Hereditary motor and sensory neuropathy-Ib (HMSN-Ib) is a common autosomal dominant disorder linked to the Duffy blood group locus on human chromosome 1. The gene for antithrombin III (AT3) is also located on the long arm of chromosome 1. Using a DNA restriction fragment length polymorphism for AT3, we have investigated the genetic linkage relationship of all three markers (HMSN-Ib, Duffy, and AT3) in two affected families. Neither HMSN-Ib nor Duffy was tightly linked to AT3. The loci for both HMSN-Ib and Duffy must be close to the centromere on chromosome 1, but precise localization and gene

The neuronal type of Charcot-Marie-Tooth disease (hereditary motor and sensory neuropathy, type II) was found to be present in ten of thirty-two members of one family. Motor nerve conduction velocities in this type of the disease may be normal in persons who are only mildly affected. Hoffmann-reflex investigation was effective in discriminating between affected and non-affected subjects. The mode of inheritance seemed to be X-linked, but this remained doubtful, because the outcome of the Hoffmann-reflex investigations was abnormal in one clinically non-affected boy. Xg(a)-typing gave no inform

A study of 7 cases of Charcot-Marie-Tooth disease associated with a dyskinesia resembling benign essential tremor is presented. In 4 patients, the family history strongly suggested an autosomal mode of transmission, 2 cases were sporadic without an established genetic pattern and 1 was probably recessive. The distal parts of the upper and lower limbs showed imparied muscle strength with slight or no atrophy in 4 patients and conspicuous weakness and wasting in another 2. One patient was a chairbound. Although essential tremor and the tremor seen in these patients are clinically (phenotypically

To investigate whether preterm birth (PTB) and extreme birth weights are associated with the prevalence of periodontal disease and tooth loss later in life, we conducted a cross-sectional analysis of 6152 adults (62.3 ± 5.6 years) from the Atherosclerosis Risk in Communities (ARIC) study, who underwent a periodontal examination at Visit 4 (1996-1998). Preterm birth (PTB) (< 37 weeks) and birth weight categories (low birth weight (LBW) < 2.5 kg and high birth weight (HBW) > 4.0 kg) were self‑reported. Periodontal status was assessed via clinical examination and classified using the Periodontal

BACKGROUND: PMP22 duplications cause Charcot-Marie-Tooth disease (CMT), whereas PMP22 deletion results in hereditary neuropathy with liability to pressure palsies (HNPP). Although PMP22 point mutations can cause a spectrum of neuropathies, including hereditary motor and sensory neuropathy (HMSN) I, congenital hypomyelinating neuropathy, HMSN III, and HNPP, they are rare, and their features in Japanese patients have not been well characterized. Therefore, we compared the genetic, clinical, and electrophysiological characteristics of patients with PMP22 point mutations.
METHODS: This multicente

INTRODUCTION: There is a paucity of nationwide population-based studies exploring the associations between chronic conditions and oral health. This study was conducted to describe the sociodemographic characteristics of a sample of Australian dentate adults with chronic conditions and to ascertain the associations of chronic conditions with tooth loss, dental caries and periodontal disease.
METHODS: Data were drawn from the National Study of Adult Oral Health (NSAOH) 2017-18, which included a random sample of Australians aged 15+ years. NSAOH used online/telephone interviews to elicit self-re

Charcot-Marie-Tooth (CMT) disease is the most common hereditary motor and sensory neuropathy. While diagnosis is typically established through clinical findings, nerve conduction studies (NCS), and genetic testing, imaging manifestations such as nerve root hypertrophy are increasingly recognized. This case report presents a 43-year-old man with lifelong progressive distal weakness who presented with severe lower extremity atrophy, pes cavus deformities, and wheelchair dependence. Neurologic examination revealed marked distal weakness and hyporeflexia, while nerve conduction studies demonstrate

BACKGROUND AND OBJECTIVES: Our aim was to report a novel variant of Guanine nucleotide-binding protein subunit beta-4 gene (GNB4) variants and to report the clinical and molecular spectrum of the GNB4-related neuropathy based on the currently reported cases.
METHODS: We performed detailed neurologic and electrophysiologic assessments and segregation analysis in a patient form single family with Charcot-Marie-Tooth (CMT) disease with novel GNB4 variant. Via literature search, we found studies that included neuropathy cases with GNB4 variants; we then reviewed and reclassified these variants in

INTRODUCTION: Charcot-Marie-Tooth disease (CMT) understanding and diagnostic rates are improving. Symptomatic management is still the only option, but many therapeutic approaches are under investigation, some in the clinical trial phase.
AREAS COVERED: Through a comprehensive search in PubMed, the ClinicalTrials.gov website, and the latest abstracts on the topic, the authors review the diagnostic advances and promising treatments, focusing on pharmacological and gene therapy/silencing approaches, and on clinical trial challenges. They also review current CMT management, including rehabilitati

BACKGROUND: Orthopaedic surgical intervention in children with Charcot-Marie-Tooth (CMT) often includes triceps surae lengthening (TSL) and foot procedures to address instability and pain due to equinus and cavovarus deformities. These surgeries may unmask underlying weakness in this progressive disease causing increased calcaneal pitch and excessive dorsiflexion in terminal stance leading to crouch. The purpose of this study was to evaluate changes in ankle function during gait following TSL surgery in children with CMT.
METHODS: Nineteen participants (11 male; age 12.8, SD 3.6 y) with CMT

Charcot-Marie-Tooth is an inherited disorder involving multiple genes, causing progressive nerve damage affecting sensation and movement. The complexity of the condition often leads to various possible diagnoses along with neuropathic diseases, sometimes resulting in significant delays in diagnosis and treatment. Thorough clinical examinations, suspicion based on symptoms, electromyography, nerve conduction tests, and specific genomic testing can expedite diagnosis. Here, we will introduce a case of Charcot-Marie-Tooth disorder, initially presenting with stridor and hoarseness due to vocal cor

Charcot-Marie-Tooth disease (CMT), an inherited peripheral nervous system disorder, causes muscle weakness, sensory deficits, decreased or absent tendon reflexes, and skeletal deformities. Although primarily a peripheral disorder, some studies indicate central nervous system (CNS) involvement. This study systematically investigated the potential structural and functional brain alterations in patients with CMT without overt CNS symptoms using magnetic resonance imaging (MRI) techniques. In this prospective cross-sectional study, 14 patients with clinically and genetically confirmed CMT and 14 a

Mutations in myelin protein zero (MPZ) are generally associated with Charcot-Marie-Tooth type 1B (CMT1B) disease, one of the most common forms of demyelinating neuropathy. Pathogenesis of some MPZ mutants, such as S63del and R98C, involves the misfolding and retention of MPZ in the endoplasmic reticulum (ER) of myelinating Schwann cells. To cope with proteotoxic ER stress, Schwann cells mount an unfolded protein response (UPR) characterized by activation of the PERK, ATF6 and IRE1α/XBP1 pathways. Previous results showed that targeting the PERK UPR pathway mitigates neuropathy in mouse models o

BACKGROUND AND PURPOSE: X-linked Charcot-Marie-Tooth disease type 1 (CMTX1) ranks as the second most prevalent hereditary neuropathy and, currently, has no definitive cure. Emerging preclinical trials offer hope for potential clinical studies in the near future. While it is widely accepted that experimental groups in these trials should be balanced for age and gender, there is a current shortfall in data regarding phenotype-genotype correlations. Our aim was to provide a more detailed understanding of these correlations to facilitate the formation of well-matched patient groups in upcoming cli
