SO2 is a well-known signal molecule and one of reactive sulfur species, which closely participates in many metabolic processes. While unbalanced metabolism of sulfur dioxide can lead to serious complications of various diseases. Therefore, a rapid and accurate monitoring of SO2 derivatives with high selectivity and sensitivity would be beneficial for their bio-analytic studies. Herein, a novel ratiometric fluorescent probe (NG-TCF) based on ICT mechanism for monitoring SO2 was developed. The probe underwent a nucleophilic addition of HSO3-/SO32- to give rise to a 120 nm blue-shift dual-emissio

Plasma tau phosphorylated at threonine 181 (p-tau181) predicts Alzheimer's disease (AD) pathology with high accuracy in the general population. In this study, we investigated plasma p-tau181 as a biomarker of AD in individuals with Down syndrome (DS). We included 366 adults with DS (240 asymptomatic, 43 prodromal AD, 83 AD dementia) and 44 euploid cognitively normal controls. We measured plasma p-tau181 with a Single molecule array (Simoa) assay. We examined the diagnostic performance of p-tau181 for the detection of AD and the relationship with other fluid and imaging biomarkers. Plasma p-tau

Regulation of membrane receptor mobility tunes cellular response to external signals, such as in binding of B cell receptors (BCR) to antigen, which initiates signaling. However, whether BCR signaling is regulated by BCR mobility, and what factors mediate this regulation, are not well understood. Here we use single molecule imaging to examine BCR movement during signaling activation and a novel machine learning method to classify BCR trajectories into distinct diffusive states. Inhibition of actin dynamics downstream of the actin nucleating factors, Arp2/3 and formin, decreases BCR mobility. C…

UV-DDB, a key protein in human global nucleotide excision repair (NER), binds avidly to abasic sites and 8-oxo-guanine (8-oxoG), suggesting a noncanonical role in base excision repair (BER). We investigated whether UV-DDB can stimulate BER for these two common forms of DNA damage, 8-oxoG and abasic sites, which are repaired by 8-oxoguanine glycosylase (OGG1) and apurinic/apyrimidinic endonuclease (APE1), respectively. UV-DDB increased both OGG1 and APE1 strand cleavage and stimulated subsequent DNA polymerase β-gap filling activity by 30-fold. Single-molecule real-time imaging revealed that UV

Luminescent nanomaterials such as semiconductor nanocrystals (NCs) and quantum dots (QDs) attract much attention to optical detectors, LEDs, photovoltaics, displays, biosensing, and bioimaging. These materials include metal chalcogenide QDs and metal halide perovskite NCs. Since the introduction of cadmium chalcogenide QDs to biolabeling and bioimaging, various metal nanoparticles (NPs), atomically precise metal nanoclusters, carbon QDs, graphene QDs, silicon QDs, and other chalcogenide QDs have been infiltrating the nano-bio interface as imaging and therapeutic agents. Super-resolution and single-molecule microscopies have been increasingly applied to complex biological systems. A major challenge of these approaches is that fluorescent puncta must be detected in the low signal, high noise, heterogeneous background environments of cells and tissue. We present RASP, Radiality Analysis of Single Puncta, a bioimaging-segmentation method that solves this problem. RASP removes false-positive puncta that other analysis methods detect and detects features over a broad range of spatial scales: from single proteins to complex cell phenotypes. RASP outperforms the stat

The clinical assessment, treatment, and prognostic evaluation of breast cancer liver metastasis (BCLM) remain clinically challenging. Hepatic involvement is often identified only after substantial intrahepatic tumor burden, radiologically evident parenchymal involvement, abnormal liver function, or symptoms attributable to hepatic impairment have developed. Standard evaluation relies on molecular subtype, tumor burden, liver function, imaging, metastatic-site biopsy, receptor reassessment, and selected genomic testing; however, these variables do not fully explain why only a subset of dissemin

Understanding how gene activity relates to other biological structures is critical to investigate tissue remodeling processes, disease, and regeneration. RNAscope in situ hybridization assay provides single-molecule detection of targeted transcripts, while label-free multiphoton microscopy enables high-resolution, quantitative imaging of extracellular matrix collagen. These modalities have not previously been combined to extract spatially resolved correlations between molecular and structural features within the same tissue section. Here, we introduce correlative multimodal imaging that integr

Fertility-sparing treatment is an established option for reproductive-age women with early-stage endometrioid endometrial cancer or atypical endometrial hyperplasia. Despite high initial response rates with progestin-based therapy, predicting treatment response and recurrence remains challenging. This narrative review summarizes current evidence on prognostic and predictive biomarkers for conservative management. Progesterone receptor positivity is the most consistent favorable predictor of response, while POLE ultra-mutated tumors show excellent remission and low recurrence. Mismatch repair d

Proper chromosome alignment and segregation during mitosis depend on cohesion between sister chromatids. Cohesion is thought to occur through the entrapment of DNA within the tripartite ring (Smc1, Smc3 and Rad21) with enforcement from a fourth subunit (SA1/SA2). Surprisingly, cohesin rings do not play a major role in sister telomere cohesion. Instead, this role is replaced by SA1 and telomere binding proteins (TRF1 and TIN2). Neither the DNA binding property of SA1 nor this unique telomere cohesion mechanism is understood. Here, using single-molecule fluorescence imaging, we discover that SA1

DNA compaction by human condensin II is crucial for the correct organization of mitotic chromosomes. Mitotic activation of condensin II is a multilayered process, controlled mainly by the opposing activity of the MCPH1 and M18BP1 proteins, acting as an interphase inhibitor and a mitotic activator, respectively. Mitotic phosphorylation favors M18BP1 binding to condensin II, resulting in effective and timely chromosome compaction. However, the molecular mechanisms underlying condensin II activation in driving DNA compaction remain uncharacterized. Here, combining cryo-electron microscopy with si

Activation-induced cytidine deaminase (AID) converts cytosines to uracils within the actively transcribed switch regions to initiate DNA repair and formation of DNA double strand breaks required for immunoglobulin class switch recombination (CSR). How AID specifically targets switch regions remains a key unanswered question. Using a multimodal live cell single molecule imaging approach, we demonstrate that intronic switch regions promote robust transcription by enhancing polymerase loading and persistent transcriptional bursts, resulting in the formation of a dynamic RNA hub consisting of nume…

Sepsis-associated acute kidney injury (SA-AKI) is characterized by tubular inflammation and impaired metabolism, yet effective treatments remain lacking. Here, we identify the purinergic receptor P2Y12 as a key regulator of these pathological processes in renal proximal tubule cells. P2Y12 expression is markedly upregulated after lipopolysaccharide challenge. Mechanistically, P2Y12 activation promotes Gi signaling, suppresses the cAMP-PKA pathway, increases proinflammatory cytokine production, and disrupts gluconeogenesis. Using single-molecule imaging, we directly visualize the interaction be

Understanding biological processes requires spatiotemporal mapping of proliferative and transcriptional dynamics. Current spatial transcriptomics methods capture only protein-coding transcripts and static snapshots, obscuring non-coding RNAs (ncRNAs) and dynamic events. We developed SPTEdU-seq, integrating spatial total transcriptomics with 5-ethynyl-2'-deoxyuridine tracking to co-profile gene expression and proliferation dynamics. SPTEdU-seq demonstrates ultrahigh sensitivity for coding and non-coding transcripts and for splicing isoforms, with single-molecule probe design eliminating optical

In addition to increased aberrant protein aggregation, inflammation has been proposed as a key element in the pathogenesis and progression of Alzheimer's disease. How inflammation interacts with other disease pathways and how protein aggregation increases during disease are not clear. We used single-molecule imaging approaches and membrane permeabilization assays to determine the effect of chronic exposure to tumour necrosis factor, a master proinflammatory cytokine, on protein aggregation in human-induced pluripotent stem cell-derived neurons harbouring monogenic Alzheimer's disease mutations

Astrocytes constantly adapt their ramified morphology in order to support brain cell assemblies. Such plasticity is partly mediated by ion and water fluxes, which rely on the water channel aquaporin-4 (AQP4). The mechanism by which this channel locally contributes to process dynamics has remained elusive. Using a combination of single-molecule and calcium imaging approaches, we here investigated in hippocampal astrocytes the dynamic distribution of the AQP4 isoforms M1 and M23. Surface AQP4-M1 formed small aggregates that contrast with the large AQP4-M23 clusters that are enriched near glutama

Agitoxin-2 (AgTx2) from scorpion venom is a potent blocker of K+ channels. The docking model has been elucidated, but it remains unclear whether binding dynamics are described by a two-state model (AgTx2-bound and AgTx2-unbound) or a more complicated mechanism, such as induced fit or conformational selection. Here, we observed the binding dynamics of AgTx2 to the KcsA channel using high-speed atomic force microscopy. From images of repeated binding and dissociation of AgTx2 to the channel, single-molecule kinetic analyses revealed that the affinity of the channel for AgTx2 increased during per

Apoptosis is a critical factor in AIDS and other viral illnesses, cerebral and myocardial ischemia, autoimmune and neurodegenerative states, organ and bone marrow transplant rejection, and tumor response to chemotherapy and radiation. Improved methods to identify sites of apoptosis are increasing our understanding of the pathophysiology and treatment of these and numerous other human disorders. Here we describe the most used method for labeling annexin V, a protein with a high affinity for apoptotic cells in vitro, with technetium-99m (99mTc) as a radionuclide imaging agent that can localize a

Epithelial cell adhesion molecule (EpCAM) is overexpressed in various epithelial malignancies and represents an attractive target for molecular imaging and radionuclide therapy. This study aimed to develop and evaluate a novel EpCAM-targeted monoclonal antibody for single-photon emission computed tomography (SPECT) imaging. To this end, Ep4-1, a high-affinity anti-EpCAM monoclonal antibody, was conjugated to the bifunctional chelator p-SCN-Bn-diethylenetriaminepentaacetic acid (DTPA) and radiolabeled with indium-111 (111In). In vitro EpCAM-binding affinity, plasma stability, and cellular inter

Multimodal phototheranostics utilizing single molecules offer a "one-and-done" approach, presenting a convenient and effective strategy for cancer therapy. However, therapies based on conventional photosensitizers often suffer from limitations such as a single photosensitizing mechanism, restricted tumor penetration and retention, and the requirement for multiple irradiations, which significantly constrain their application. In this report, we present an aggregation-induced emission luminogen (AIEgen) bacteria hybrid bionic robot to address above issues. This bionic robot is composed of multif
