Implications of insecticide resistance for malaria vector control with long-lasting insecticidal nets: evidence from health facility data from Benin.

BACKGROUND: Insecticide-based interventions have averted more than 500 million malaria cases since 2000, but insecticide resistance in mosquitoes could bring about a rebound in disease and mortality. This study investigated whether insecticide resistance was associated with increased incidence of clinical malaria. METHODS: In an area of southern Benin with insecticide resistance and high use of insecticide-treated nets (ITNs), malaria morbidity and insecticide resistance were measured simultaneously in 30 clusters (villages or collections of villages) multiple times over the course of 2 years

Zika virus incidence, preventive and reproductive behaviors: Correlates from new survey data.

During the outbreak of the Zika virus, Brazilian health authorities recommended that pregnant women take meticulous precaution to avoid mosquito bites and that women in general use contraceptive methods to postpone/delay pregnancies. In this article, we present new estimates on the Zika virus incidence, its correlates and preventive behaviors in the Northeast of Brazil, where the outbreak initiated, using survey data collected between March 30th and June 3rd of 2016. The target population were women aged 15-49 in the capital cities of the nine states of the Northeast region of Brazil. We find

Enhanced protection against malaria by indoor residual spraying in addition to insecticide treated nets: is it dependent on transmission intensity or net usage?

BACKGROUND: Insecticide treated nets (ITNs) and indoor residual spraying (IRS) are effective vector control tools that protect against malaria. There is conflicting evidence regarding whether using ITNs and IRS in combination provides additional benefit over using either of these methods alone. This study investigated factors that may modify the effect of the combined use of IRS and ITNs compared to using ITNs alone on malaria infection prevalence. METHODS: Secondary analysis was carried out on data from a cluster randomised trial in north-west Tanzania. 50 clusters received ITNs from a unive

Incubation periods of Yellow fever virus.

Yellow fever virus is a global health threat due to its endemicity in parts of Africa and South America where human infections occur in residents and travelers. To understand yellow fever dynamics, it is critical to characterize the incubation periods of the virus in vector mosquitoes and humans. Here, we compare four statistical models of the yellow fever incubation periods fitted with historical data. The extrinsic incubation period in the urban vector Aedes aegypti was best characterized with a temperature-dependent Weibull model with a median of 10 days at 25 degrees C (middle 95% = 2.0-37

Protective behavior survey, West Nile virus, British Columbia.

We investigated personal protective behaviors against West Nile virus infection. Barriers to adopting these behaviors were identified, including the perception that DEET (N,N-diethyl-m-toluamide and related compounds) is a health and environmental hazard. Televised public health messages and knowing that family or friends practiced protective behaviors were important cues to action.

West Nile Virus.

West Nile Virus (WNV), a mosquito transmitted disease, made its first appearance in North America in 1999 and quickly spread westward. It arrived in Kansas in 2002, re-emerged in 2003, and is expected again in 2004. The natural cycle is from infected bird to mosquito, with humans and horses as accidental (dead-end) hosts. WNV is usually asymptomatic in humans, but can cause a wide spectrum of disease. There is currently no human vaccine and treatment is symptomatic only. A best preventive measure combines personal protection to prevent mosquito bites with reducing the number of mosquitoes.

Epizootic activity of Murray Valley encephalitis and Kunjin viruses in an aboriginal community in the southeast Kimberley region of Western Australia: results of mosquito fauna and virus isolation studies.

We undertook annual surveys of flavivirus activity in the community of Billiluna in the southeast Kimberley region of Western Australia between 1989 and 2001 [corrected]. Culex annulirostris was the dominant mosquito species, particularly in years of above average rains and flooding. Murray Valley encephalitis (MVE) virus was isolated in 8 of the 13 years of the study from seven mosquito species, but more than 90% of the isolates were from Cx. annulirostris. The results suggest that MVE virus is epizootic in the region, w ith activity only apparent in years with average or above average rainfa

Queenslanders' use of personal strategies to minimise risk of mosquito-borne disease.

OBJECTIVE: To describe Queenslanders' awareness, knowledge and attitudes towards mosquito-borne diseases and their transmission, and to determine which factors influence the adoption of effective individual prevention strategies. METHODS: In 1995-6, cross-sectional surveys of adult residents in the western suburbs of Brisbane and registered voters in Cairns were conducted. Forced entry logistic regression was used to predict use of personal protection and elimination of domestic breeding sites in the two cities. RESULTS: Final sample sizes were 347 in Cairns and 165 in Brisbane with response

Introduction of an integrated community-based bancroftian filariasis control program into the Mt Bosavi region of the Southern Highlands of Papua New Guinea.

In mid-1987 a baseline microfilarial prevalence survey was conducted among five villages in the Mt Bosavi region of the Southern Highlands Province of Papua New Guinea. Through use of the Nucleopore filtration technique, it was determined that 48% of villagers had detectable microfilaraemia. The highest prevalence was documented in Fogomaiyu, where the microfilaraemia rate was 92%. On the basis of this initial survey and the expressed interest of the community, the Division of Health in the Southern Highlands Province undertook an integrated community-based pilot control program. The project u

A progress report in advancements of heterocyclic compounds as novel antimalarial agents over the last 5 years.

Malaria, caused by Plasmodium parasites and transmitted by Anopheles mosquitoes, remains a significant global health challenge, especially in tropical and subtropical regions where the disease is endemic. The complex Plasmodium lifecycle, involving stages in both the liver and bloodstream, leads to symptoms such as high fever, anemia, and, in severe cases, life-threatening complications, particularly P. falciparum infections. While historical treatments such as quinine and modern therapies such as artemisinin-based combination therapies (ACTs) have been effective, the growing issue of drug and

Aedes aegypti Hemocytes Mediate Antiviral Immunity.

Hemocytes play several key roles in the mosquito's immune response. Despite most of our understanding regarding their immunological role concerns their responses against bacteria, fungi, and Plasmodium, our knowledge of hemocyte's role in antiviral defense is poorly understood. We performed a comprehensive comparative transcriptomic analysis between the dengue vector Aedes aegypti's two major immune cell types, hemocytes and fat body, revealing a plethora of differentially expressed immune genes that indicates a high level of functional specialization as well as complementation between the two

microRNAs: An opportunity to overcome significant challenges in malaria detection and control.

Organ damage and pathological disease states lead to the rapid release of microRNAs (miRNAs), a class of endogenous small non-coding RNAs, into the blood circulation. Because secreted miRNAs can be detected in biologic fluids such as plasma, they are currently being explored as promising non-invasive biomarkers of infectious and non-infectious diseases. Malaria remains a major global health challenge but still the potential of miRNAs has not been explored extensively in the context of malaria compared to other diseases. Here, we highlight important miRNAs found during different phases of the m

Recent Advances in Understanding the Inflammatory Response in Malaria: A Review of the Dual Role of Cytokines.

Malaria is a serious and, in some unfortunate cases, fatal disease caused by a parasite of the Plasmodium genus. It predominantly occurs in tropical areas where it is transmitted through the bite of an infected Anopheles mosquito. The pathogenesis of malaria is complex and incompletely elucidated. During blood-stage infection, in response to the presence of the parasite, the host's immune system produces proinflammatory cytokines including IL-6, IL-8, IFN-γ, and TNF, cytokines which play a pivotal role in controlling the growth of the parasite and its elimination. Regulatory cytokines such as

Optimal Control of Mitigation Strategies for Dengue Virus Transmission.

Dengue virus is transmitted by Aedes mosquitoes, posing threat to people's health and leading to great economic cost in many tropical and subtropical regions. We develop an ordinary differential equation model taking into account multiple strains of dengue virus. Using the model, we assess the effectiveness of human vaccination considering its waning and failure. We derive the lower bound and upper bound for the final size of the epidemic. Sensitivity analysis quantifies the impact of parameters on the basic reproduction number. Different scenarios of vaccinating humans show that it is better

Confirmation of Zika virus infection through hospital-based sentinel surveillance of acute febrile illness in Uganda,  2014-2017.

Zika virus (ZIKV), transmitted by Aedes species mosquitoes, was first isolated in Uganda in 1947. From February 2014 to October 2017, the Uganda Virus Research Institute, in collaboration with the US Centers for Diseases Control and Prevention, conducted arbovirus surveillance in acute febrile illness (AFI) patients at St Francis hospital in Nkonkonjeru. Three hundred and eighty-four serum samples were collected and tested for IgM antibodies to yellow fever virus (YFV), West Nile virus (WNV), dengue virus (DENV), chikungunya virus (CHIKV) and ZIKV. Of the 384 samples, 5 were positive for ZIKV

Detection of the Wolbachia protein WPIP0282 in mosquito spermathecae: implications for cytoplasmic incompatibility.

Cytoplasmic incompatibility (CI) is a conditional sterility induced by the bacterium Wolbachia pipientis that infects reproductive tissues in many arthropods. Although CI provides a potential tool to control insect vectors of arthropod-borne diseases, the molecular basis for CI induction is unknown. We hypothesized that a Wolbachia-encoded, CI-inducing factor would be enriched in sperm recovered from spermathecae of female mosquitoes. Using SDS-PAGE and mass spectrometry, we detected peptides from the 56 kDa hypothetical protein, encoded by wPip_0282, associated with sperm transferred to femal

Human genetic determinants of dengue virus susceptibility.

Dengue virus (DENV) is an emerging mosquito-borne pathogen that produces significant morbidity worldwide resulting in an estimated 50-100 million infections annually. DENV causes a spectrum of illness ranging from inapparent infection to life-threatening hemorrhagic fever and shock. The varied DENV disease outcome is determined by complex interactions between immunopathologic, viral, and human genetic factors. This review summarizes these interactions with a focus on human genetic determinants of DENV susceptibility, including human leukocyte antigens, blood type, and single nucleotide polymor

Understanding malaria vector dynamics and transmission in the Pouth-Kelle community, Nyong et Kelle division, Centre Region, Cameroon.

INTRODUCTION: Malaria remains a significant public health challenge in tropical regions. The disease is driven by multiple Anopheles vector species with varied ecological and behavioral traits. In transitional zones like the forested Pouth-Kelle community, detailed knowledge of vector dynamics and disease transmission remains limited. This study aimed to characterize local Anopheles species composition, biting behavior, and malaria transmission indicators to guide control strategies. MATERIALS AND METHODS: From February 2021 to January 2022, adult mosquitoes were monthly collected indoors and

Investigating genetic differentiation between brackish and fresh water collections of the arboviral vector Aedes aegypti.

BACKGROUND: Aedes aegypti is typically regarded as a freshwater mosquito; however, recent studies have documented its development in brackish water habitats in coastal regions, including Sri Lanka's Jaffna Peninsula. Compared with freshwater populations, brackish water samples in Jaffna display enhanced salt tolerance throughout larval-to-adult development, along with differences in gene expression, cuticle morphology and composition, and insecticide susceptibility. METHODS: To explore the genetic basis of these differences, we performed a comparative genomic analysis using 5135 genome-wide s
