Multivariate Genome-Wide Analysis Delineates Shared Genetic Determinants Across Periodontal and Oral Microbial Phenotypes.

BACKGROUND: Periodontitis affects approximately 10% of the global population, posing a serious threat to tooth retention and quality of life. The disease emerges from bidirectional host-microbiome interactions that can shift from homeostasis to dysbiosis and chronic inflammation, yet conventional single-phenotype analyses cannot resolve pleiotropic loci acting across interrelated periodontal and microbial traits. We aimed to jointly model the shared genetic structure of six periodontitis-related phenotypes and to prioritise pleiotropic loci with translational relevance. METHODS AND MATERIALS:

The Genetic Landscape of Plasma P-Selectin Glycoprotein Ligand Levels and Bidirectional Mendelian Randomization to Assess Role in Proinflammatory Cytokine Levels.

BACKGROUND: Polymorphonuclear (PMN) leukocyte recruitment to activated pulmonary endothelium is a central mechanism in acute respiratory distress syndrome (ARDS). This process is mediated by selectins and their counter-ligand, P-selectin glycoprotein ligand-1 (PSGL-1), encoded by SELPLG. Genetic variation in SELPLG has been associated with ARDS susceptibility, while disruption of PSGL-1/P-selectin interactions attenuates lung injury in preclinical models. Because inflammatory stimuli increase both SELPLG expression and circulating PSGL-1 levels, PSGL-1 represents a promising biomarker and ther

X-linked hypophosphatemic rickets: Description of seven new variants in patients followed up in reference hospitals in Rio de Janeiro.

BACKGROUND: X-linked hypophosphatemic rickets (XLHR) is a rare genetic disease, often delayed in diagnosis due to the low degree of suspicion and limited access to sophisticated diagnostic tools that confirm the diagnosis, such as genetic testing. METHODS: Through a cross-sectional and observational study, 26 patients with a previously presumptive diagnosis of X-linked hypophosphatemic rickets (based on clinical history, laboratory findings, and physical examination), were followed for approximately 12 months. During 12 months of follow-up, only 16 patients underwent genetic testing and enrol

SCN1A testing for epilepsy: application in clinical practice.

This report is a practical reference guide for genetic testing of SCN1A, the gene encoding the α1 subunit of neuronal voltage-gated sodium channels (protein name: Nav 1.1). Mutations in this gene are frequently found in Dravet syndrome (DS), and are sometimes found in genetic epilepsy with febrile seizures plus (GEFS+), migrating partial seizures of infancy (MPSI), other infantile epileptic encephalopathies, and rarely in infantile spasms. Recommendations for testing: (1) Testing is particularly useful for people with suspected DS and sometimes in other early onset infantile epileptic encephal

Insights into the genetic epidemiology of Crohn's and rare diseases in the Ashkenazi Jewish population.

As part of a broader collaborative network of exome sequencing studies, we developed a jointly called data set of 5,685 Ashkenazi Jewish exomes. We make publicly available a resource of site and allele frequencies, which should serve as a reference for medical genetics in the Ashkenazim (hosted in part at https://ibd.broadinstitute.org, also available in gnomAD at http://gnomad.broadinstitute.org). We estimate that 34% of protein-coding alleles present in the Ashkenazi Jewish population at frequencies greater than 0.2% are significantly more frequent (mean 15-fold) than their maximum frequency

Single nucleotide polymorphisms associated with sporadic brain arteriovenous malformations: where do we stand?

Brain arteriovenous malformations are characterized by a tangle of abnormal vessels directly shunting blood from the arterial to venous circulation. They are known to occur either sporadically or in the context of well-defined genetic disorders. Haemorrhage represents the most severe clinical manifestation, whereas other common symptoms include headache, seizures and neurological deficits. Although sporadic forms do not recognize a specific genetic cause, in recent years, it has been hypothesized that genes involved in angiogenesis and inflammation or coding for proteins, such as fibronectins,

The retrovirus HTLV-1 inserts an ectopic CTCF-binding site into the human genome.

Human T-lymphotropic virus type 1 (HTLV-1) is a retrovirus that causes malignant and inflammatory diseases in ∼10% of infected people. A typical host has between 10(4) and 10(5) clones of HTLV-1-infected T lymphocytes, each clone distinguished by the genomic integration site of the single-copy HTLV-1 provirus. The HTLV-1 bZIP (HBZ) factor gene is constitutively expressed from the minus strand of the provirus, whereas plus-strand expression, required for viral propagation to uninfected cells, is suppressed or intermittent in vivo, allowing escape from host immune surveillance. It remains unknow

Genetic findings in ten Ecuadorian patients with suspected Wilson's disease.

BACKGROUND: Wilson disease is a rare autosomal recessive disorder caused by variations in ATP7B, leading to copper accumulation and multisystemic damage. Diagnosis is often delayed due to its heterogeneous clinical presentation and limited genetic data in underrepresented populations. METHODS: We characterized ten Ecuadorian patients with clinical suspicion of Wilson disease using whole-exome sequencing (WES), selected to enable simultaneous assessment of ATP7B and other metabolic genes relevant to Wilson-like phenotypes. Variants were interpreted following HGNC and ACMG/AMP guidelines. Ances

Regulatory roles of microRNAs in epigenetic and immune gene networks in psoriasis.

Psoriasis is a chronic, immune-mediated skin disorder characterized by keratinocyte hyperproliferation, impaired differentiation, and sustained inflammation driven by complex interactions between the immune system and the epidermis. Emerging evidence indicates that microRNAs (miRNAs) are central post-transcriptional regulators orchestrating these processes, linking environmental cues, genetic predisposition, and immune signaling. Dysregulated miRNAs in keratinocytes, immune cells, and the circulation contribute to disease onset and progression. Specific miRNAs, such as miR-21, miR-31, miR-155,

A Joint Survival Modeling and Therapy Knowledge Graph Framework to Characterize Opioid Use Disorder Trajectories.

MOTIVATION: Opioid use disorder (OUD) often emerges following prescription opioid exposure and follows a dynamic clinical course characterized by onset, remission, and relapse. Large-scale cohorts linking electronic health records (EHR) with rich longitudinal survey data, such as the All of Us Research Program, enable computational modeling of stage-specific risk and provide an opportunity to connect predictive factors with potential treatment options. RESULTS: We developed a multi-stage computational framework to model transitions across OUD onset, treatment-related remission, and relapse, a

Mechanism of Action of circRNA/miRNA Network in DLBCL.

Circular RNAs (circRNAs) make up approximately 10% of the human transcriptome. CircRNAs belong to the broad group of non-coding RNAs and characteristically are formed by backsplicing into a stable circular loop. Their main role is to regulate transcription through the inhibition of miRNAs' expression, termed miRNA sponging. CircRNAs promote tumorigenesis/lymphomagenesis by competitively binding to miRNAs at miRNA binding sites. In diffuse large B-cell lymphoma (DLBCL), several circRNAs have been identified and their expression is related to both progression and response to therapy. DLBCL is th

Codon-Usage Bias and Genomic Architecture of 79 Psoriasis-Susceptibility Genes: Implications for Expression Efficiency and Therapeutic Targeting.

INTRODUCTION: Psoriasis is a chronic autoimmune skin disorder with a complex genetic basis. However, the codon usage patterns and nucleotide features of psoriasis-related genes remain unexplored, despite their potential to influence gene expression and disease progression. METHODS: We analyzed 79 psoriasis-associated genes to investigate codon usage bias (CUB) and nucleotide composition. Metrics included GC content, effective number of codons (ENC), and relative synonymous codon usage (RSCU). Evolutionary influences were assessed using correspondence analysis, parity rule 2 (PR2) plots, and n

Adaptive neuro-fuzzy inference systems for improved mastitis classification and diagnosis.

For modeling dairy cattle data, fuzzy logic offers the capability to manage uncertainty, enhance accuracy, facilitate informed decision-making, and optimize resource allocation. A critical aspect of dairy cattle production is the modeling of mastitis, an udder infection that affects milk quality and yields significant economic consequences. The aim of this study was to compare the performance of three adaptive neuro-fuzzy inference systems (ANFIS) classification methodologies in classifying mastitis in Holstein dairy cattle: gradient descent (GD)-based ANFIS (GD-ANIFIS), particle swarm optimiz
