A Novel Phthalimide-Based Turn-On Fluorescent Probe for Detection of Hydrogen Sulfide and Its Application in Real Samples.

In this paper, we rationally designed a turn-on fluorescent probe PSH for the selective and efficient detection of hydrogen sulfide (H2S) over other biologically relevant species, particularly glutathione (GSH), cysteine (Cys), and homocysteine (Hcy). Consisting of a phthalimide derivative linked with the 2,4-dinitrobenzenesulfonyl (DNBS) group through a sulfonyl ether bond, PSH exhibits a turn-on fluorescence response toward H2S with distinct advantages: a broad pH range (pH 5-9), rapid response kinetics (within 5 min), high selectivity against interfering substances (accompanied by an 8.5-fo

Ratiometric "on/off"-type fluorescence sensors based on nitrogen and silicon co-doped graphene quantum dots for identification and semi-quantitative visualized detection of dopamine, epinephrine, and norepinephrine.

Catecholamines (CAs) play pivotal physiological in the human body, and the aberrant levels of them are closely associated with various diseases. Therefore, developing a rapid, simple, and accurate method for CAs determination is of considerable clinical importance for precise disease diagnosis. In this work, a turn-off fluorescence sensor based on nitrogen and silicon co-doped graphene quantum dots (N,Si-GQDs) was first established for the detection of total CAs, leveraging the inner filter effect (IFE). To achieve the selective identification and separate quantification of dopamine (DA), epin

Sustainable Synthesis of Biomass-Based Carbon Quantum Dots for Selective Fluorescent Recognition of Cr3+ and In Vitro Antioxidant Applications.

The development of cost-effective, eco-friendly, sensitive, and efficient analytical platforms for the monitoring of metal ions holds profound practical value. In this work, edible fungus carbon quantum dots (Ef-CQDs) are synthesized via a facile hydrothermal route using edible fungus as a green carbon precursor, and a novel fluorescence sensing strategy is established for the rapid and selective detection of Cr3+ in environmental water matrices. Systematic optical investigations revealed that the as-prepared Ef-CQDs displayed outstanding selectivity toward Cr3+ over other coexisting metal ion

A Dual Responsive Fluorescent Probe Exhibiting ICT Based Colorimetric and Fluorimetric Detection of Cyanide.

2-(pyridin-4-yl)-8H-thieno[2,3-b] indole (ITP) fluorescent probe has been synthesized and utilized as a ratiometric sensor for cyanide anion. The color change upon the addition of cyanide anion is easily noticeable with naked eyes. The greater difference between the absorption and emission spectrum due to the inclusion of cyanide is attributed to the enhanced intramolecular charge transfer (CT) by hydrogen bonding of CN¯ with ITP, which greatly increased the electron density on the donor thieno[2,3-b] indole. The cyanide sensing mechanism is further confirmed by 1H NMR and DFT studies. Further

Tetrazine-based AIE probes for specific detection of superoxide anion.

Combining the single-electron recognition property of tetrazine with aggregation-induced emission (AIE), two fluorescent probes named SWJT-48 and SWJT-49 were rationally designed and synthesized. This molecular design relied on a specific reduction recognition mechanism to eliminate interference from various substances. Both probes exhibited rapid response, high sensitivity and excellent selectivity toward superoxide anion (O2•-). Both exogenous and endogenous O2•- in HeLa cells were successfully detected using SWJT-49. Furthermore, SWJT-49 achieved fluorescence imaging of O2•- in HT22 cells v

Simultaneous dual-mode detection and removal of acrolein in food samples via a fluorescent PAA-CDs/Sulfhydryl cotton/SA hydrogel composite.

A multifunctional fluorescent carbon dot/sulfhydryl cotton/sodium alginate composite hydrogel (PAA-CDs/Sulfhydryl Cotton/SA hydrogel) had been developed, which could integrate in dual-mode optical responses with fluorescence quenching and colorimetric signals to detect and simultaneous remove acrolein in food samples. The fluorescence detection exhibited a linear range spanning from 0 to 70 μM for acrolein, along with a relatively low LOD of 20 nM. In contrast, smartphone-based RGB colorimetric quantification presented a linear range of 0-70 μM, with an LOD of 350 nM. Meanwhile, the adsorption

Accurate Detection of Hcy in Human Serum and Two-Photon Visualization of Atherosclerosis Using a Highly Specific Fluorescent Probe.

As an important metabolic intermediate of sulfur-containing amino acids in human body, homocysteine (Hcy) is regarded as an independent risk factor for atherosclerotic cardiovascular disease. Therefore, real-time monitoring of the fluctuation of Hcy level is of great importance for the early diagnosis as well as the treatment of atherosclerosis. Herein, a new two-photon (TP) fluorescent probe (RH-2) was developed via a hydrogen bond-assisted strategy, which had a high specificity for detecting Hcy over cysteine (Cys) and glutathione (GSH) in solution, cells, and tissue. Probe RH-2 was applied

Y3+@CdTe quantum dot nanoprobe as a fluorescence signal enhancement sensing platform for the visualization of norfloxacin.

Quinolone antibiotics (norfloxacin) pose a serious threat to animal and human health due to their misuse and difficulty in being broken down in surface water and food. Rapid and effective detection of norfloxacin (NOR) is essential for environmental testing and ecosystems. In this study, yttrium was coordinated with mercaptopropionic acid (MPA)-modified CdTe quantum dots (QDs) to obtain a novel fluorescence sensor Y3+@CdTe QDs for the sensitive detection of NOR. NOR can bind to Y3+ to form a complex (NOR-Y3+). This complex enhances the luminescence of NOR and blue-shifts to 423 nm. The fluores

Fluorescent probe for detecting hydrogen sulfide based on disulfide nucleophilic substitution-addition.

In view of the importance of hydrogen sulfide (H2S) in the organism, a fast, noninvasive method for the detection of H2S in situ is needed. Fluorescent probes based on disulfide-bond nucleophilic substitution-addition can selectively detect H2S in vivo, which is very popular because it allows quick response for H2S, thus it will be a useful tool for monitoring H2S in the vivo. We developed a dicyanoisopentanone-based H2S fluorescent probe (EW-H) that used a disulfide group as a self-destructive linker reaction site. Under the nucleophilic substitution of H2S, the disulfide bond of EW-H was cle

Rational Design of a Red-Emissive Fluorophore with AIE and ESIPT Characteristics and Its Application in Light-Up Sensing of Esterase.

The development of red fluorophores with efficient solid-state emission is still challenging. Herein, a red fluorophore 1 with aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics is rationally designed and facilely synthesized by attaching an electron-donor diethylamine and an electron-acceptor maleonitrile group to salicyladazine. In contrast to many red fluorophores which undergo serious aggregation-caused quenching (ACQ), compound 1 emits bright red fluorescence (λem = 650 nm, ΦF = 24.3%) in the solid state with a large Stokes shift of

The fluorescence regulation mechanism of the paramagnetic metal in a biological HNO sensor.

Paramagnetic metals are frequently used to regulate fluorescence emissions in chemical and biological probes. Accurate quantum calculations offer the first regulation theory that quenching is through the competitive nonradiative decay of the mixed fluorophore/metal (3)ππ*/dd state isoenergetic to the fluorophore-localized (1)ππ* state.

Convenient and highly effective fluorescence sensing for Hg2+ in aqueous solution and thin film.

A quinolinocyclodextrin, that is, MQAS-beta-cyclodextrin (MQAS = N-(2-methyl-8-amino- quinolyl)-p-aminobenzene sulfonamide) was synthesized. Further experiments showed that it could form very stable stoichiometric 2:1 complex with Zn(2+) in water. Significantly, the resultant quinolinocyclodextrin/Zn(2+) complex showed the specific fluorescence response to Hg(2+) over other metal ions, which could be readily distinguished in either aqueous solution or the PVA-based thin film. This finding would enable Zn x 3(2) complex as a convenient and highly efficient fluorescence sensor for the detection

Amphoteric phosphorous(V)-phthalocyanines as proton-driven switchable fluorescers toward deep-tissue bio-imaging.

Spectral (optical absorption and emission) properties of three amphoteric phosphorous(V)-phthalocyanine derivatives, [P(Pc)(O)OH], where Pc=tetra(tert-butyl)phthalocyaninate (tbpc), tetrakis(2',6'-dimethylphenoxy)phthalocyaninate (tppc), and octakis(4'-tert-butylphenoxy)phthalocyaninate (obppc), have been investigated in ethanolic solutions. Spectral changes upon protonation/deprotonation (the reaction sites have been determined to be their axial ligands by magnetic circular dichroism study) are drastic and rapid. All the initial ([P(Pc)(O)OH]), protonated ([P(Pc)(OH)2]+), and deprotonated ([P

Determination of the substrate specificity of tripeptidyl-peptidase I using combinatorial peptide libraries and development of improved fluorogenic substrates.

Classical late-infantile neuronal ceroid lipofuscinosis is a fatal neurodegenerative disease caused by mutations in CLN2, the gene encoding the lysosomal protease tripeptidyl-peptidase I (TPP I). The natural substrates for TPP I and the pathophysiological processes associated with lysosomal storage and disease progression are not well understood. Detailed characterization of TPP I substrate specificity should provide insights into these issues and also aid in the development of improved clinical and biochemical assays. To this end, we constructed fluorogenic and standard combinatorial peptide

Visual and fast detection of trace copper ions using biosensor based on FRET.

The development of a simple, rapid and sensitive sensor to detect copper ion is very important for environmental detection. Here, we constructed a fluorescence biosensor based on fluorescence resonance energy transfer (FRET) between copper resistance operon coded protein C (CopC), a copper binding protein, and dansyl chloride (DNS-Cl) to selectively detect copper ion. At alkaline conditions, DNS-Cl was contently attached to CopC forming biosensor of DNS-Cl/CopC, in which fluorescence emission of CopC at 320nm was absorbed by DNS-Cl. After binding with copper ion, the fluorescence of DNS-Cl was

Sustained glycolytic oscillations--no need for cyanide.

Using fluorescence spectroscopy we detected long trains of macroscopic oscillations in the glycolytic pathway, in whole cell suspensions of Saccharomyces cerevisiae, without addition of cyanide. Such oscillations may be induced if argon or another inert gas is bubbled through the yeast cell suspension. This supports that the synchronizing agent is a volatile compound secreted by the yeast cells, e.g. CO2 and/or acetaldehyde. Our results show that the rate of acetaldehyde removal is not a crucial parameter to the synchronization of the yeast cells. The sample cell was connected to a membrane in

The formation of glutathione-chromium complexes and their possible role in chromium disposition.

The reduction of Cr(VI) to Cr(III) by glutathione (GSH) in vitro resulted in the formation of two different GSH-chromium complexes with an approximate molecular-weight of 10,000 and 5000, respectively. Both complexes exhibited strong fluorescence. The molar ratio between oxidized glutathione (GSSG) and chromium was calculated by determining the chromium content of the two isolated complex species by flameless Atomic Absorption Spectrometry (AAS).

Fluorescent biosensing of glucose and ATP in biological systems: Photophysical mechanisms and AI-assisted probe design.

Glucose and adenosine triphosphate (ATP) are essential metabolic biomarkers involved in cellular energy regulation, physiological homeostasis, and disease progression. Accurate and real-time detection of these molecules is therefore critically important in biological and biomedical research. Although conventional analytical techniques provide excellent sensitivity and accuracy, their dependence on sophisticated instrumentation and extensive sample preparation limits their application in continuous and point-of-care monitoring. Fluorescence spectroscopy has emerged as a powerful analytical appr

Novel zinc nanoclusters based ratiometric fluorescence sensing strategy for sensitive detection of thiram.

Thiram residue has drawn widespread attention due to its severe threat to the agricultural safety. Herein, a ratiometric fluorescence probe was designed for thiram detection based on glutathione capped zinc nanoclusters (GSH-ZnNCs) and o-phenylenediamine (OPD). OPD was oxidized by Ag+ to produce 2, 3-diaminophenazine (DAP), and the fluorescence of GSH-ZnNCs was quenched by DAP through fluorescence resonance energy transfer (FRET) effect. When exposed to thiram, Ag+ led to the cleavage of SS bond in thiram to form Ag+-dimethyl thiocarbamate complex, inhibiting the oxidation of OPD. Accordingly,
