Vaginal malformations: a proposed classification based on embryological, anatomical and clinical criteria and their surgical management (an analysis of 167 cases).

BACKGROUND: Although it is virtually impossible to formulate a scheme that can satisfactorily collect all different types of vaginal malformations, a simple classification would be of a considerable value and would permit logical operative decisions. Many classifications of anomalies of uterus and vagina have been proposed: we find them unsatisfactory and confusing, being either too simple or too complex. We propose a new classification, focused only on vagina and based on embryological, anatomical, clinical and surgical criteria. METHODS: In over 30 years, 167 females with vaginal malformati

Report of a case of Cobb syndrome: multimodality imaging.

Cobb syndrome is a rare vascular disorder characterized by vascular skin lesions distributed in a dermatomal pattern, with corresponding muscular, osseous, paraspinal, and/or spinal vascular lesions occurring at the same body somite (metamere). We present a case of a 25-year-old man who presented with a history of right upper limb paresthesia followed by bilateral progressive upper and lower limb weakness and heaviness. Physical examination showed large cutaneous port wine stains on the right side of the chest, the nape, and along the whole right upper limb in a dermatomal distribution, with n

Congenital pelvic skeletal anomalies: Clinical and radiographic evaluation of newborns with gastrointestinal malformation.

BACKGROUND: Congenital pelvic skeletal anomalies (CPSA) may appear as isolated defects or in association with other anomalies like congenital malformations of the digestive system (CMDS). Minor CPSA in non-syndromic patients are often overlooked. We aimed to assess the frequency of CPSA in newborns with CMDS to review the diagnostic approaches. STUDY DESIGN: A retrospective review of medical records of 201 newborns who underwent X-rays for different neonatal indications was conducted. In 122 patients CMDS were diagnosed and classified according to the ICD-10 classification; 79 non-CMDS patien

Fertilization, hatching, and embryogenesis of diploid and triploid eggs of Anabas testudineus (Bloch, 1792).

SummaryThis study investigated the breeding parameters and embryogenic development of diploid and heat shock-induced triploid eggs of Anabas testudineus (Bloch, 1792). To this effect, broodstocks of A. testudineus were induced to spawn using the Ovaprim® hormone. After fertilization, the eggs were divided into two groups and one portion heat shocked at 41°C (for 3 min), at approximately 4 min after fertilization. Results of fertilization, hatchability, as well as the sequence and timing of embryogenic development were collated from three breeding trials. Fertilization percentages were similar

Rare variants in the notch signaling pathway describe a novel type of autosomal recessive Klippel-Feil syndrome.

Klippel-Feil syndrome is a rare disorder represented by a subgroup of segmentation defects of the vertebrae and characterized by fusion of the cervical vertebrae, low posterior hairline, and short neck with limited motion. Both autosomal dominant and recessive inheritance patterns were reported in families with Klippel-Feil. Mutated genes for both dominant (GDF6 and GDF3) and recessive (MEOX1) forms of Klippel-Feil syndrome have been shown to be involved in somite development via transcription regulation and signaling pathways. Heterotaxy arises from defects in proteins that function in the de

A new species of Colobomatus (Copepoda, Phylichthyidae) parasitic on Mullus argentinae (Perciformes, Mullidae) from South American Atlantic coast.

A new species of Colobomatus Hesse, 1873 is described from pores of the cephalic sensory system and nostrils of Argentine goatfish, Mullus argentinae Hubbs et Marini, 1933 (Perciformes: Mullidae), living along the southwestern Atlantic coast. The fish were collected at different latitudes, stretching from the State of Rio de Janeiro in the north, through Santa Catarina, Rio Grande do Sul (Brazil) to of Mar del Plata (Argentina) in the south. The prevalence of the infection ranged from 42% through 84%. The new species look alike to two other species, parasites of mullids (C. steenstrupi and C.

Primary body axes of vertebrates: generation of a near-Cartesian coordinate system and the role of Spemann-type organizer.

A rationale for the complex-appearing generation of the primary body axes in vertebrates can be obtained if this process is divided into two … crucial role. This oscillation also leads to the repetitive nature of the trunk pattern as seen in somites or segments. The function of the Spemann-type organizer is not to specify the dorsoventral (DV) positional information directly but to initiate the formation of a stripe-shaped midline organizer, realized with different structures in the brain and i …

Expression of Sonic Hedgehog downstream genes is modified in rat embryos exposed in utero to a distal inhibitor of cholesterol biosynthesis.

Holoprosencephaly is a common developmental anomaly of the forebrain and midface, that has been associated with mutations in the Sonic Hedgehog gene, and with perturbations of cholesterol synthesis and metabolism in mammalian embryos. The study presented here was aimed to evaluate the functional relationship between these two causal agents in the genesis of the phenotype. Therefore, we used rat embryos exposed in utero to a distal inhibitor of cholesterol biosynthesis (AY9944) in which we analyzed different Shh-dependent processes, as evaluated by the expression of eight target genes. In addit

Absence of ventral cell populations in the developing brain in a rat model of the Smith-Lemli-Opitz syndrome.

The Smith-Lemli-Opitz syndrome (SLOS) is an autosomal recessive condition involving craniofacial and central nervous system malformations with occasional holoprosencephaly (HPE). It is caused by a defect in the 7-dehydrocholesterol (7-DHC) reductase, the enzyme catalyzing the last step of cholesterol biosynthesis. Treatment of pregnant rats with inhibitors of 7-DHC reductase, either AY9944 or BM15.766, has provided a valuable model to study the pathogenesis in SLOS. Recently, cholesterol has been shown to be involved in the post-translational activation of the signaling protein Sonic Hedgehog.

Evidence that spontaneous situs inversus in cultured neural plate staged rat embryos is additive with and not mediated through adrenergic mechanisms.

Approximately 50% of untreated presomite rat embryos in culture have demonstrated inversions of cardiac looping (laeval instead of dextral) or tail flexure (left-sided instead of right-sided), or both. This spontaneous situs inversus (SI) was not accompanied by growth inhibition or any other observable defects. The incidence of SI was directly related to the stage at dissection, and all heart defects and most flexure defects were eliminated by delaying explantation to the early somite stage. The incidence of SI was not lowered significantly either by removal of endogenous catecholamines from t

Sonic Hedgehog repression underlies gigaxonin mutation-induced motor deficits in giant axonal neuropathy.

Growing evidence shows that alterations occurring at early developmental stages contribute to symptoms manifested in adulthood in the setting of neurodegenerative diseases. Here, we studied the molecular mechanisms causing giant axonal neuropathy (GAN), a severe neurodegenerative disease due to loss-of-function of the gigaxonin-E3 ligase. We showed that gigaxonin governs Sonic Hedgehog (Shh) induction, the developmental pathway patterning the dorso-ventral axis of the neural tube and muscles, by controlling the degradation of the Shh-bound Patched receptor. Similar to Shh inhibition, repressio

Diagnosis of a variant of Mayer-Rokitansky-Kuster-Hauser syndrome: useful MRI findings.

Mayer-Rokitansky-Kuster-Hauser (MRKH) syndrome consists of vaginal aplasia associated with other müllerian duct abnormalities. Its penetrance varies, as does the involvement of other organ systems. Type I MRKU syndrome is characterized by an isolated absence of the proximal two thirds of the vagina, whereas type II is marked by other malformations which include vertebral, cardiac, urologic (upper tract), and otologic anomalies. In both types, the extent of vaginal aplasia varies, ranging from virtually absent to a length much more inferior than the normal one (2-5 cm). MRKU syndrome usually re

Transient morphologies at muscle attachments to the future medial margin of the developing scapula: a histological study using human embryos and early fetuses.

BACKGROUND: The medial margin of the scapula and its related muscles [the serratus anterior (SA), rhomboideus, levator scapulae (LS)] have been among the major interests of embryologists because of their common somitic origin. A systematic and synchronous mechanism appeared to operate during the formation of muscle attachments. MATERIALS AND METHODS: Sagittal and horizontal histological sections from 24 human embryos and fetuses at approximately 5-7 weeks of gestation were examined. RESULTS: Instead of the upper half of the medial margin, the mesenchymal or cartilaginous scapula had a supero

How snake embryos coil.

Snake embryos are often tightly coiled, but the biological basis for this developmental trait remains a puzzle. To understand how and why they coil, we first investigated directions of coiling within and across 39 snake and other limbless squamate species. In early developmental stages, coiling orientation appears to be nearly fixed right-handed across multiple species. We present evidence that accelerated somitogenesis outpaces gastrointestinal extension at these stages, causing the axial column and somitic series to coil around the delayed visceral tissues. To allow this, the body axis and t

Typical Thermochromic Dye Crystal Violet Lactone Acts as an Endocrine Disruptor: Nontargeted Screening, Prioritization, and In Vivo Experimental Verification.

Identification and characterization of endocrine disruptors and their associated ecological and health risks are critical areas of environmental toxicology. In this study, we aimed to identify previously unrecognized endocrine disruptors in thermal paper by employing a nontargeted screening coupled ToxPi Ranking (NSTR) approach. Among the substances identified with high confidence in thermal paper, Crystal Violet Lactone (CVL), a typical thermochromic dye, was prioritized with a high endocrine disruptive potential. CVL can be absorbed through the skin and excreted in the urine. In addition, we

Dephosphorylation of Y228 and Y217 and phosphorylation of Y335 in p120 catenin activate convergent extension during zebrafish gastrulation.

BACKGROUND: The cadherin-associated protein p120 catenin regulates convergent extension through interactions with cadherin proteins, Cdc42, and Rac1, as we previously showed in zebrafish (Danio rerio). Phosphorylation of p120 catenin changes the nature of its activity in vitro but is virtually unexplored in embryos. We used our previously developed antisense RNA splice-site morpholino targeted to endogenous p120 catenin-δ1 to cause defects in axis elongation probing the functions of three p120 catenin tyrosine-phosphorylation sites in gastrulating zebrafish embryos. RESULTS: The morpholino-in
