Children exposed to hydroxychloroquine in utero show no signs of retinal toxicity at age 5.

OBJECTIVE: Limited examinations from prior studies support the absence of retinal toxicity in infants exposed to maternal hydroxychloroquine (HCQ). However, the infant retina does not have adult morphology that could show typical signs of HCQ retinopathy. Using the Preventive Approach To Congenital Heart Block with Hydroxychloroquine (PATCH) study, children at age 5 (when adult morphology has been reached) were evaluated using modern imaging techniques. METHODS: Ocular coherence tomography (OCT) was performed (CIRRUS HD-OCT, Carl Zeiss Meditec) at age 5 on 10 PATCH children exposed to materna

Functional analysis of retinal-binding protein in the visual system of the nocturnal marine shellfish Pacific abalone (Haliotis discus hannai).

The Pacific abalone (Haliotis discus hannai) is a nocturnal species that inhabits marine benthic ecosystems, characterized by its aversion to light and preference for darkness during daylight hours, with movement and feeding predominantly occurring at night. However, the morphology and structure of its visual organs and the mechanisms underlying its perception of cyclical light variations are unknown. This study analyzed the Retinal-binding protein (RALBP) gene to elucidate its role in the abalone's visual system. Using the rapid amplification of cDNA ends technique, the RALBP gene was cloned,

Choroid and choriocapillaris changes in early-stage Parkinson's disease: a swept-source optical coherence tomography angiography-based cross-sectional study.

BACKGROUND: Parkinson's disease (PD) is one of the most common neurodegenerative diseases in the aging population. Previous literature has reported thinning of the retinal nerve fiber layer, ganglion cell layer, inner plexiform layer, and photoreceptor layer in PD patients. However, very few studies have used swept-source optical coherence tomography (SS-OCT) to study the choroid and choriocapillaris vascular changes in PD and their correlations with altered contrast sensitivity. METHODS: PD patients and controls were enrolled in the current study. We used a CSV-1000E instrument to assess con

Melanopsin retinal ganglion cell loss in Alzheimer disease.

OBJECTIVE: Melanopsin retinal ganglion cells (mRGCs) are photoreceptors driving circadian photoentrainment, and circadian dysfunction characterizes Alzheimer disease (AD). We investigated mRGCs in AD, hypothesizing that they contribute to circadian dysfunction. METHODS: We assessed retinal nerve fiber layer (RNFL) thickness by optical coherence tomography (OCT) in 21 mild-moderate AD patients, and in a subgroup of 16 we evaluated rest-activity circadian rhythm by actigraphy. We studied postmortem mRGCs by immunohistochemistry in retinas, and axons in optic nerve cross-sections of 14 neuropath

Assessment of night vision problems in patients with congenital stationary night blindness.

Congenital Stationary Night Blindness (CSNB) is a retinal disorder caused by a signal transmission defect between photoreceptors and bipolar cells. CSNB can be subdivided in CSNB2 (rod signal transmission reduced) and CSNB1 (rod signal transmission absent). The present study is the first in which night vision problems are assessed in CSNB patients in a systematic way, with the purpose of improving rehabilitation for these patients. We assessed the night vision problems of 13 CSNB2 patients and 9 CSNB1 patients by means of a questionnaire on low luminance situations. We furthermore investigated

Tuberculous choroidal granuloma in an HIV-positive patient.

The circadian and neurobehavioral effects of light are primarily mediated by a retinal ganglion cell photoreceptor in the mammalian eye containing the photopigment melanopsin. Nine action spectrum studies using rodents, monkeys, and humans for these responses indicate peak sensitivities in the blue region of the visible spectrum ranging from 459 to 484 nm, with some disagreement in short-wavelength sensitivity of the spectrum. The aim of this work was to quantify the sensitivity of human volunteers to monochromatic 420-nm light for plasma melatonin suppression. Adult female (n=14) and male (n=

Mechanism of protection from light-induced retinal degeneration by the synthetic antioxidant phenyl-N-tert-butylnitrone.

PURPOSE: A prior study demonstrated that pretreatment with phenyl-N-tert-butylnitrone (PBN), a synthetic antioxidant and free radical trapping agent, protects rats from light-induced photoreceptor cell death. The objective of the present study was to elucidate the molecular mechanism of PBN neuroprotection. METHODS: Sprague-Dawley rats (5-6 weeks old) raised in dim (5 lux) cyclic light (12 hours ON/OFF) from birth were injected intraperitoneally with PBN or water 30 minutes before exposure to three columns of fluorescent light (approximately 2700 lux intensity) for 0, 3, 6, 12, or 24 hours. m

Activation of Sigma 1 Receptor Extends Survival of Cones and Improves Visual Acuity in a Murine Model of Retinitis Pigmentosa.

PURPOSE: Retinitis pigmentosa (RP), a retinal photoreceptor degeneration, typically affects rod function and subsequently cones. Activation of sigma 1 receptor (Sig1R) has been shown to preserve cone function through 6 weeks in the rd10 mouse model of RP, when mice were treated systemically with the Sig1R ligand (+)-pentazocine (PTZ). This study determined the extent to which cone function is preserved in rd10 mice when Sig1R is activated. METHODS: Rd10 mice were administered (+)-PTZ (alternate days beginning at postnatal day [P]14) over a period of 180 days. Mouse visual function and structu

Gene Therapy Fully Restores Vision to the All-Cone Nrl(-/-) Gucy2e(-/-) Mouse Model of Leber Congenital Amaurosis-1.

Mutations in GUCY2D are the cause of Leber congenital amaurosis type 1 (LCA1). GUCY2D encodes retinal guanylate cyclase-1 (retGC1), a protein expressed exclusively in outer segments of photoreceptors and essential for timely recovery from photoexcitation. Recent clinical data show that, despite a high degree of visual disturbance stemming from a loss of cone function, LCA1 patients retain normal photoreceptor architecture, except for foveal cone outer segment abnormalities and, in some patients, foveal cone loss. These results point to the cone-rich central retina as a target for GUCY2D replac

Abnormal photoresponses and light-induced apoptosis in rods lacking rhodopsin kinase.

Phosphorylation is thought to be an essential first step in the prompt deactivation of photoexcited rhodopsin. In vitro, the phosphorylation can be catalyzed either by rhodopsin kinase (RK) or by protein kinase C (PKC). To investigate the specific role of RK, we inactivated both alleles of the RK gene in mice. This eliminated the light-dependent phosphorylation of rhodopsin and caused the single-photon response to become larger and longer lasting than normal. These results demonstrate that RK is required for normal rhodopsin deactivation. When the photon responses of RK-/- rods did finally tur

Morphologic effects on the retina of massive exchange transfusion with stroma-free hemoglobin solution.

Retinal morphology serves as an extremely sensitive end point to hypoxic and ischemic conditions manifested by circulatory perturbations. Therefore, the effect of massive transfusion of stroma-free hemoglobin solution (SFHS) on retinal morphology of rats was evaluated. Seventy-three rats were divided into three groups. Two groups were transfused to 75% blood volume replacement--one (n = 30) with 7% SFHS one (n = 26) with 7% albumin solution and the third group (n = 17) was cannulated, not transfused (controls). Animals in each group were killed at 1, 5, 12, and 24 hours and 2 months after the
