Conjugation of chitosan nanoparticles with biogenic and synthetic polyamines: A delivery tool for antitumor polyamine analogues.

We report the conjugation of chitosan nanoparticles with biogenic polyamines spermine (spm), spermidine (spmd) and synthetic polyamines 3,7,11,15-tetrazaheptadecane.4HCl (BE-333) in aqueous solution. Multiple spectroscopic methods, thermodynamic parameters and molecular modeling were used to analyse polyamine bindings to chitosan nanoparticles. Thermodynamic parameters ΔS, ΔH and ΔG showed that polyamines bind protein through H-bonding and hydrophobic contacts with biogenic polyamines form more stable conjugates than synthetic polyamines. As polymer size increases the stability of polyamine-ch…

Assessment and supplementation of antioxidant amines in male infertility-are we approaching clinical implementation?

Male infertility remains unexplained in a substantial proportion of cases, despite extensive clinical and biological investigations. Oxidative stress has emerged as a central pathophysiological mechanism underlying impaired sperm function, yet current diagnostic and therapeutic strategies inadequately capture the complexity of redox regulation within the male reproductive tract. In this narrative review, we revisit both the historical foundations and recent advances supporting a pivotal role for a specific class of metabolites: polyamines (spermine, spermidine, and putrescine) and carnitine, w…

The telencephalon is a neuronal substrate for systemic inflammatory responses in teleosts via polyamine metabolism.

Systemic inflammation elicits sickness behaviors and fever by engaging a complex neuronal circuitry that begins in the preoptic area of the … amine metabolism changes, characterized by accumulation of putrescine and decreases in spermine and spermidine, are recorded in the telencephalon but not hypothalamus upon systemic injection of bacteria. While systemic inflammation causes canonical behavioral fever in trout, blockade of bacterial polyamine metabolism prior to injection abrogates behavioral f …

The importance of the urea cycle and its relationships to polyamine metabolism during ammonium stress in Medicago truncatula.

The ornithine-urea cycle (urea cycle) makes a significant contribution to the metabolic responses of lower photosynthetic eukaryotes to episodes of high nitrogen availability. In this study, we compared the role of the plant urea cycle and its relationships to polyamine metabolism in ammonium-fed and nitrate-fed Medicago truncatula plants. High ammonium resulted in the accumulation of ammonium and pathway intermediates, particularly glutamine, arginine, ornithine, and putrescine. Arginine decarboxylase activity was decreased in roots, suggesting that the ornithine decarboxylase-dependent produ

Putrescine controls the formation of Escherichia coli persister cells tolerant to aminoglycoside netilmicin.

Persisters are suggested to be the products of a phenotypic variability that are quasi-dormant forms of regular bacterial cells highly tolerant to antibiotics. Our previous investigations revealed that a decrease in antibiotic tolerance of Escherichia coli cells could be reached through the inhibition of key enzymes of polyamine synthesis (putrescine, spermidine). We therefore assumed that polyamines could be involved in persister cell formation. Data obtained in our experiments with the polyamine-deficient E. coli strain demonstrate that the formation of persisters tolerant to netilmicin is h…

Polyamines modulate the binding of GABAA-benzodiazepine receptor ligands in membranes from the rat forebrain.

The effects of spermine, spermidine and putrescine on the binding of the GABAA-benzodiazepine receptor complex were examined in the hippocampus and frontal cortex membranes of the rat. The results demonstrated modulatory effects of polyamines on the binding of diazepam and flunitrazepam but not on that of GABA, muscimol and Ro 15-1788. When membranes were prepared without detergent, the polyamines enhanced the binding of diazepam. However, while the binding capacity increased after homogenization in the presence of the non-ionic detergent Triton X-100, the polyamines did not enhance the bindin…

Concomitantly elevated concentrations of the polyamines putrescine, spermidine and spermine reached a maximum on the first day after infection and then decreased. A model is presented to integrate these and other results to explain how RNA synthesis may be regulated during infection.

Effects of infection with Coxiella burnetii on synthesis of RNA in L cells.

Experimental infections with Coxiella burnetii augment rates of ribonucleic acid (RNA) synthesis in guinea pigs. The activity of deoxyribonucleic acid-dependent RNA polymerase in L cells persistently infected with C. burnetii was threefold greater than that in unifected cells; the polymerase activity in infected cells was predominantly of class I, whereas that in uninfected cells was predominantly of class II. A search for regulatory factors of polymerase activity revealed that preincubation of uninfected L cells with lipopolysaccharide of C. burnetii or with putrescine, spermidine, or spermin…

Pharmacological and dietary agents for colorectal cancer chemoprevention: effects on polyamine metabolism (review).

Chemoprevention is the long-term use of different chemical agents, both synthetic and natural, to prevent or delay the onset of disease. Since colorectal cancer has a significant environmental component, it is an ideal disease in which to evaluate the potential benefits of chemopreventive agents. The polyamines, spermine, spermidine and putrescine have been involved in almost all the steps of colorectal tumorigenesis. Consequently, polyamine biosynthesis and catabolism can be considered as promising targets for cancer chemoprevention. A variety of drug formulations have been tested for their e…

Assessing the polyamine metabolism of Plasmodium falciparum as chemotherapeutic target.

More than 30 years ago the potent ornithine decarboxylase inhibitor difluoromethylornithine (DFMO) was designed as new anticancer drug. Its efficacy was not as expected since the polyamine metabolism in mammalian cells seemed to be far more complex. However when DFMO was applied to African trypanosomes its effect on this protozoan parasite was highly convincing. Thenceforward many researchers tested DFMO and also other polyamine synthesis inhibitors against different parasites among them the causative agent of malaria Plasmodium. This review recapitulates the different attempts to interfere ch

[Effect of polyamines on actomyosine-ATP-ase activity in smooth muscles of the uterus].

The ability of the aliphatic polyamines to inhibit [figure: see text] the ATPase activity of smooth muscle actomyosine satisfies the succession: spermine > spermidine > putrescine that is correlated with magnitude of positive charge at physiological value of pH. The most effective inhibitor of the ATP hydrolysis process is the spermine, which highest inhibitory action is manifested at 10(-3) M concentration, in lesser concentration (10(-5) M) activates the actomyosine ATPase. While defining the kinetic parameters of the ATP hydrolysis reaction catalyzed by uterus myometrium the correlation bet…

Effect of alpha-difluoromethylornithine on rectal mucosal levels of polyamines in a randomized, double-blinded trial for colon cancer prevention.

BACKGROUND: Polyamines (e.g., putrescine, spermidine, and spermine) are required for optimal cell growth. Inhibition of polyamine synthesis suppresses carcinogen-induced epithelial cancers, including colon cancer, in animal models. In a short-term phase IIa trial, we determined that low doses of alpha-difluoromethylornithine (DFMO), an inhibitor of ornithine decarboxylase (an enzyme involved in polyamine synthesis), reduced the polyamine content of normal-appearing rectal mucosa of subjects with a prior history of resected colon polyps. In a follow-up study, we have attempted to determine the …

Exogenous Polyamines and Plant Salinity Tolerance: A Systematic Review, Evidence Map, and Family-Aware Quantitative Synthesis.

Salinity disrupts plant redox balance, ion homeostasis, and water relations, and exogenous polyamines have been widely investigated as potential mitigators of these effects. However, it remains unclear which polyamine-mediated responses are reproducible across independent studies and whether redox protection consistently translates into downstream physiological recovery. We therefore conducted an integrated systematic review, evidence map, and publication-family-aware quantitative synthesis to evaluate the effects of exogenous putrescine, spermidine, and spermine in salt-stressed plants. We sy…

Nitrogen waste metabolism as a locus of nitrergic co-transmission in the brain.

Nitrogen detoxification pathways in the central nervous system supply a range of neurotransmitters, ranging from long-appreciated examples like nitric oxide and agmatine, to emergent neurotransmitters including spermidine, spermine, and polyamine-derived GABA. This review summarizes specialized nitrogen detoxification pathways in the brain, and evidence supporting several of these pathways' metabolites as co-transmitters in neurons and glia. Known functional roles of these nitrergic co-transmitters in learning, sleep, addiction, and other neurological disorders will be discussed to elucidate t…
