Plasma Phospholipid Fatty Acids, FADS1 and Risk of 15 Cardiovascular Diseases: A Mendelian Randomisation Study.

Whether circulating fatty acids (FAs) play a causal role in the development of cardiovascular disease (CVD) remains unclear. We conducted a Mendelian randomisation study to explore the associations between plasma phospholipid FA levels and 15 CVDs. Summary-level data from the CARDIoGRAMplusC4D, MEGASTROKE, and Atrial Fibrillation consortia and UK Biobank were used. Sixteen single-nucleotide polymorphisms (SNPs) associated with ten plasma FAs were used as instrumental variables. SNPs in or close to the FADS1 gene were associated with most FAs. We performed a secondary analysis of the associatio

Antioxidants and fatty acids in the amelioration of rheumatoid arthritis and related disorders.

The generation of reactive oxygen species (free radicals) is an important factor in the development and maintenance of rheumatoid arthritis in humans and animal models. One source of free radicals is nitric oxide produced within the synoviocytes and chondrocytes and giving rise to the highly toxic radical peroxynitrite. Several cytokines, including tumour necrosis factor-alpha (TNFalpha) are involved in the formation of free radicals, partly by increasing the activity of nitric oxide synthase. Indeed, nitric oxide may mediate some of the deleterious effects of cytokines on bone resorption. Asp

Nutrient assessment of sea buckthorn residues as potential feed ingredients.

This study aimed to evaluate the suitability of sea buckthorn by-products, including pomace without seeds (SBPW), pomace with seeds (SBPS), and leaves (SBL) as potential livestock feed ingredients. Proximate composition, amino acids, fatty acids, minerals, vitamins, and metabolizable energy (ME) were assessed and compared to the daily requirements of livestock, assuming an inclusion rate of 2.5% of dry matter intake in different animal species. SBPW exhibited the highest metabolizable energy (13.7-17 MJ/kg DM across species), exceeding 2.5% of daily requirements in most animal species, suggest

Human cerebrospinal fluid fatty acid levels differ between supernatant fluid and brain-derived nanoparticle fractions, and are altered in Alzheimer's disease.

BACKGROUND: Although saturated (SAFA), monounsaturated (MUFA), and polyunsaturated (PUFA) fatty acids are important structural components of neuronal membranes and precursors of signaling molecules, knowledge of their metabolism in Alzheimer's disease (AD) is limited. Based on recent discovery that lipids in cerebrospinal fluid (CSF) are distributed in both brain-derived nanoparticles (NP) and supernatant fluid (SF), we hypothesized that fatty acid (FA) abundance and distribution into these compartments is altered in early AD pathology. METHODOLOGY AND FINDINGS: We assayed the FA composition

[Impact of controlling risk factors after myocardial infarction].

The relative risks of each factors and the benefits of their reduction after myocardial infarction are comparable to those observed in primary prevention. However, because of an overexposure to the risk, the absolute gains are five times greater. The impact of diet is one of the most important: in addition to the limitation of polyunsaturated fats and global calory intake, especially in cases of central obesity, the increase in dietary alpha-linolenic acid and in omega-3, has been shown to reduce the risk of myocardial infarction and mortality by up to 70%. Supplements of vitamins A and E coul…

Reduced intestinal FADS1 gene expression and plasma omega-3 fatty acids following Roux-en-Y gastric bypass.

BACKGROUND & AIMS: Roux-en-Y gastric bypass (RYGB) limits food ingestion and may alter the intestinal expression of genes involved in the endogenous synthesis of polyunsaturated fatty acids (PUFAs). These changes may decrease the systemic availability of bioactive PUFAs after RYGB. To study the impact of RYGB on the dietary ingestion and plasma concentration of PUFAs and on the intestinal expression of genes involved in their endogenous biosynthesis in severely obese women with type 2 diabetes. METHODS: Before, and 3 and 12 months after RYGB, obese women (n = 20) self-reported a seven-day die

Allosteric activation of a cell-type-specific GPR120 inhibits amyloid pathology of Alzheimer's disease.

Black rice diets are enriched with unsaturated fatty acids that are thought to be beneficial for neurodegenerative disorders in aging. Here we find that α-linolenic acid (ALA) and 11,14-eicosadienoic acid (EDA), which are naturally enriched in black rice, inhibit amyloid pathology, rescue cognition and extend lifespan in mouse preclinical models of Alzheimer's disease via allosteric activation of G protein-coupled receptor 120 (GPR120) in plaque-associated macrophages and activated microglia. We generate the structures of GPR120 bound to ALA and EDA. We demonstrate that ALA and EDA allosterica

Nutritional Imbalance between Omega-6 and Omega-3 Polyunsaturated Fatty Acids during Pregnancy Increases the Number of Pyramidal Neurons in the Basolateral Amygdala and Anxiety-Related Behavior in Offspring.

Modern agriculture allows for the production of foods that are high in n-6 linoleic acid and low in n-3 α-linolenic acid (LAhigh/ALAlow), which are suggested to be associated with an increased risk for the onset of anxiety disorders. However, there is not sufficient evidence to understand its underlying brain mechanism. Given that mouse offspring derived from mothers fed a LAhigh/ALAlow diet during gestation and early lactation showed increased anxiety-related behaviors and that rodents exposed to a LAhigh/ALAlow diet are more vulnerable to stress, in this study, we investigated the effects of

Influence of Maternal Obesity and Gestational Weight Gain on Maternal and Foetal Lipid Profile.

Fatty acids (FAs) are fundamental for a foetus's growth, serving as an energy source, structural constituents of cellular membranes and precursors of bioactive molecules, as well as being essential for cell signalling. Long-chain polyunsaturated FAs (LC-PUFAs) are pivotal in brain and visual development. It is of interest to investigate whether and how specific pregnancy conditions, which alter fatty acid metabolism (excessive pre-pregnancy body mass index (BMI) or gestational weight gain (GWG)), affect lipid supply to the foetus. For this purpose, we evaluated the erythrocyte FAs of mothers a

Fatty acid-specific lipid remodeling shapes inflammatory responses and intestinal epithelial barrier functions.

Intestinal epithelial cells are the primary site for the absorption of dietary unsaturated fatty acids, yet the distinct effects of these fatty acids on epithelial barrier integrity and lipid homeostasis remain unclear. Here, we compared the effects of oleic acid (OA), linoleic acid (LA), and α-linolenic acid (ALA), respectively, in differentiated Caco-2 monolayers. LA markedly impaired epithelial barrier integrity, as indicated by disrupted tight junction organization, enhanced oxidative stress, and activation of TLR4/MyD88/NF-κB inflammatory signaling. By contrast, OA induced relatively mild
