ETHNOPHARMACOLOGICAL RELEVANCE: In tropical nations like India, the herb Costus pictus, belonging to the family Costaceae, is widely distributed. Because of its many therapeutic benefits, C. pictus is used extensively by different ethnic communities. In addition to its possible antibacterial, anti-inflammatory, and antioxidant qualities, C. pictus leaf powder has long been used to treat diabetes. Even though C. pictus leaves have several therapeutic uses and are widely used in traditional medicine; there is no empirical evidence of their efficacy or toxicity.
AIMS OF THE STUDY: This investiga

Early-life stage (ELS) toxicity tests are recognized as an advancement over current testing methodologies in terms of cost, animal use, and biological relevance. However, standardized ELS tests are not presently available for some vertebrate taxa, including birds. The present study describes a Japanese quail (Coturnix japonica) ELS test that is a promising candidate for standardization and applies it to test 8 environmental chemicals (ethinylestradiol, benzo[a]pyrene, chlorpyrifos, fluoxetine, lead(II)nitrate, trenbolone, seleno-L-methionine, hexabromocyclododecane). Individual chemicals were …

Over the past 20 years or more, investigators have been developing non-animal test methods for use in assessing the skin sensitisation potential of chemicals. In parallel with this effort, the key biological events of skin sensitisation have been well-characterised in an Adverse Outcome Pathway (AOP) proposed by the Organisation for Economic Co-operation and Development (OECD). The key molecular initiating event of this AOP is haptenation or covalent modification of epidermal proteins. In this review, the strengths and limitations of the Direct Peptide Reactivity Assay (DPRA) are described, an

UNLABELLED: As a general practice in rodent toxicology studies, satellite animals are used for toxicokinetic determinations, because of the potential impact of serial blood sampling on toxicological endpoints. Besides toxicological and toxicokinetic determinations, blood samples obtained longitudinally from a same animal may be used for the assessment of additional parameters (e.g., metabolism, pharmacodynamics, safety biomarkers) to maximize information that can be deduced from rodents. We investigated whether removal of up to 6 × 200 μL of blood over 24h can be applied in GLP rat toxicology

Quantitative structure-toxicity relationships (QSTR's) are derived for an extensive series of halogenated benzenes, anilines, phenols, nitrobenzenes, toluenes and other substituted benzenes against Vibrio fischeri using a wide range of whole molecule and atom-based descriptors derived from semi-empirical molecular-orbital calculations. In terms of direct statistical correlation with toxicity it was found that the molar refractivity was the most important parameter, closely followed by the solvent accessible surface area of the compound. The accuracy of these descriptors in fitting the numerous

ETHNOPHARMACOLOGICAL RELEVANCE: Petiveria alliacea L. (Phytolaccaceae) is used in folk medicine due to its antispasmodic, diuretic, hypoglycemic, abortive, anti-inflammatory and anticancerogenic properties. Although P. alliacea is considered toxic by people, its toxicity remains a concern since it is strongly dependent on the extraction method and the part of the plant used during tests. Even if some healers prefer to use the aerial parts in a liquefied form or by chewing them, instead of decoctions or infusions, no toxicological studies exist using whole dried stems and leaves.

Sediment toxicity tests in the laboratory are an important part of ecological risk assessments, yet how they relate to sediment toxicity in situ has rarely been explored. Using meta-analysis, we examined differences in the toxicity of sediment tested in the laboratory and in situ. Data from four published studies were subjected to rigorous statistical analyses. Overall, the toxicity of sediments in laboratory tests was substantially less than their toxicity in situ. Differences between laboratory and in situ toxicity, expressed using the log odds ratio effect size, varied significantly among p…

The effects of exposure to environmental tobacco smoke on pulmonary function were assessed in 21 subjects with asthma who claimed respiratory complaints (cough, shortness of breath, and chest tightness) on previous exposure to cigarette smoke. Exposure to mechanically produced tobacco smoke was performed in a static inhalation chamber for two-hour intervals at two distinct smoke levels (as measured by carbon monoxide, nicotine, and particulate levels). Seven of the 21 smoke-challenged subjects experienced a significant (greater than 20%) decline in FEV1 during passive exposure to tobacco smoke

High salmonid toxicity of 6PPD-quinone, a transformation product of the tire antioxidant 6PPD, has accelerated the search for safer tire alternatives. We evaluated the toxicity of the quinone transformation products of two candidate alternatives, 7PPD-quinone (7PPD-Q) and 77PD-quinone (77PD-Q), in zebrafish (Danio rerio) early life stages. Embryos and sac-fry were exposed to these chemicals under OECD Test Guideline 212 conditions. Endpoints included 8-day survival, hatching, heartbeat, eye and head sizes, body length, morphological deformities, and yolk-sac pigmentation. The results showed th

The current approach to qualify NMIs (non-mutagenic impurities) exceeding the qualification threshold is to perform genotoxic, general toxicity and other specific studies in animals. Furthermore, these studies are rarely conducted on clean impurity samples. As a result, any toxicity identified in safety tests cannot be unambiguously attributed to the impurity alone, and hence the lack of impurity-specific safety data prevents results from being extrapolated to conditions such as increased impurity levels in the same drug product during shelf life. That is, animal safety tests would often need

Pharmaceuticals are a class of emerging organic contaminants found in freshwaters. Potential chronic effects of many legacy drugs (those developed pre-2006) on freshwater biota were not investigated thoroughly before their market release. Carbamazepine, a drug used to treat epilepsy and certain types of neuropathic pain, is a legacy contaminant commonly detected in surface freshwaters worldwide, yet its potential impacts on aquatic invertebrates are largely unknown. Our study investigated the chronic effects (21-d exposure) of environmentally relevant concentrations of carbamazepine (measured

Assessment and characterization of bone toxicity during drug development is important to ensure the safety of new therapeutics. Drugs can affect bone composition and quality either directly on bone cells or indirectly via systemic effects, leading to alterations in bone density, remodeling, and fracture risk. Drug classes known to have harmful effects on bone include antidiabetics, non-steroidal anti-inflammatory drugs, antivirals, chemotherapeutics, and steroids. Various methods are available to assess and investigate bone toxicity, including in vivo animal models, ex vivo organ cultures, and

Ocular toxicity studies may encompass a variety of ocular routes of administration. Because the retina and optic nerve are an extension of the central nervous system, safety assessment of ocular compounds must include thorough examination of extraocular visual pathways, as ocular administration may result in primary toxicities in the nervous system due to its exposure to therapeutics via the retina and optic nerve, or changes that are secondary to primary ocular effects. Here we provide a practical guide to sampling extraocular visual pathway structures for commonly utilized ocular toxicology
