The longitudinal and cross-sectional systemic surveillance have been conducted for 9-11 consecutive years in six counties (cities) of Guangxi Zhuang Autonomous Region after basic elimination of bancroftian filariasis. The two different control regimens had been used with DEC selective treatment followed by mass treatment of all persons and selective treatment followed by taking DEC medicated salt. During the former 6 years, residual microfilaremia cases could still be detected; whereas during the latter 5 years, no microfilaremia cases could be detected at all. The natural infection of vector

The decrease of tissue glutathione (GSH) concentrations in different senescent organisms gave rise to our hypothesis that a glutathione deficiency is a biochemical cause of the aging process. A rigorous test of this notion would be the correction of the deficiency and concomitant increase in life span. To this end, adult mosquitoes were fed magnesium thiazolidine-4-carboxylic acid, and their GSH levels and life spans were determined. The GSH levels increased 50-100% (P less than 0.005) regardless of the age when feeding was initiated or whether the feeding period extended over 2 days or the en

BACKGROUND: Long-lasting insecticidal nets are the primary malaria vector control measure in coastal Kenya. In 2018, phenotypic resistance to pyrethroids and low frequency of L1014S kdr mutation were reported in the Anopheles gambiae complex. Since then, additional pyrethroid-treated nets were distributed in 2021. The objectives of this study were to determine the insecticide resistance profiles of An. gambiae and Anopheles funestus from Kwale County and evaluate potential resistance mechanisms.
METHODS: From July 2023 to May 2024, adult and larval collections of An. funestus and An. gambiae

The mouse model of West Nile virus (WNV), which is a leading cause of mosquito-borne encephalitis worldwide, has provided fundamental insights into the host and viral factors that regulate viral pathogenesis and infection outcome. In particular, CD8+ T cells are critical for controlling WNV replication and promoting protection against infection. Here, we present the characterization of a T cell receptor (TCR)-transgenic mouse with specificity for the immunodominant epitope in the WNV NS4B protein (here referred to as transgenic WNV-I mice). Using an adoptive-transfer model, we found that WNV-I

As the world confronts simultaneous climate and health emergencies, the spread of emerging infectious diseases, particularly arboviruses, underscores the intersection of planetary health, global mobility, and disease risk. Viral pathogens like Oropouche, dengue, and chikungunya are extending their reach, with expanding vector habitats (ticks, mosquitoes, and others) driven by global warming and changing ambient humidity. Arboviral risks due to these unfavorable vector dynamics are exacerbated by voluntary and involuntary migration of people, urbanization with attendant crowding, and suboptimal

Anopheles stephensi, an invasive vector is aggressively expanding its geographic range across Africa, posing significant threat to malaria control. Surveillance of its spread is crucial for mitigating its impact on malaria transmission. Here, we report for the first time incursion of An. stephensi into Gayi, a rural area in southern Niger Republic. A combination of morphological identification, end point PCR and sequencing of fragments of COXI and ITS2 genes confirmed An. stephensi. This species was found resting indoor together with other mosquitoes, including An. coluzzii, An. gambiae s.s.,

To persist in the blood circulation and to be available for mosquitoes, Plasmodium falciparum gametocytes modify the deformability and the permeability of their erythrocyte host via cyclic AMP (cAMP) signaling pathway. Cyclic nucleotide levels are tightly controlled by phosphodiesterases (PDE), however in Plasmodium these proteins are poorly characterized. Here, we characterize the P. falciparum phosphodiesterase delta (PfPDEδ) and we investigate its role in the cAMP signaling-mediated regulation of gametocyte-infected erythrocyte mechanical properties. Our results revealed that PfPDEδ is a du

Reproduction of the dominant vector of Zika and dengue diseases, Aedes aegypti mosquito, is controlled by an active heterodimer complex composed of the 20-hydroxyecdysone receptor (EcR) and ultraspiracle protein. Although A. aegypti EcR shares the structural and functional organization with other nuclear receptors, its C-terminus has an additional long F domain (AaFEcR). Recently, we showed that the full length AaFEcR is intrinsically disordered with the ability to specifically bind divalent metal ions. Here, we describe the details of the exhaustive structural and thermodynamic properties of

Dengue virus is a mosquito borne Flavivirus and the most prevalent arbovirus in tropical and subtropical regions around the world. The incidence of dengue has increased drastically over the last few years at an alarming rate. The clinical manifestation of dengue ranges from asymptomatic infection to severe dengue. Even though the viral kinetics of dengue infection is lacking, innate immune response and humoral immune response are thought to play a major role in controlling the virus count. Here, we developed a computer simulation mathematical model including both innate and adaptive immune res

Malaria remains one of the leading causes of death worldwide, despite decades of public health efforts. The recent commitment by many endemic countries to eliminate malaria marks a shift away from programs aimed at controlling disease burden towards one that emphasizes reducing transmission of the most virulent human malaria parasite, Plasmodium falciparum. Gametocytes, the only developmental stage of malaria parasites able to infect mosquitoes, have remained understudied, as they occur in low numbers, do not cause disease, and are difficult to detect in vivo by conventional methods. Here, we

Over 61,000 anophelines collected between January 1988 and October 1989 in three villages in western Venezuela were assayed by ELISA for Plasmodium vivax circumsporozoite (CS) protein. The six specimens confirmed positive belonged to three species: Anopheles (Nyssorhynchus) nuneztovari Gabaldón, 1940, A. albitarsis Arribalzaga, 1878 sensu lato and A. oswaldoi (Peryassu, 1922). The estimated CS protein rate for all species combined was 0.01% (95% confidence limits 0.004-0.02%). This CS protein rate and the mean number of bites received by the collectors indicated a sporozoite inoculation rate o

The transmission role of the residual microfilaremia cases was studied in Pingle Village, Liangqing Township, Yongning County where bancroftian filariasis had been virtually eradicated during 1984-1988. The results indicated that after the microfilarial rate of the human population met the criterion of elimination of filariasis, namely below 1% of the total population in all the administrative villages, and the average density of microfilaria being around 5 per 60 microliters peripheral blood, subsequent control measures might be no longer instituted while the microfilaria rate and its density

This review summarizes the impact of insecticide resistance in urban pests and its implications for 'One Health' systems. Mosquitoes are the major vector of human disease via blood feeding, followed by houseflies and cockroaches via physical contact, and finally bed bugs, ants, and termites, which are not typically disease vectors. Besides overt disease, other impacts in the context of One Health include nuisance biting and resultant infections, as well as allergic reactions. Control of urban pests is still primarily achieved with insecticides, which have continually improved over time. Howeve

BACKGROUND: Anopheles mosquitoes pose notable public health concerns as competent vectors of malaria and other diseases. Although malaria is no longer endemic in the United States, recent locally acquired cases in states including Maryland highlight the need to better understand Anopheles dynamics in the region. This study aimed to identify geographic hotspots of Anopheles presence in Maryland and evaluate how land cover, microclimatic conditions, and socioeconomic vulnerability shape their spatial and temporal distribution.
METHODS: Monthly Anopheles occurrence data (1999-2024) from Global B

BACKGROUND: Malaria transmission hinges on infected Anopheles mosquitoes biting humans, with carbon dioxide (CO2), host odor, and body heat acting as key attractants. Along the Thai-Myanmar border, Anopheles minimus (the Funestus Group), a primary malaria vector, exhibits a stronger preference for human hosts than species of the Maculatus Group. Elucidating the genetic basis of this feeding behavior is essential for improving malaria control strategies.
METHODS: Wild Anopheles mosquitoes were collected in Tha Song Yang district, Tak province, Thailand, from July 2019 to November 2020, using c

As of November 2016, the Florida Department of Health (FDH) and the Centers for Disease Control and Prevention have confirmed more than 4000 travel-related Zika virus (ZIKV) infections in the United States with >700 of those in Florida. There have been 139 cases of locally acquired infection, all occurring in Miami, Florida. Within the US territories (eg, Puerto Rico, US Virgin Islands), >30,000 cases of ZIKV infection have been reported. The projected number of individuals at risk for ZIKV infection in the Caribbean and Latin America approximates 5 million. Similar to Dengue and Chikungunya v

Gametocytogenesis is a crucial process in which malaria parasites transition from their asexual stage to the sexual-stage gametocytes. This transformation enables the parasite to infect and multiply within the Anopheles mosquito, the vector responsible for transmitting the disease between hosts. Understanding how gametocytogenesis works and how it can be controlled offer insights into the malaria life cycle and potential strategies for controlling the transmission. Significant efforts have been dedicated to gametocytogenesis induction in the laboratory using in vitro parasite cultures over the

Human and animal pathogens that are transmitted by arthropods are a global concern, particularly those vectored by ticks (e.g. Borrelia burgdorferi and tick-borne encephalitis virus) and mosquitoes (e.g. malaria and dengue virus). Breaking the circulation of pathogens in permanent foci by controlling vectors using acaricide-based approaches is threatened by the selection of acaricide resistance in vector populations, poor management practices and relaxing of control measures. Alternative strategies that can reduce vector populations and/or vector-mediated transmission are encouraged worldwide.

Temephos is considered the gold standard by the Ministry of Health for controlling the larvae of the mosquito Aedes aegypti. The present study evaluated the effects of Temephos larvicide on the reproductive performance, embryo-fetal development and DNA integrity of Swiss mice. This study used 30 pregnant female mice: 10 were controls treated with drinking water at a dosage of 0.1 mL/10 g (body weight - b.w., administered orally - a.o.), and 20 were treated with Temephos at doses of 0.0043 mg/kg and 0.043 mg/kg (b.w., a.o.) during the gestational period. Statistical analysis showed that Temepho

INTRODUCTION: Neglected tropical diseases (NTDs) in developing countries like the Caribbean, negatively affect multiple income-generating sectors, including the tourism industry upon which island states are highly dependent. Insect-transmitted NTDs include, but are not limited to, malaria, dengue and lymphatic filariasis. Control measures for these disease, are often ignored because of the associated cost. Many of the developing country members are thus retained in a financially crippling cycle, balancing the cost of prophylactic measures with that of controlling an outbreak. The purpose of the
