MsM-DPM: Multiscale Mamba Diffusion Probabilistic Model for Medical Image Segmentation.

Diffusion probabilistic models (DPMs) have recently demonstrated promising performance in medical image segmentation. However, traditional DPM has difficulty handling the irregular structure of images and the inherent similarity between lesions and surrounding tissues. To overcome these challenges, we propose an innovative architecture, the multiscale Mamba DPM (MsM-DPM), designed to enhance medical image segmentation. Specifically, MsM-DPM introduces a multiscale attention fusion module (MSAFM) in a multiscale denoising UNet (Ms-DU) to capture lesion deformations from multilevel features, the

Incorporating Auxiliary Information into Assessment of Accuracy and Discrimination of Risk Models When Some Predictors are Missing.

Validating a risk model in independent data is an important step before recommending it for broad use. However, some model predictors may be missing for some subjects in the validation data, either randomly or by design. Missingness by design occurs for case-cohort or nested case-control studies, in which some predictors are measured only for sub-sampled subjects. Standard approaches to handling missing data are weighting and imputation. We propose adjusting known sampling weights by incorporating auxiliary information available for all cohort members to improve the efficiency of weighted esti

Calibration of Priors for Bayesian Model-Based Dose-Finding Trial Designs With Joint Outcomes.

The goal of dose-finding oncology trials is to assess the safety of anti-cancer treatments across multiple doses and to recommend dose(s) for subsequent trials. As patients' outcomes accrue, trialists dynamically recommend new doses for further investigation during the trial. This adaptive decision-making lends itself to Bayesian learning, with Bayesian frameworks increasingly guiding dose recommendations in model-based dose-finding designs, including the Continual Reassessment Method (CRM). However, such approaches introduce increased complexity, not least when additional outcomes are incorpo

Cure rate joint model for time-to-event data and longitudinal tumor burden with potential change points.

In non-small cell lung cancer (NSCLC) clinical trials, tumor burden (TB) is a key longitudinal biomarker for assessing treatment effects. Typically, standard-of-care (SOC) therapies and some novel interventions initially decrease TB; however, many patients subsequently experience an increase-indicating disease progression-while others show a continuous decline. In patients with an eventual TB increase, the change point marks the onset of progression and must occur before the time of the event. To capture these dynamics, we propose a cure-rate joint model that integrates time-to-event and longi

A comparative study of statistical methods for identifying differentially expressed genes in spatial transcriptomics.

Spatial transcriptomics (ST) provides unprecedented insights into gene expression patterns while retaining spatial context, making it a valuable tool for understanding complex tissue architectures, such as those found in cancers. Seurat, by far the most popular tool for analyzing ST data, uses the Wilcoxon rank-sum test by default for differential expression analysis. However, as a nonparametric method that disregards spatial correlations, the Wilcoxon test can lead to inflated false positive rates and misleading findings. This limitation highlights the need for a more robust statistical appro

A tree-based kernel for densities and its applications in clustering DNase-seq profiles.

Modeling multiple sampling densities within a hierarchical framework enables borrowing of information across samples. These "density random effects" can act as kernels in latent variable models to represent exchangeable subgroups or clusters. A key feature of these kernels is the (functional) covariance they induce, which determines how densities are grouped in mixture models. Our motivating problem is clustering chromatin accessibility profiles from high-throughput DNase-seq experiments to detect transcription factor (TF) binding. TF binding typically produces footprint profiles with spatial

Confidence Set for the Cluster of a Spatial Scan Statistic.

Spatial cluster detection is widely used in fields such as epidemiology and environmental science to identify spatial units that exhibit patterns differing from background variation. While traditional methods like spatial scan statistics are effective in detecting clusters, they often lack tools to quantify and visualize the uncertainty surrounding detected clusters. In this work, we propose a novel framework for constructing confidence sets for spatial clusters, applicable to both Poisson and Normal models. Building on likelihood-based methods, we leverage the p value of the likelihood ratio

Onset and Risk Period of Functional Sound Tooth Loss: A Hyperbolic Tangent Model.

OBJECTIVES: Existing percentile analyses describe age-related tooth loss trajectories but do not quantify when accelerated loss begins or how long the high-risk period lasts. This study aimed to apply a hyperbolic tangent model to functional sound tooth loss data to derive clinically interpretable transition parameters. METHODS: A hyperbolic tangent regression was fitted to normalised functional sound tooth loss data from 27,215 adults across 5 cycles of the Korean National Health and Nutrition Examination Survey (2010-2015), covering the 10th, 25th, 50th, and 75th percentile groups. Spatio-temporal analysis and geostatistical modelling of onchocerciasis prevalence in Nigeria to support elimination efforts.

Nigeria has made significant progress toward the elimination of onchocerciasis through mass drug administration (MDA) of ivermectin, with ten states recently declared eligible to stop treatment following WHO-recommended epidemiological and entomological assessments. However, reliable spatial prevalence estimates remain necessary to guide elimination strategies, particularly in areas with limited surveillance. We applied model-based geostatistical analysis using Monte Carlo Maximum Likelihood Estimation to assess the spatio-temporal distribution of onchocerciasis prevalence across Nigeria from

Deep tobit model: an integrated framework for high-dimensional censored regression with variable selection.

High-dimensional data with left-censored responses are increasingly common in modern applications, yet existing methods for analyzing them are limited. Classical Tobit models fail to handle nonlinear relationships or perform high-dimensional variable selection, whereas deep learning approaches often prioritize prediction performance but lack selection and interpretation capabilities. To address this gap, we propose an integrated deep learning framework, the Deep Tobit model, which employs the negative Tobit log-likelihood as its loss function to properly account for data censoring. A two-stage

A Bayesian Log-Cauchy Mixture Cure Fraction Model for Heavy-Tailed Survival Data: Implementation via OpenBUGS.

Bayesian estimation implemented via OpenBUGS is used to establish the novelty and practical relevance of the log-Cauchy distribution within a mixture cure fraction modeling framework. Mixture cure fraction models are widely used in survival analysis to accommodate the presence of long-term survivors; however, standard parametric assumptions often fail when survival data exhibit heavy-tailed behavior and substantial right censoring. In such settings, conventional distributions such as the exponential, Weibull, gamma, and lognormal may lead to biased estimation and inadequate uncertainty quantif

Estimating Prevalence of Opioid Misuse in North Carolina Counties From 2016 to 2021: An Integrated Abundance Model Approach.

BACKGROUND: The overdose epidemic remains largely driven by opioids, but the county-level prevalence of opioid misuse is unknown. Without this information, public health and policy responses are limited by a lack of knowledge on the scope of the problem. METHODS: Using an integrated abundance model, we estimate the annual county-level prevalence of opioid misuse for counties in North Carolina from 2016 to 2021. The model integrates county-level observed counts of illicit opioid overdose deaths, people receiving prescriptions for buprenorphine, and people served by treatment programs. It also

Improving necrosis estimation in MR-guided prostate focused ultrasound ablations with a probabilistic thermal dose model.

False positive necrosis classification close to ablation zones in magnetic resonance (MR)-guided transurethral ultrasound ablation of the prostate poses an issue for real-time assessment of the treatment. Typically, apriori knowledge about transducers is utilized to filter temperature maps, thereby reducing false positives. Our innovation, however, lies in the ability of our probabilistic CEM43 thermal dose model (pCEM43) to decrease false positives without the need for prior knowledge beyond the noise statistics estimated from baseline images. The original pCEM43 was modified improving statis

Bayesian Borrowing With Multiple Heterogeneous Historical Studies Using Order Restricted Normalized Power Prior.

The recent U.S. Food and Drug Administration guidance on complex innovative trial designs acknowledges the use of Bayesian strategies to incorporate historical information based on clinical expertise and data similarity. Also, data from multiple previous studies with similar settings often qualify for historical borrowing. Although several classes of informative priors can semi-automatically leverage historical information based on data compatibility, it is common that some exogenous factors, such as the year of patient enrollment, can also influence the relevance of each historical study to t

Prediction of transition probabilities in multi-state models with nested case-control data.

Multi-state models are widely used to study complex interrelated life events. In resource-limited settings, nested case-control (NCC) sampling may be employed to extract subsamples from a cohort for an event of interest, followed by a conditional likelihood analysis. However, conditioning restricts the reuse of NCC data for studying additional events. An alternative approach constructs pseudolikelihoods using inverse probability weighting (IPW) for inference with NCC data. Existing IPW-based pseudolikelihood methods focus primarily on estimating relative risks for multiple outcomes or secondar

A multilevel probabilistic dose response model for radiation-induced Harderian gland tumor prevalence-single beam and mixed field effects.

Space radiation exposure is one of the main risks faced by crewmembers during exploration missions. Currently, the greatest uncertainty in radiation-induced cancer risk estimation for spaceflight missions is in the calculation of the quality factor (QF) largely due to limited ground-based experimental data for the vast range of radiation types found in space. Such data are needed to guide and validate dose-response models and subsequent calculations of relative biological effectiveness (RBE) used for QF assignment. In this work, a dose-response model is developed on the basis of probabilistic

Causal Inference Over a Subpopulation: The Effect of Malaria Vaccine in Women During Pregnancy.

Preventing malaria during pregnancy is of critical importance, yet there are no approved malaria vaccines for pregnant women due to lack of efficacy results within this population. Conducting a randomized trial in pregnant women throughout the entire duration of pregnancy is impractical. Instead, a randomized trial was conducted among women of childbearing potential (WOCBP), and some participants became pregnant during the 2-year study. We explore a statistical method for estimating vaccine effect within the target subpopulation-women who can naturally become pregnant, namely, women who can be

Compatibility of Linehan's biosocial theory and the DSM-5 Alternative Model of Personality Disorders for borderline personality disorder.

Borderline personality disorder (BPD) is characterized by affective, interpersonal, and identity instability, as well as marked impulsivity. There is evidence that BPD may be best operationalized dimensionally using models such as the Alternative Model for Personality Disorders (AMPD) described in Section III of the Diagnostic and Statistical Manual for Mental Disorders (DSM). Moreover, biosocial theory is a well-known etiological theory of BPD emphasizing emotion dysregulation, inherited impulsivity, and development within invalidating contexts as key etiological mechanisms. Given that curren

Analysing HCV incidence trends in people who inject drugs using serial behavioural and seroprevalence data: A modelling study.

INTRODUCTION: The introduction of new direct-acting antivirals for hepatitis C virus (HCV) infection, has enabled the formulation of a HCV elimination strategy led by the World Health Organisation (WHO). Guidelines for elimination of HCV target a reduction in incidence, but this is difficult to measure and needs estimating. METHODS: Serial cross-sectional bio-behavioural sero-surveys provide information on an individual's infection status and duration of exposure and how these change over time. These data can be used to estimate the rate of first infection through appropriate statistical mode

A Causal Mediation Approach to Account for Interaction of Treatment and Intercurrent Events: Using Hypothetical Strategy.

Hypothetical strategy is a common strategy for handling intercurrent events (IEs). No current guideline or study considers treatment-IE interaction to target the estimand in any one IE-handling strategy. Based on the hypothetical strategy, we aimed to (1) assess the performance of three estimators with different considerations for the treatment-IE interaction in a simulation and (2) compare the estimation of these estimators in a real trial. Simulation data were generalized based on realistic clinical trials of Alzheimer's disease. The estimand of interest was the effect of treatment with no I
