Antinociceptive and hypnotic activities of pregabalin in a neuropathic pain-like model in mice.

To evaluate the antinociceptive and hypnotic effects of pregabalin, we established a neuropathic pain-like model in mice using partial sciatic nerve ligation (PSNL), and examined thermal hyperalgesia, mechanical allodynia, electroencephalogram, rota-rod testing, and c-Fos expression in the anterior cingulate cortex. Gabapentin was used as a reference drug in the study. Pregabalin administered i.g. at 12.5 and 25mg/kg prolonged the duration of thermal latencies by 1.4- and 1.6-fold and increased the mechanical threshold by 2.2- and 3.1-fold 3h after administration, respectively, but did not aff

Progressive sleep and electroencephalogram changes in mice carrying the Huntington's disease mutation.

Sleep disturbances in Huntington's disease may be deleterious to the cognitive performance, affective behaviour, and general well-being of patients, but a comprehensive description of the progression of changes in sleep and electroencephalogram in Huntington's disease has never been conducted. Here we studied sleep and electroencephalogram disturbances in a transgenic mouse model of Huntington's disease (R6/2 mice). We implanted 10 R6/2 mice and five wild-type littermates with electromyography electrodes, frontofrontal and frontoparietal electroencephalogram electrodes and then recorded sleep/

Outcome of restless legs severity after continuous positive air pressure (CPAP) treatment in patients affected by the association of RLS and obstructive sleep apneas.

BACKGROUND AND PURPOSE: The association of obstructive sleep apnea syndrome (OSAS) and restless legs syndrome (RLS) has been reported in the literature for many years. Nevertheless, this pathological association has not yet been extensively studied and many questions remain unanswered. The primary concern of this study is to evaluate the influence of nasal continuous positive airway pressure (n-CPAP) therapy on daytime sleepiness and fatigue as well as the presence of a possible long-term beneficial effect on RLS severity and, secondarily, to ascertain the clinical, neurophysiological and poly

High-frequency oscillations arise through distinct mechanisms in models of α- synucleinopathy and channelopathy-induced epilepsy.

Background Pathological high-frequency oscillations (HFOs), widely researched in epilepsy, are increasingly being investigated as potential biomarkers of network hyperexcitability in neurodegenerative disorders. This interest is driven by growing evidence of comorbidity between epilepsy and neurodegenerative diseases, particularly Alzheimer's disease (AD) and dementia with Lewy bodies (DLB), suggesting that these HFOs could be useful biomarkers in these disorders. Patients with AD and mouse models of AD exhibit elevated HFOs during neurophysiology recordings, but whether HFOs occur in DLB rema

N-terminal proVGF modifications related to Amyotrophic Lateral Sclerosis.

Objective: Amyotrophic Lateral Sclerosis (ALS) is a progressive neurodegenerative disorder characterized by motor dysfunction, for which effective therapies and early diagnostic biomarkers remain limited. This study comprehensively investigated ALS-associated alterations in the N-terminal region of the proVGF precursor protein, evaluating their potential as novel diagnostic and prognostic biomarkers and exploring their precise correlations with clinical severity and plasma neurofilament light chain (NfL), an established indicator of neuroaxonal injury. Methods: N-terminal proVGF levels were qu

α5-containing nicotinic acetylcholine receptors regulate state transitions in cortical networks and modulate susceptibility to epileptiform discharges.

Spike-and-wave discharges (SWDs) of absence seizures and slowly oscillating Up-Down states of non-REM sleep and are both manifestations of large-scale synchronized cortical activity, yet the circuit mechanisms differentiating these phenomena remain poorly understood. Cholinergic modulation via nicotinic acetylcholine receptors (nAChRs) plays a key role in shaping cortical excitability. Here, we investigated the role of α5 subunit-containing nAChRs in regulating spontaneous cortical activity and the transition to paroxysmal states. We used acute brain slices from wild type (wt) and α5 knockout

Salience and frontoparietal network impairments across disease stages in dementia with Lewy bodies: A comparative functional MRI study with Alzheimer's disease.

BackgroundResting-state functional magnetic resonance imaging (fMRI) studies in dementia with Lewy bodies (DLB) and Alzheimer's disease (AD) have described connectivity alterations in large-scale brain networks. However, little is known about functional changes across disease stages, particularly in DLB.ObjectiveTo investigate functional connectivity of key brain networks in DLB patients at different stages, compare them to AD patients and healthy controls (HC), and examine associations with core clinical symptoms.MethodsNinety DLB patients (63 with mild cognitive impairment [MCI-DLB] and 27 w

A case of Homer-3 IgG cerebellar ataxia & literature review of 15 reported cases.

Homer-3-IgG autoimmunity is a rare diagnosis typically presenting with subacute cerebellar ataxia. Here, we report a patient presenting insidiously with cerebellar ataxia and mild cognitive impairment, referred to the Mayo Clinic for a second opinion of her initial diagnosis of parkinsonism. Considering autoimmune cerebellar ataxia as the correct diagnosis, the patient's cerebrospinal fluid (CSF) and serum samples were sent for Movement Disorders Autoimmune/Paraneoplastic Evaluation neural antibody testing. Murine tissue-based indirect immunofluorescence assay of serum and CSF revealed a uniqu

No anti-IgLON5 antibody in carefully diagnosed PSP patients with "atypical" or variant clinical features.

BACKGROUND: "Atypical" features including REM sleep behaviour disorder (RBD) symptoms and visual hallucinations, and non-Richardson's syndrome (non-RS) variants, are increasingly recognized in progressive supranuclear palsy (PSP). A concern in clinical practice is missing PSP mimics, like anti-IgLON5 disease, an autoimmune encephalitis which can reportedly present with "PSP-like" features and is amenable to immunotherapy. We aimed to determine if anti-IgLON5 antibody is detectable in Movement Disorder Society criteria-diagnosed PSP patients exhibiting "atypical" features and variant phenotypes

Novel mouse model of alternating hemiplegia of childhood exhibits prominent motor and seizure phenotypes.

Pathogenic variants in ATP1A3 encoding the neuronal Na/K-ATPase cause a spectrum of neurodevelopmental disorders including alternating hemiplegia of childhood (AHC). Three recurrent ATP1A3 variants are associated with approximately half of known AHC cases and mouse models of two of these variants (p.D801N, p.E815K) replicated key features of the human disorder, which include paroxysmal hemiplegia, dystonia and seizures. Epilepsy occurs in 40-50 % of individuals affected with AHC, but detailed investigations of seizure phenotypes were limited in the previously reported mouse models. Using gene

Optimising classification of Parkinson's disease based on motor, olfactory, neuropsychiatric and sleep features.

Olfactory loss, motor impairment, anxiety/depression, and REM-sleep behaviour disorder (RBD) are prodromal Parkinson's disease (PD) features. PD risk prediction models typically dichotomize test results and apply likelihood ratios (LRs) to scores above and below cut-offs. We investigate whether LRs for specific test values could enhance classification between PD and controls. PD patient data on smell (UPSIT), possible RBD (RBD Screening Questionnaire), and anxiety/depression (LADS) were taken from the Tracking Parkinson's study (n = 1046). For motor impairment (BRAIN test) in PD cases, publish

Clinical and Dopamine Depletion Patterns in Hyposmia- and Dysautonomia-Dominant Parkinson's Disease.

BACKGROUND: Olfactory or autonomic dysfunction is one of the earliest prodromal symptoms of Parkinson's disease (PD). It has not been investigated whether PD patients have different phenotypes depending on the presence of these prodromal symptoms. OBJECTIVE: To investigate whether hyposmia-dominant and dysautonomia-dominant patients with early PD have different clinical manifestations and nigrostriatal degeneration. METHODS: This cross-sectional study recruited 168 drug-naive PD patients and 34 control subjects. PD patients were classified as patients without hyposmia and dysautonomia (PD-H-

Association of specific biotypes in patients with Parkinson disease and disease progression.

OBJECTIVE: To identify biotypes in patients with newly diagnosed Parkinson disease (PD) and to test whether these biotypes could explain interindividual differences in longitudinal progression. METHODS: In this longitudinal analysis, we use a data-driven approach clustering PD patients from the Parkinson's Progression Markers Initiative (n = 314, age 61.0 ± 9.5, years 34.1% female, 5 years of follow-up). Voxel-level neuroanatomic features were estimated with deformation-based morphometry (DBM) of T1-weighted MRI. Voxels with deformation values that were significantly correlated (p < 0.01) wit

Inflammation and asthma control in children with comorbid obstructive sleep apnea.

OBJECTIVES: A bi-directional relationship exists between asthma and obstructive sleep apnea (OSA) in which presence of one is associated with increased prevalence and severity of the other. Our objective was to determine whether OSA accounted for differences in airway and systemic inflammation in asthmatic children and whether inflammation was associated with asthma control. We hypothesized that greater severity of SDB would correlate with increased upper airway and systemic inflammation and result in reduced asthma control. METHODS: Non-obese children aged 4-12 years with persistent asthma,

Latent Cognitive Phenotypes in De Novo Parkinson's Disease: A Person-Centered Approach.

OBJECTIVES: Cognitive impairment is an important aspect of Parkinson's disease (PD), but there is considerable heterogeneity in its presentation. This investigation aims to identify and characterize latent cognitive phenotypes in early PD. METHODS: Latent class analysis, a data-driven, person-centered, cluster analysis was performed on cognitive data from the Parkinson's Progressive Markers Initiative baseline visit. This analytic method facilitates identification of naturally occurring endophenotypes. Resulting classes were compared across biomarker, symptom, and demographic data. RESULTS:

Characteristics, correlates, and assessment of psychosis in Parkinson disease without dementia.

INTRODUCTION: Considering that psychosis in Parkinson disease (PD) is associated with worse outcomes, including dementia, we aimed to study the characteristics, correlates, and assessment of PD psychosis in those without dementia. METHODS: 101 PD subjects without dementia (Montreal Cognitive Assessment ≥21/30) were recruited to participate in a study of neuropsychiatric symptoms in PD. This study included a baseline standard neurological exam and common PD symptom assessments. Using the Scale for the Assessment of Positive Symptoms (SAPS) and separate assessment of visual illusions and sense

Gamma band activity in the developing parafascicular nucleus.

The parafascicular nucleus (Pf) receives cholinergic input from the pedunculopontine nucleus, part of the reticular activating system involved in waking and rapid eye movement (REM) sleep, and sends projections to the cortex. We tested the hypothesis that Pf neurons fire maximally at gamma band frequency (30-90 Hz), that this mechanism involves high-threshold voltage-dependent P/Q- and N-type calcium channels, and that this activity is enhanced by the cholinergic agonist carbachol (CAR). Patch-clamped 9- to 25-day-old rat Pf neurons (n = 299) manifested a firing frequency plateau at gamma band
