In vivo albendazole treatment of Taenia crassiceps cysticerci strain WFU: proliferation, damage, and recovery.

Taenia crassiceps has been widely experimented as a model for in vitro and in vivo studies on drug responses. The purpose of this study was to treat BALB/c mice infected with T. crassiceps strain WFU with commercially available albendazole and to analyze the reduction in parasite infrapopulations. Here, we describe the reduction and apparent damage of T. crassicceps WFU cysticerci in infected mice after antihelminthic drug treatment and subsequent inoculation of those treated parasites into a naïve host. We were able to reduce significantly the parasite counts to 33 and 48% after albendazole t…

Novel albendazole-chitosan nanoparticles for intestinal absorption enhancement and hepatic targeting improvement in rats.

To improve the treatment of helminthiasis, filariasis, and colorectal cancer, albendazole-associated chitosan nanoparticles (ABZ-CS-NPs) were prepared using the emulsion crosslinking volatile technique with contained sodium tripolyphosphate as the crosslinking agent and Poloxamer 188 as the auxiliary solvent. The structural characteristics of the NPs were determined using X-ray diffraction to analyze the interaction between CS and the drug. The NPs were then evaluated in terms of their physicochemical characteristics, drug release behavior, in vivo pharmacokinetic parameters, and biodistributi…

Continuous albendazole therapy in alveolar echinococcosis: long-term follow-up observation of 20 cases.

Twenty patients with primary hepatic alveolar echinococcosis were treated with continuous long-term albendazole at a dose of 20 mg/kg/d for an average of 5.7 years (1.5-13.5 years) and were followed up for an average of 12.7 years (4.1-13.5 years). The therapeutic effects were evaluated by clinical, laboratory (haemogram, liver function tests), ultrasonography and computed tomography (CT) examinations. The CT pattern of hepatic lesions was classified into three types: solid form (5 cases), pseudocystic form (6 cases) and 'geographic map' (mixed) form (9 cases). Short-term results were encourag…

[Excessive subcutaneous gnathostomosis after a six months stay in Peru. Successful treatment with albendazole].

A 27-year-old female primatologist returned from a trip to Peru with painful swellings on the right jaw, the mandible and at the axillary gland. Later on her right upper arm and forearm showed big, painful swellings with reddish stripes, while the former swellings gradually disappeared. Finally the back of the right hand, the right forefinger and the left forearm showed the same swellings. Diagnosis was established by the evidence of special antibodies against Gnathostoma spinigerum. Two courses of a 4-week treatment with albendazole cured the patient.

Randomized trial of albendazole versus tiabendazole plus flubendazole during an outbreak of human trichinellosis.

To determine the therapeutic usefulness of benzimidazoles in trichinellosis, 117 patients from a single outbreak were treated either with albendazole alone (N = 59) or with a regimen including tiabendazole followed by flubendazole (N = 58). The criteria of disease activity were evaluated at days 1, 7, 15, and 45. No difference was found between the two groups with regard to the evolution of myalgia, fever, fatigue, new clinical manifestations, or laboratory and serologic data. Both treatment regimens were well tolerated. In all, 30 patients of the albendazole group and 29 of the tiabendazole-f…

Albendazole Treatment of Alveolar Echinococcosis in a Trial Based on a Cross Sectional Ultrasound Study in Kyrgyzstan.

BACKGROUND: Human alveolar echinococcosis (AE) is a potentially fatal parasitic disease. In the low-resource country of Kyrgyzstan, it is essential to develop protocols to manage the large number of AE cases being diagnosed. METHODS: A randomised trial was undertaken to explore the hypothesis that subjects with small AE lesions, detected by ultrasound, a wait and see approach could be adopted. Alternatively, subjects could be managed by albendazole and avoid invasive surgical treatment. One group of subjects with small P1N0M0 lesions was treated with albendazole. A second group remained untre…

Synergistic Effects of Combined Caryophyllene and Albendazole against Invaginated and Evaginated Protoscoleces of Echinococcus granulosus.

BACKGROUND: Natural compounds have garnered increasing attention as candidate antiparasitic therapeutics. We aimed to evaluate the protoscolicidal efficacy of caryophyllene (CAP), alone and in combination with albendazole (ALZ), against hydatid cyst protoscoleces. METHODS: Suspensions of 10,000 invaginated and evaginated protoscoleces (400 μL normal saline) were exposed to CAP (62.5-500 μg/mL), ALZ (250 μg/mL), or CAP+ALZ combinations in 24-well culture plates. Protoscolex viability was assessed by eosin exclusion test. Reactive oxygen species (ROS) induction was quantified using the DCFH-DA …

Studies on absorption mechanism and pharmacokinetic properties of albendazole-bile acid conjugate: In vivo and in vitro.

PURPOSE: To improve the oral bioavailability of albendazole (ABZ), a series of albendazole-bile acid conjugates (ABCs) were synthesized. ABC's transmembrane transport mechanism and in vivo pharmacokinetic properties were preliminarily studied. METHODS: The transmembrane transport mechanism of ABCs was studied using the Caco-2 monolayer cell model and intestinal perfusion model. The concentration of ABCs and ABZ were evaluated using High-Performance Liquid Chromatography (HPLC) and HPLC-Mass Spectrometry (HPLC-MS/MS). RESULTS: Compared to ABZ, better permeability was observed for different ty…

Evaluation of the In-Vitro Effects of Albendazole, Mebendazole, and Praziquantel Nanocapsules against Protoscolices of Hydatid Cyst.

Cystic echinococcosis still remains a serious health and economic problem worldwide. The etiologic agent is Echinococcus granulosus sensu lato, giving origin to a fluid-filled cystic lesion. Therapy faces several challenges. Nanodrugs have shown promise as chemotherapeutics against hydatid cysts. The present study evaluated a highly safe lipid nano-polymeric capsule for its superior efficacy and ability to overcome drug resistance. Nanocapsule drugs were formulated into six groups: Albendazole, mebendazole, praziquantel, albendazole + mebendazole, albendazole + praziquantel, and praziquantel +…

Factors associated with variation in single-dose albendazole pharmacokinetics: A systematic review and modelling analysis.

BACKGROUND: Albendazole is an orally administered anti-parasitic medication with widespread usage in a variety of both programmatic and clinical contexts. Previous work has shown that the drug's pharmacologically active metabolite, albendazole sulfoxide, is characterised by substantial inter-individual pharmacokinetic variation. This variation might have implications for the efficacy of albendazole treatment, but current understanding of the factors associated with this variation remains incomplete. METHODOLOGY/PRINCIPAL FINDINGS: We carried out a systematic review to identify references cont…

In vivo efficacy of combination therapy with albendazole and atovaquone against primary hydatid cysts in mice.

Alveolar echinococcosis (AE) is caused by the larval stage of Echinococcus multilocularis. Chemotherapy for AE involves albendazole (ABZ), which has shown insufficient efficacy. More effective chemotherapy for AE is needed. Previously, we have demonstrated that atovaquone (ATV), an antimalarial, inhibits mitochondrial complex III of E. multilocularis and restricts the development of larval cysts in in vivo experiments. Therefore, in this study, we evaluated the efficacy of ABZ and ATV combination therapy on E. multilocularis in culture and in vivo experiments. Protoscoleces were treated with 5…

Albendazole solid dispersions against alveolar echinococcosis: a pharmacotechnical strategy to improve the efficacy of the drug.

Alveolar echinococcosis is a neglected parasitic zoonosis caused by Echinococcus multilocularis. The pharmacological treatment is based on albendazole (ABZ). However, the low water solubility of the drug produces a limited dissolution rate, with the consequent failure in the treatment of the disease. Solid dispersions are a successful pharmacotechnical strategy to improve the dissolution profile of poorly water-soluble drugs. The aim of this work was to determine the in vivo efficacy of ABZ solid dispersions using poloxamer 407 as a carrier (ABZ:P407 solid dispersions (SDs)) in the murine intr…
