Strains of outbred (ICR/CD1 and S--W) and inbred (BALB/C and C57BL/6) mice vaccinated subcutaneously (SQ) with 500, 1,000, or 2,000 exsheathed Heligmosomoides polygyrus larvae developed varying levels of protection upon subsequent oral challenge with larvae. In contrast, the inbred C3H/HEJ strain failed to develop protection at any dosage level tested. ICR/CD1 mice vaccinated intraperitoneally with exsheathed larvae developed a high level of resistance but exhibited extensive adhesions of the viscera. When ensheathed larvae were used for vaccination, ICR/CD1 mice developed a moderate level of

Antibodies to Nippostrongylus brasiliensis acetylcholinesterase were found in sera from infected rats and in one pooled sample were associated with IgG1. Antibodies were formed against all three isoenzymes of worm acetylcholinesterase and antisera taken from animals which had experienced multiple infections had the strongest activity, in particular to isoenzyme A. No antibodies were found against the worms' non-specific esterases, aminopeptidases or acid phosphatases. It appeared that acetylcholinesterase production by the worms was increased in the presence of antibodies and decreased in thei

BACKGROUND: Anthelmintic drugs are vital for controlling soil-transmitted helminths (STH), but treatment failures remain common. The egg reduction rate (ERR) is a key metric for assessing anthelmintic efficacy. This study calculated individual ERRs (ERRi) to evaluate a new anthelmintic drug and identify factors associated with treatment failure.
METHODOLOGY: Data were collected from 369 children infected with T. trichiura and 226 infected with hookworms, who participated in a Phase II/III clinical trial. Participants received a single dose of ivermectin/albendazole 9/400 mg or 18/400 mg fixed

BACKGROUND: Parasitic worms remain a serious public health problem in developing countries. Mebendazole is a widely used anthelmintic, but variations in drug efficacy among different brands of mebendazole may impact treatment outcomes. This trial investigated efficacy of two different brands of mebendazole (Vermox® and X-WORM®) used in the deworming program and available in local pharmaceutical shops.
METHODS: A randomized, open label trial was conducted among 212 children in Alember and Addis Zemen primary schools. Convenient sampling technique was used to select schools and study participan

Intestinal parasitic infections (IPIs) remain a public-health problem worldwide, including in countries of the Lower Mekong subregion. Increases in human migration from neighboring countries might cause reemerging parasitic infections, leading to spread of parasites in the landscape. Here, we conducted a cross-sectional study to identify the prevalence of IPIs in migrant workers from Myanmar, Lao PDR, and Cambodia who were dwelling in Nakhon Ratchasima Province, northeastern Thailand. The identification of Strongyloides species and genetic differentiation of worms from migrant workers with dif

A number of gastrointestinal parasites have been reported to infect dogs in Nigeria, some of which have zoonotic potential. Of these, hookworms are the most prevalent, with both Ancylostoma caninum and Uncinaria stenocephala reported in the country. In this study, we subjected 203 hookworm microscopy-positive samples of the 885 individual faecal samples collected from dogs in Nigeria to a recently developed multiplex qPCR for the detection and characterisation of canine hookworm species. The qPCR demonstrated a diagnostic sensitivity of 98% (95% CI 95-99.4) allowing the detection of A. caninum

BACKGROUND: National mapping of soil-transmitted helminth infections (STH) was conducted for the first time in all of the 77 districts of Benin (West Africa) from 2013 to 2015. This mapping aimed to provide basic epidemiological data essential for the implementation of the national strategy against the neglected tropical diseases (NTDs) in the context of achieving the WHO target of controlling these infections by 2020.
METHODS: In each district, 5 schools were purposively selected in 5 villages and 50 school-children (25 girls and 25 boys) from ages 8 to 14 years were randomly enrolled in eac

We created a mobile phone microscope and assessed its accuracy for the diagnosis of soil-transmitted helminths compared with conventional microscopy. Mobile phone microscopy has a sensitivity of 69.4% for detecting any helminth egg and sensitivities of 81.0%, 54.4%, and 14.3% for the diagnosis of Ascaris lumbricoides, Trichuris trichiura and hookworm respectively.

The Klato-Katz method is widely used for diagnosing helminth infections in epidemiological surveys, but is known to have a low sensitivity. In the case of Schistosoma mansoni, statistical methods have been developed to compensate for the poor sensitivity, but the same is not true of any other helminth parasite, or infections with multiple-helminth species. We screened 101 schoolchildren from a rural area of Côte d'Ivoire over 5 consecutive days and made 5 Kato-Katz readings from each stool specimen. We estimated single and dual-species infections with S. mansoni and hookworm based on raw egg c

Mesenteric lymph node cells obtained from highly inbred donor rats (Lewis strain), resistant to Nippostrongylus brasiliensis infection, were syngeneically transferred by intravenous injection into previously uninfected recipients. The adoptively immunized recipients were then challenged with either 1500 or 3000 third stage N. brasiliensis larvae on the day of cell transfer. The degree of resistance transferred was assessed by monitoring daily faecal egg output, differential worm burdens on days 6 and 10 of infection and the number of eggs per uterus in gravid worms. The syngeneic transfer of 1

The potentiated reagin response induced by infection with the nematode Nippostrongylus brasiliensis after sensitization with egg albumin and Bordetella pertussis vaccine was investigated to reveal the time course of the potentiated reagin response and the maximum reagin titre attained. The potentiated reagin response was found to be transitory but during this period high reagin titres to egg albumin were recorded. Rat sensitized to egg albumin and treated with N. brasiliensis crude worm extract instead of an infection did not show a potentiated response.

BACKGROUND: The results of the latest national survey on important human parasitic diseases in 2015-2016 showed Guangdong Province is still a moderately endemic area, with the weighted prevalence of soil-transmitted helminths (STHs) higher than the national average. High-resolution age- and gender-specific spatial-temporal risk maps can support the prevention and control of STHs, but not yet available in Guangdong.
METHODOLOGY: Georeferenced age- and gender-specific disease data of STH infections in Guangdong Province was derived from three national surveys on important human parasitic diseas

BACKGROUND: Despite the existence of a population-based control program using single dose albendazole or mebendazole as a preventive chemotherapy, hookworm transmission remains high. It causes a negative impact on the growth and school performance of children. In connection to this preventive chemotherapy, different studies produced conflicting results. This study aimed at evaluating the efficacy of single (500 mg) versus multiple doses (100 mg twice a day during three consecutive days) of mebendazole against hookworm infections among school-aged children.
METHODS: This randomized open-label

BACKGROUND: Soil-transmitted helminths are more prevalent in tropics and sub-tropics including Ethiopia. Despite their high prevalence, direct saline microscopy with its low sensitivity has been used as a diagnostic method in almost all health facilities in Ethiopia. Alternative diagnostic methods which have higher sensitivity are not yet implemented. Therefore, this study aimed to compare and evaluate the performance of diagnostic methods for soil transmitted helminths.
METHODS: A cross-sectional study among 520 school children was conducted from October to December, 2019 in Amhara National

Strongyloidiasis is caused by the human infective nematodes Strongyloides stercoralis, Strongyloides fuelleborni subsp. fuelleborni and Strongyloides fuelleborni subsp. kellyi. The zoonotic potential of S. stercoralis and the potential role of dogs in the maintenance of strongyloidiasis transmission has been a topic of interest and discussion for many years. In Australia, strongyloidiasis is prevalent in remote socioeconomically disadvantaged communities in the north of the continent. Being an isolated continent that has been separated from other regions for a long geological period, descripti
