Relation of epicardial fat thickness and brachial flow-mediated vasodilation with coronary artery disease.

OBJECTIVE: The purpose of this study is to investigate the presence of a statistical association between epicardial fat thickness (EFT) and coronary artery disease (CAD) and between flow-mediated vasodilation (FMD) and CAD. METHODS: We measured the EFT and FMD in 64 subjects with suspected stable angina pectoris. The patients were separated into two groups according to their coronary angiography results: 34 patients with CAD and 30 patients with normal coronary arteries (NCA). RESULTS: EFT was significantly higher in the patients with CAD than the NCA group (6.43 ± 0.90 mm vs. 5.35 ± 0.75 mm…

Acyloxy nitroso compounds as nitroxyl (HNO) donors: kinetics, reactions with thiols, and vasodilation properties.

Acyloxy nitroso compounds hydrolyze to nitroxyl (HNO), a nitrogen monoxide with distinct chemistry and biology. Ultraviolet-visible spectroscopy and mass spectrometry show hydrolysis rate depends on pH and ester group structure with the observed rate being trifluoroacetate (3) > acetate (1) > pivalate (2). Under all conditions, 3 rapidly hydrolyzes to HNO. A combination of spectroscopic, kinetic, and product studies show that addition of thiols increases the decomposition rate of 1 and 2, leading to hydrolysis and HNO. Under conditions that favor thiolates, the thiolate directly reacts with th

Role of nitrosyl factors in the hindlimb vasodilation elicited by baroreceptor afferent nerve stimulation.

This study determined whether electrical stimulation (ES) of the baroreceptor afferent fibers in the aortic depressor nerve (ADN) produces hindlimb vasodilation in pentobarbital-anesthetized rats via the release of nitric oxide (NO)-containing (nitrosyl) factors from NO synthase-positive lumbar sympathetic nerve terminals. ES of the ADN (1-10 Hz for 15 s) produced frequency-dependent reductions in mean arterial blood pressure (MAP) and mesenteric and hindlimb vascular resistance (MR and HLR, respectively). The falls in resistance were substantially smaller in hindlimb beds in which the ipsilat…

Low-renin hypertension with relative aldosterone excess is associated with impaired NO-mediated vasodilation.

Recent studies suggest that hypertension associated with low renin status and hyperaldosteronism is associated with increased risk for end-o … onstrates that low-renin hypertension is associated with marked impairment of nitric oxide-mediated vasodilation of resistance vessels in the forearm vasculature of humans. This impairment could contribute to adverse outcomes in patients with low-renin hypertension and relative aldosterone excess.

Haldane, hot dogs, halitosis, and hypoxic vasodilation: the emerging biology of the nitrite anion.

While it has long been known that the reduction of nitrite to nitric oxide (NO) forms iron-nitrosyl-myoglobin and is the basis of meat curing, a greater biological activity of the nitrite anion has only recently been appreciated. In the stomach, NO is formed from acidic reduction of nitrite and increases mucous barrier thickness and gastric blood flow (see the related study beginning on page 106). Nitrite levels in blood reflect NO production from endothelial NO synthase enzymes, and recent data suggest that nitrite contributes to blood flow regulation by reaction with deoxygenated hemoglobin

Impaired endothelium-mediated vasodilation in heart failure: clinical evidence and the potential for therapy.

Numerous studies in the last decade have clearly shown an attenuated endothelium-dependent vasodilation in patients with chronic heart failure. This abnormality has been demonstrated in the peripheral, pulmonary, and coronary circulation in patients with both ischemic and nonischemic cardiomyopathy; its magnitude correlates with the severity of symptoms. Endothelial dysfunction in patients with cardiomyopathy and a relatively new onset of symptoms suggests that change in endothelial function occurs early in the course of the disease. In contrast to other circulatory beds, renal circulation has…

Role of EDHF in conduction of vasodilation along hamster cheek pouch arterioles in vivo.

We tested whether local and conducted responses to ACh depend on factors released from endothelial cells (EC) in cheek pouch arterioles of anesthetized hamsters. ACh was delivered from a micropipette (1 s, 500 nA), while arteriolar diameter (rest, approximately 40 microm) was monitored at the site of application (local) and at 520 and 1,040 microm upstream (conducted). Under control conditions, ACh elicited local (22-65 microm) and conducted (14-44 microm) vasodilation. Indomethacin (10 microM) had no effect, whereas N(omega)-nitro-L-arginine (100 microM) reduced local and conducted vasodilati…

However, the combined results showed an exponential increase in 10(-5) M Ach-induced vasodilation as liver copper concentration increases from 0 microg Cu/g dry wt. Role of smooth muscle activation in conduction of vasodilation along isolated hamster feed arteries.

Vasodilation originating within a muscle is conducted into feed arteries, but the determinants of conduction along feed arteries are undefined. We have investigated the effect of vasomotor tone on conducted vasodilation in feed arteries of the hamster retractor muscle. Unbranched segments (length, 2-3 mm) were isolated, cannulated, and studied in vitro at 37 degreesC while pressurized to 25 mm Hg, 75 mm Hg (in vivo pressure), or 120 mm Hg in the absence and presence of norepinephrine (NE, 10(-7) M + 10(-6) M propanolol). Effect of nitric oxide inhibition on capsaicin-elicited vasodilation in the rat oral circulation.

The effects of local application of capsaicin on the vascular conductance of the oral structures (upper gingiva, lower gingiva, tongue, right and left submandibular glands) were studied with and without pretreatment with NG-nitro-L-arginine methyl ester (L-NAME), a potent inhibitor of nitric oxide formation in rats. Alterations in tissue blood flow were measured by laser-Doppler-flowmetry; systemic blood pressure was monitored continuously during the experiments. Local application of increasing concentrations of capsaicin (1.0; 3.33; 10.0; 33.3 mM; in a volume of 5 microliters) resulted in a d

Impaired endothelium-mediated vasodilation in the peripheral vasculature of patients with congestive heart failure.

Impaired endothelial-dependent vasodilation has been demonstrated in two animal models of congestive heart failure and in the coronary circulation of patients with idiopathic dilated cardiomyopathy. To determine whether this impairment contributes to the abnormal peripheral vasomotor tone in patients with congestive heart failure, the local vascular response to intraarterial infusions of graded concentrations (10(-8) M to 10(-5) M) of acetylcholine (an endothelial-dependent vasodilator) and nitroglycerin (a direct-acting vasodilator) was studied in the superficial femoral artery of 19 patients…

Nerve-induced tachykinin-mediated vasodilation in skeletal muscle is dependent on nitric oxide formation.

Nerve-induced vasodilatation was studied by intravital microscopy of the rabbit tenuissimus muscle, pretreated with pancuronium, phentolamine, and guanethidine. Nerve stimulation of the tenuissimus nerve induced a vasodilatation which was frequency and pulse duration-dependent and insensitive to atropine and propanolol but abolished by tetrodotoxin. The nitric oxide synthase inhibitor, N omega-nitro-L-arginine methyl ester (L-NAME, 100 microM), but not its enantiomer, D-NAME, markedly inhibited the vasodilation induced by nerve stimulation or by exogenous substance P or neurokinin A. Vasodilat…

H(+)-induced vasodilation of rat aorta is mediated by alterations in intracellular calcium sequestration.

Acidosis induces vasodilation both in vivo and in vitro. Although it is commonly surmised that acidosis alters contractility by affecting contractile proteins and calcium entry, the exact role of these mechanisms in acidosis-induced vasodilation has not been determined. In the present study, we demonstrated that a novel mechanism, involving increased calcium sequestration into intracellular sites sensitive to norepinephrine, mediates the vasodilation associated with relatively modest decreases in pH. The effects of changing pH from 7.4 to 7.0 on tension development, 45Ca fluxes, and the norepi…

Simultaneous determination of brachial and femoral arterial pressures during reactive hyperemia and papaverine vasodilation.

Accurate assessment of aortoiliac occlusive disease often requires direct intraarterial pressure determination. Since these measurements may alter systemic pressure, brachial arterial pressure (BAP) and femoral arterial pressure (FAP) were obtained simultaneously to quantitate these changes. BAP and FAP were measured at rest, and then during vasodilation produced by postischemic reactive hyperemia and intra-arterial injection of papaverine. The gradient between FAP and BAP was used to assess the significance of an aortoiliac stenosis. Sixty-eight observations were performed in 19 limbs. During…
