The present authors observed and treated a siblings case of normocalcemic tetany, which is considered as belonging to Bartter's syndrome. As far as we know, there are a number of familial cases of tetany in literature, but none of them spreads over more than two generations, so that the tetany appears to be recessive in hereditary characters including our patients. Both of them presented tetanic seizures in the course of Bartter's syndrome and they were regarded as one of various manifestations of the syndrome. In other words, the Bartter's syndrome or the hypopotassemia should be one of the f…

These siblings of a Czech family aged 21, 19 and 6 years, respectively, with congenital dyserythropoietic anemia, type II, (HEMPAS) are reported. In two elder siblings ferrokinetic studies revealed a rapid plasma 59Fe clearance, markedly decreased erythrocyte incorporation and shortened 51Cr red-cell survival. Direct anti-globulin test was found positive in one of them. Further investigations revealed low values of blood plasma cholesterol, total lipids, beta-lipoproteins, beta-carotine and vitamin E and A as well as low values of the prothrombin complex. Liver biopsy demonstrated siderosis an…

Carney complex (CNC), an inherited disorder, is rarely diagnosed in children. We present two siblings diagnosed with CNC. The younger sister presented with primary amenorrhoea, while the brother sought evaluation for weight gain. Both siblings were cushingoid. The sister displayed characteristic features of CNC, including facial lentigines, adrenocorticotropic hormone (ACTH)-independent Cushing's syndrome (CS), a paradoxical cortisol rise in response to dexamethasone and primary pigmented nodular adrenocortical disease (PPNAD). The second case was more challenging as he had CS but normal ACTH.…

OBJECTIVES: Friedreich ataxia (FRDA) is an autosomal recessive disorder caused by FXN gene mutations involving GAA trinucleotide repeat expansions. This study explores phenotypic heterogeneity between siblings, focusing on differences in age at onset (AAO) and shorter GAA repeat (GAA1) length to improve understanding of disease variability.
METHODS: We analyzed AAO and genotype of siblings with FRDA. Linear regression examined AAO differences and genetic predictors (GAA1 length and SIRT6 S46N single nucleotide polymorphism), while logistic regression assessed discordant clinical manifestation…

Cherubism is an uncommon, multilocular cystic condition of the jaws. This autosomal dominant, nonneoplastic, fibro-osseous condition of the jaws affects children and is self-limiting. Clinically, it shows up as an expansion or distortion of the jaw together with an unnatural teeth alignment. In their most severe forms, these deformations might cause phonation difficulties, decreased pharynx lumen, visual issues, and psychological effects. This case report describes a case report of two siblings involving the maxilla and the mandible presenting with a painless bilateral symmetrical enlargement.…

BACKGROUND: Typically-developing siblings of individuals with Down Syndrome often experience complex emotions towards their sibling. This study explored how social support, personal resources (optimism, sense of coherence [SOC]), and individual variables (sex, religious affiliation, siblings' functionality) may impact emerging adult siblings' emotions toward their sibling with Down Syndrome.
METHODS: Participants were 292 siblings of individuals with DS ranging in age from 18-27 (M=21.54, SD=2.50). Participants completed self-report questionnaires exploring optimism, SOC, support, and accepta…

INTRODUCTION: Poretti-Boltshauser Syndrome (PBS) is a rare neuro-ophthalmological disorder with autosomal recessive inheritance. It is characterized by non-progressive cerebellar ataxia, delay in neuropsychomotor development, intellectual disability, and vision abnormalities. PBS is caused by mutations in the LAMA1 gene, resulting in cerebellar abnormalities, including cerebellar cysts in most cases.
CASE PRESENTATION: We present two siblings with LAMA1 mutations and distinct phenotypic presentation, with one of them showing no evidence of cerebellar cysts on magnetic resonance imaging (MRI).…

Pyloric atresia is a rare gastrointestinal anomaly with an incidence of 1/100,000 in live births. It is usually seen as an isolated condition or in combination with other congenital or hereditary anomalies. Autosomal recessive inherited either fatal or non-fatal variants of pyloric atresia with epidermolysis bullosa are known due to mutations in ITGA6, ITGB4, and PLEC genes. ITGB4 gene mutation was recently identified in 5 siblings in 2 families associated with familial isolated pyloric atresia. Herein, we present two siblings who had pyloric atresia together with a homozygous variant in the I…

Cerebral folate deficiency (CFD) is a rare progressive neurological condition characterized by normal blood folate level and low 5-methyltetrahydrofolate (5-MTHF) levels in the cerebrospinal fluid. Patients present with different neurological findings including hypotonia and microcephaly. Later, patients develop ataxia, seizures, para or quadri-plagia. Herein, we report two siblings; born to consanguineous parents; who had normal neurological development in early childhood. Subsequently they developed drug-resistant seizures, neurological regression, and spastic quadriplegia. After thorough in…
