Emergence of PER and VEB extended-spectrum beta-lactamases in Acinetobacter baumannii in Belgium.

OBJECTIVES: Strains of Acinetobacter baumannii producing the extended-spectrum beta-lactamase (ESBL) PER-1 are widespread in Turkey and have also been reported from Korea and France. In contrast, A. baumannii producing the ESBL VEB-1 have only been reported from France, where one strain was responsible for a nationwide outbreak in 2003-2004. Here we describe the emergence of strains of A. baumannii producing VEB-1 and PER- 1 in Belgium. METHODS: Belgian hospitals were alerted in December 2003 to the emergence in France of VEB-1-producing A. baumannii susceptible only to meropenem and colistin…

Partial characterization of a transposon containing the tet(A) determinant in a clinical isolate of Acinetobacter baumannii.

A genomic library from one clinical isolate of Acinetobacter baumannii was obtained to find genes responsible for tetracycline resistance. Escherichia coli DH5alpha-MDR transformants, selected on Mueller-Hinton agar supplemented with tetracycline 18 mg/L, were thoroughly characterized. In one clone, with an insert of 7,070 bp, it was found that the resistance to tetracycline was mediated by the tet(A) gene (1,200 bp) which encodes a tetracycline efflux pump. This gene was recovered together with tetR(A) (651 bp), the tet(A) repressor. Moreover, the partial sequence (2,008 bp) of a transposase …

Verification of a PCR-based typing method for Acinetobacter baumannii in a pseudo-outbreak investigation.

Acinetobacter spp. isolates were increasingly obtained from clinical specimens and sterility samples, and a subsequent epidemiological investigation implicated an intermittently contaminated supply of commercially acquired enrichment broths. Typing was performed with DNA amplification by the polymerase chain reaction (PCR) using enterobacterial repetitive intergenic consensus sequence primers, ERIC2 and reverse ERIC1R. The reliability of this PCR-based typing method was verified by the ability of the technique to demonstrate homology and differences among isolates from an epidemiologically wel

Thermostability Engineering of a Chitin-Utilizing GlcNAc Deacetylase from Acinetobacter baumannii via Distal Mutations.

The deacetylation of N-acetylglucosamine (GlcNAc) to glucosamine (GlcN) is a key step in enzymatic chitin valorization. We identified a novel GlcNAc deacetylase (AbNGD) from Acinetobacter baumannii with high substrate specificity and catalytic activity but poor thermal stability (half-life of 5.5 h at 37 °C). Using the FireProt platform, we generated a combinatorial mutant, AbNGD-M3 (T93P-C153T-A244M). Its optimal temperature increased to 37 °C, and its half-life reached 28.1 h (5.1-fold improvement), while retaining 97% of wild-type catalytic efficiency (kcat/Km = 3.4 mM-1·s-1). Site-directed…

KEGG orthology-based machine learning reveals functional determinants of antimicrobial resistance in Acinetobacter baumannii.

UNLABELLED: Antimicrobial resistance represents a critical global health threat, with Acinetobacter baumannii classified as a priority pathogen due to its extensive multidrug resistance. Currently, antimicrobial susceptibility testing relies on phenotypic characterization, which provides limited mechanistic insights into resistance mechanisms and significantly delays appropriate therapy. While machine learning approaches have shown promise for rapid resistance prediction, existing computational methods rely predominantly on gene presence-absence matrices that inadequately capture functional ca…

Disulfiram Attenuates Acinetobacter baumannii-Induced Hyperinflammation by Regulating Inflammatory Pathways: Insights from RNA Sequencing.

The emergence of carbapenem-resistant Acinetobacter baumannii (CRAB) poses a major obstacle in treating sepsis, highlighting the urgent need for new therapeutic approaches. Recent studies have reported that disulfiram (DSF) exhibits antibacterial activity and potentiates meropenem activity against resistant A. baumannii (A. baumannii) isolates. The mechanisms by which DSF modulates inflammation during A. baumannii infection remain unclear, although DSF possesses anti-inflammatory properties by regulating the pyroptosis pathway. The present study aimed to investigate the potential effect of DSF…

Nutrient-driven small-colony variants as an adaptive outcome in Carbapenem-resistant Acinetobacter baumannii.

Carbapenem-resistant Acinetobacter baumannii (CRAB) is a serious threat, and resistance is emerging to cefiderocol, one of the few remaining treatment options. However, how host micronutrients drive bacterial adaptation under antibiotic pressure is poorly defined. Using the AB5075 and CDC AR Bank #0033 (M27835) CRAB isolates, we combined time-kill assays, whole-genome sequencing, gene expression analysis, and phenotyping to test how vitamin B12 promotes small-colony variants (SCVs) -like adaptive derivatives and alters cefiderocol response. Vitamin B12 reproducibly selected slow-growing SCV-li…

Antibacterial efficacy and mechanism of the novel antimicrobial peptide lachnospirin-1 against Acinetobacter baumannii.

The treatment of carbapenem-resistant A. baumannii (CRAB) infections has become a major global medical challenge; therefore, there is an urgent need for new antimicrobial development. Antimicrobial peptides hold great promising potential due to their rapid bactericidal activity and low induction of drug resistance. In this study, we screened and synthesized the antimicrobial peptide lachnospirin-1, which exhibited significant bactericidal activity against CRAB and can also effectively eliminate its biofilms and persister cells. Mechanism studies, including fluorescent probe detection, transmis
