescence secretome.

Polypyrimidine Tract Binding protein 1 (PTB) is an alternative splicing factor linked to neuronal induction and maturation. Previously, knockdown experiments supported a model in which PTB can function as a potent reprogramming factor, able to elicit direct glia-to-neuron conversion in vivo, in both the brain and retina. However, later lineage tracing and genetic knockouts of PTB did not support direct neuronal reprogramming. Nevertheless, consistent with the PTB depletion experiments, we show that antisense knockdown of PTB (ptbp1a) in the zebrafish retina can activate Müller glia-derived pro
