A simple classification method for residual antibiotics using E. coli cells transformed by the calcium chloride method and drug resistance plasmid DNA.

Using three different plasmid DNA codings for kanamycin (KM), chloramphenicol (CP), and ampicillin- (AMP) and tetracycline- (TC) resistance, four different competent Escherichia coli strains were transformed by the calcium chloride method to produce KM-, CP- and AMP- and TC-resistant strains. Evaluation of minimum inhibitory concentrations (MIC) of 22 antibiotics, showed KM-resistant E. coli to be cross resistant only to fradiomycin (FRM); CP-resistant E. coli, especially HB101 and JM109 strains, exhibited cross-resistance only to thiamphenicol (TP). On the other hand, AMP- and TC-resistant E.

A gene controlling segregation of the Bacillus subtilis plasmid pC194.

Plasmid pC194-1, a mutant of pC194, and chimeric derivatives of pC194-1 are segregationally unstable in B. subtilis. Such instability could be enhanced by exposure of pC194-1-carrying cells to methyl methanesulfonate. pC194-1 is distinct from pC194 in the addition of two A:T base pairs within the previously defined D region of pC194. Complementation experiments between pC194-1 and other plasmids suggest that the mutation of pC194-1 interferes with the production of a diffusible gene product required for plasmid maintenance.

Helicobacter pylori and gastroduodenal pathology: new threats of the old friend.

The human gastric pathogen Helicobacter pylori causes chronic gastritis, peptic ulcer disease, gastric carcinoma, and mucosa-associated lymphoid tissue (MALT) lymphoma. It infects over 50% of the worlds' population, however, only a small subset of infected people experience H. pylori-associated illnesses. Associations with disease-specific factors remain enigmatic years after the genome sequences were deciphered. Infection with strains of Helicobacter pylori that carry the cytotoxin-associated antigen A (cagA) gene is associated with gastric carcinoma. Recent studies revealed mechanisms throug

Impact of Fermentation on Bacterial and Fungal Microbiome Interactions in Pla-Ra, a Traditional Thai Food.

Pla-ra, a traditional Thai fermented fish product, undergoes complex microbial and biochemical transformations that contribute to its unique flavor and aroma. This study investigated the effects of fermentation on the microbiome of pla-ra, focusing on bacteria-fungi interactions. Freshwater fish, combined with salt and roasted rice, were sampled after 1 and 6 months of fermentation. Bacterial and fungal communities in these pla-ra samples were characterized using next-generation sequencing of the 16S rRNA (V3-V4 region) and ITS2 regions, respectively. Results demonstrated that initial bacteria

Antibacterial efficacy of iron oxide and silver nanoparticles against bacterial wilt pathogen Ralstonia solanacearum.

The security of vegetable plants worldwide is threatened by bacterial wilts, one of the most infectious soil-borne bacterial plant diseases. This is caused by R. Solanacearum. Overuse of bactericides and antibiotics to combat bacterial wilt results in pesticide resistance and toxicity to beneficial living organisms. Consequently, nanoparticles are more beneficial, safe for the environment, and have strong antibacterial properties than conventional pesticides. In the present work, iron oxide nanoparticles (IONPs) and silver nanoparticles (AgNPs) were prepared by simple chemical, eco-friendly pr

Exposure to bacterial cell wall products triggers an inflammatory phenotype in hepatic stellate cells.

Activated hepatic stellate cells (HSCs) secrete extracellular matrix components during hepatic fibrosis, but recent studies have shown that HSCs can also release a variety of proinflammatory cytokines. Moreover, bacterial endotoxemia is not only associated with systemic complications in the late stages of liver failure but is also a direct cause of liver damage, activating resident inflammatory cells. In this study, we investigated whether HSCs can respond directly to bacterial cell wall products acquiring a new phenotype. RT-PCR and immunocytochemistry assays were used to show that murine HSC

[Cloning of Mycoplasma gallisepticum Genome in Saccharomyces cerevisiae Cells].

To date, the genomes of several mycoplasmas have been cloned in Saccharomyces cerevisiae cells, including Mycoplasma genitalium, M. pneumoniae, and M. mycoides. In this work, the 1-Mb genome of Mycoplasma gallisepticum S6 was cloned in S. cerevisiae. A transposon vector was constructed to deliver a synthetic S. cerevisiae replicon and a selective marker into M. gallisepticum genome. Transplantation of the M. gallisepticum genome with the integrated yeast replicon to yeast cells was performed using two approaches, polyethylene glycol (PEG)-induced transformation of yeast spheroplasts with intac

Fabrication of antibacterial poly (L-lactic acid)/tea polyphenol blend films via reactive blending using SG copolymer.

Poly (L-lactic acid) (PLLA) composite materials with both excellent antibacterial properties and mechanical properties are highly desirable for both food packaging and biomedical applications. However, a facile method to prepare transparent PLLA composite films with both excellent antibacterial and mechanical properties is still lacking. In this work, blend films based on PLLA, tea polyphenols (TP) and poly (styrene-co-glycidyl methacrylate) (SG) copolymers (PLLA/TP/SG) were prepared by melt blending using twin screw extruder. The blend films showed high transparency with a brownish color orig

New Semisynthetic Penicillins Obtained by Coupling of the 6-Aminopenicillanic Acid with 5-Mercapto-1,2,4-triazoles-3,4-disubstituted.

In a basic medium, 5-Mercapto-1,2,4-triazoles pass into the thiol form, allowing their transformation into sodium salts, which, in reaction with sodium monochloroacetate, lead to sodium 5-thioacetates of 1,2,4-triazoles-3,4-disubstituted. Sulfur derivatives converted to pivalic mixed anhydrides were used as active forms in the acylation of 6-amino penicillanic acid (6-AP) to obtain new semisynthetic penicillins. They contain in the molecule, together with the β-lactam ring, the nucleus 3-[(5-nitroindazol-1'-yl-methyl)]-4-aryl-5-mercapto-1,2,4-triazole, both contributing to an important antibac

[Tracing the past half century of my microbial transformation studies].

In the middle of 1950's, microbial transformation technology was introduced into the field of synthetic chemistry as a new methodology. There was a sudden interest in research on the problems of producing steroid hormones by microbial transformation. At that time, the first project entitled "The Study for Microbial Transformation of Steroids", the "Tsuda Project", was established in the Institute of Applied Microbiology (IAM), University of Tokyo, in the spring 1956, in which I took part. This paper summarizes a number of results of our microbial transformation reactions not only in the synthe

Genetic and physical studies of restriction-deficient mutants of the Inc FIV plasmids R124 and R124/3.

R124 and R124/3 are R plasmids that carry the genes for two different restriction and modification systems. The phenotype of strains carrying either of these plasmids along with the F'lac+ plasmid, is restriction-deficient (Res-). The Res- phenotype is not due to selection of pre-existing mutants but rather to a complex mutational event caused by the F plasmid. Restriction-deficient mutants carry extensive deletions and other DNA rearrangements. Tn7 insertion is used to locate the restriction gene. Many of the Res- mutants are genetically unstable and revert at exceptionally high frequencies.

Engineering of Salmonella Phages into Novel Antimicrobial Tailocins.

Due to the extensive use of antibiotics, the increase of infections caused by antibiotic-resistant bacteria is now a global health concern. Phages have proven useful for treating bacterial infections and represent a promising alternative or complement to antibiotic treatment. Yet, other alternatives exist, such as bacteria-produced non-replicative protein complexes that can kill their targeted bacteria by puncturing their membrane (Tailocins). To expand the repertoire of Tailocins available, we suggest a new approach that transforms phages into Tailocins. Here, we genetically engineered the vi

Proteolytic Landscapes in Gastric Pathology and Cancerogenesis.

Gastric cancer is a leading cause of cancer-related death, and a large proportion of cases are inseparably linked to infections with the bacterial pathogen and type I carcinogen Helicobacter pylori. The development of gastric cancer follows a cascade of transformative tissue events in an inflammatory environment. Proteases of host origin as well as H. pylori-derived proteases contribute to disease progression at every stage, from chronic gastritis to gastric cancer. In the present article, we discuss the importance of (metallo-)proteases in colonization, epithelial inflammation, and barrier di

Targeted insertion of the neomycin phosphotransferase gene into the tubulin gene cluster of Trypanosoma brucei.

Kinetoplastids are unicellular eukaryotes that include important parasites of man, such as trypanosomes and leishmanias. The study of these organisms received a recent boost from the development of transient transformation allowing the short-term expression of genes reintroduced into parasites like Trypanosoma brucei, the causative agent of African trypanosomiasis. We have obtained long-term stable transformants of T. brucei that have acquired the ability to grow in medium containing the drug G418, following the targeted insertion of the bacterial gene for neomycin phosphotransferase (neo(r) g

Production and Structural/Physicochemical Characteristics of Exopolysaccharides from the Halotolerant Strain Oceanobacillus sp. DT-Z6.

Halotolerant strains isolated from extreme environments rely on exopolysaccharides (EPSs) to maintain cellular integrity under high salinity and ensure survival under harsh conditions. Although continuous screening efforts have identified numerous EPS-producing halotolerant strains, research on the structural characteristics and physicochemical properties of their EPSs remains limited. In this study, an EPS-producing strain yielding 2.59 ± 0.15 g/L was isolated from saline soil and identified as Oceanobacillus sp. DT-Z6. The optimal conditions for key fermentation parameters were determined us

Development of Rhizobium rhizogenes-induced hairy root cultures of Vitex negundo for vitexin production.

Vitex negundo L. is a significant indigenous medicinal plant with diverse therapeutic potentials owing to its rich reservoir of bioactive compounds including sitosterol, vitexoside, agnuside, isovitexin and vitexin which are predominantly present in the root. To induce hairy roots, explants of V. negundo L. were transformed using two strains of Rhizobium rhizogenes: MTCC 532 and MTCC 2364. Among them, the MTCC 2364 strain exhibited the highest transformation efficiency (94.44%). Genetic transformation of the induced hairy roots was confirmed by PCR amplification using primers specific for the

Transformation of Anaplasma phagocytophilum.

BACKGROUND: Tick-borne pathogens cause emerging zoonoses, and include fastidious organisms such as Anaplasma phagocytophilum. Because of their obligate intracellular nature, methods for mutagenesis and transformation have not been available. RESULTS: To facilitate genetic manipulation, we transformed A. phagocytophilum (Ap) to express a green fluorescent protein (GFP) with the Himar1 transposase system and selection with the clinically irrelevant antibiotic spectinomycin. CONCLUSION: These transformed bacteria (GFP/Ap) grow at normal rates and are brightly fluorescent in human, monkey, and t
