l-Amphetamine improves poor sustained attention while d-amphetamine reduces overactivity and impulsiveness as well as improves sustained attention in an animal model of Attention-Deficit/Hyperactivity Disorder (ADHD).

BACKGROUND: ADHD is currently defined as a cognitive/behavioral developmental disorder where all clinical criteria are behavioral. Overactiv … the present research was to test effects of a range of doses of the catecholamine agonists d- and l-amphetamine on ADHD-like symptoms in SHR. METHODS: The present study tested behavioral effects of 0.64 to 1.91 mg/kg d-amphetamine; and 1.27 to 3.81 mg/kg l-amphetamine base/kg i.p. in male SHRs and their controls, the Wistar Kyoto rat (WKY). ADHD-like behavio …

A review of pretreatment and analytical techniques for amphetamine type stimulants in complex samples.

Amphetamine-type stimulants (ATSs) are among the most widely abused drugs globally, posing serious threats to public health through addiction, physical and psychological harm, and associated social problems such as increased crime rates. Effective clinical management of ATSs overdose and abuse monitoring demand analytical methods that are not only sensitive, accurate, but also rapid and adaptable to real‑world settings. However, conventional detection approaches often face challenges including limited sensitivity, prolonged analysis time, insufficient selectivity in complex matrices, and poor …

AT1 receptors mediate the effects of cold stress and amphetamine on neurocognitive performance.

Brain angiotensin II (Ang II) modulates catecholamine neurotransmission via its AT1 receptors (AT1-R) and plays a critical role in neurocognitive processes. We previously showed that amphetamine (Amph) exposure induced cognitive alterations, many times unmasked by further Amph or Ang II challenge administration. In this work, we aimed to evaluate whether Amph exposure modifies the future neurocognitive response to cold stress, an environmental cue, and the possible role of the AT1-R. Male Wistar rats received AT1-R blocker (Candesartan)/Vehicle from day 0 to 5, and Amph/Saline from day 6 to 10…

Anorectic and anxiogenic actions of cocaine- and amphetamine-regulated transcript in the lateral septum.

Cocaine- and amphetamine-regulated transcript (CART) is produced in several brain regions including the hypothalamus where it is made in cells that also produce melanin-concentrating hormone (MCH). CART-expressing MCH cells densely innervate the lateral septum (LS), which integrates food- and mood-related behaviours. However, while MCH typically promotes feeding and anxiolysis, CART suppresses feeding and promotes anxiogenesis. The LS is a target site of orexigenic MCH actions, but it is not known if the actions of CART converge or oppose that of MCH in the LS. We implanted a bilateral cannula…

Impulsiveness, suicide, and aggression in a sample of patients with disorders of methyl amphetamine use.

BACKGROUND: Research has showed a link between patients with methamphetamine dependence and the risk of impulsiveness, aggression, and the risk of suicide. But, this link has not been studied in patients abusing methamphetamine, and it is unknown how impulsiveness, aggression, and the risk of suicide affect them. MATERIALS AND METHODS: This cross-sectional study included 130 adult patients diagnosed with the disorder of the use of amphetamine, methamphetamine, cannabinoids, alcohol, other substances, and polysubstance admitted in the Addiction Department for Mental Health. Participants were i…

The Role of Cocaine- and Amphetamine-Regulated Transcript (CART) in Cancer: A Systematic Review.

The functions of cocaine- and amphetamine-regulated transcript (CART) neuropeptide encoded by the CARTPT gene vary from modifying behavior and pain sensitivity to being an antioxidant. Putative CART peptide receptor GPR160 was implicated recently in the pathogenesis of cancer. However, the exact role of CART protein in the development of neoplasms remains unclear. This systematic review includes articles retrieved from the Scopus, PubMed, Web of Science and Medline Complete databases. Nineteen publications that met the inclusion criteria and describe the association of CART and cancer were ana…

The effect of environment on cross-sensitization between methylphenidate and amphetamine in female rats.

Methylphenidate (MPD) and amphetamine (AMP) are both psychostimulants that are often used to treat behavioral disorders. More recently, it has also been increasingly used illicitly for recreation as well as to improve intellectual performance. Many factors such as age, gender, genetic background, and environment govern the development of behavioral sensitization to MPD and cross-sensitization with other drugs, which are experimental behavioral markers indicating potential of substance dependence and abuse. This study examines the effects of the environment and age when MPD was exposed in adult…

G protein βγ subunits play a critical role in the actions of amphetamine.

Abnormal levels of dopamine (DA) are thought to contribute to several neurological and psychiatric disorders including drug addiction. Extracellular DA levels are regulated primarily via reuptake by the DA transporter (DAT). Amphetamine, a potent psychostimulant, increases extracellular DA by inducing efflux through DAT. Recently, we discovered that G protein βγ subunits (Gβγ) interact with DAT, and that in vitro activation of Gβγ promotes DAT-mediated efflux. Here, we investigated the role of Gβγ in the actions of amphetamine in DA neurons in culture, ex vivo nucleus accumbens (NAc), and free…

Amphetamine-induced activation of neurons within the rat nucleus of the solitary tract.

Despite generally being a reinforcing drug of abuse, amphetamine (amph) also produces effects such as hypophagia and conditioned taste avoidance (CTA), which may indicate that amph acts as an aversive homeostatic stressor. Stress-responsive prolactin-releasing peptide (PrRP)-positive noradrenergic and glucagon-like peptide-1 (GLP-1)-positive neurons in the caudal nucleus of the solitary tract (cNTS) are modulated by metabolic state, and are prime candidates for mediating amph-induced hypophagia and CTA. The present study used dual immunolabeling and fluorescent in situ hybridization (RNAscope)…

Interplay Between Amphetamine and Activity Level in Gene Networks of the Mouse Striatum.

The psychostimulant amphetamine can be prescribed to ameliorate the symptoms of narcolepsy, attention-deficit hyperactivity disorder and to facilitate weight loss. This stimulant can also have negative effects including toxicity and addiction risk. The impact of amphetamine on gene networks is partially understood and this study addresses this gap in consideration of the physical activity. The striata of mice exposed to either amphetamine or saline treatment were compared in a mouse line selected for home cage physical overactivity, a phenotype that can be mitigated with amphetamine, and in a …

Sweet taste liking is associated with subjective response to amphetamine in women but not men.

RATIONALE AND OBJECTIVE: Preference for sweet taste rewards has been linked to the propensity for drug use in both animals and humans. Here, we tested the association between sweet taste liking and sensitivity to amphetamine reward in healthy adults. We hypothesized that sweet likers would report greater euphoria and stimulation following D-amphetamine (20 mg) compared to sweet dislikers. METHODS: Men (n = 36) and women (n = 34) completed a sweet taste test in which they rated their liking of various concentrations of sucrose and filtered water (0.05, 0.10, 0.21, 0.42, and 0.83 M). Participan…

Bupropion abuse resulting in hypomania in a geriatric amphetamine user: A case report.

BACKGROUND: Bupropion is a commonly prescribed antidepressant which acts on norepinephrine and dopamine neurotransmission. It is structurally similar to amphetamine. Several cases of recreational bupropion ingestion, insufflation, and injection have been reported in the literature. METHODS: Here we report a case of bupropion abuse in a 79-year-old gentleman with a history of alcohol and amphetamine use disorders, resulting in hypertension and hypomanic symptoms. RESULTS: To our knowledge, this is the first case of bupropion abuse documented in an older adult. The literature with respect to b…
