BACKGROUND: Nut consumption has been associated with reduced cancer-related mortality. However, it is unclear whether nut consumption also reduces the risk of esophageal and gastric cancer subtypes. We prospectively investigated the relationship of tree nut, peanut, and peanut butter intake with risk of esophageal squamous cell carcinoma (ESCC), esophageal adenocarcinoma (EAC), gastric cardia adenocarcinoma (GCA), and gastric non-cardia adenocarcinoma (GNCA) in the Netherlands Cohort Study.
METHODS: In 1986, 120,852 males and females, aged 55-69 years, completed a baseline questionnaire on di

BACKGROUND: Serum tryptase is useful in diagnosing drug and venom anaphylaxis. Its utility in food anaphylaxis is unknown.
OBJECTIVE: The objective of this study was to determine whether tryptase rises in food allergic reactions, optimal sampling time points, and a diagnostic cutoff for confirming a clinical reaction.
METHODS: Characterized peanut allergic patients were recruited and underwent up to 4 peanut challenges and 1 placebo challenge each. Tryptase was measured serially on challenge days both before (baseline) and during the challenge. The peak percentage tryptase rise (peak/baselin

BACKGROUND: Food allergies are increasing in prevalence but no treatment strategies are currently available to cure dogs with food allergy. Over the past decade, experimental food allergen-specific sublingual immunotherapy (FA-SLIT) has emerged as a potential treatment for food allergies in human medicine. However, FA-SLIT has not been investigated in dogs. Therefore, the objective of this study was to prospectively evaluate the safety, tolerability and dispenser sterility of FA-SLIT in healthy dogs before testing it in food allergic dogs. Eight experimental healthy beagle dogs, never orally e

BACKGROUND: There has been a growing interest in measuring allergen-specific immunoglobulin E (sIgE) directly from the nasal mucosal epithelium of individuals with sinonasal disease. However, there is currently no normative data with which to estimate sensitivity, specificity, and the appropriate positive cutoff level for new testing methods.
METHODS: Twenty individuals with no history of sinonasal disease or food allergies underwent serum IgE testing and nasal mucosal brush biopsy (MBB) testing for total and sIgE to 7 common airborne allergens and 1 food allergen. The correlation between sIg

BACKGROUND: Peanut allergies are common and can be life-threating for sensitised individuals. Peanut allergens share significant amino acid homology with those of other legumes and tree nuts, but their cross-reactivity still remains unclear.
OBJECTIVE: We sought to determine the clinical significance of the cross-reactivity of peanut allergens with those of walnut and soybean.
METHODS: Pooled sera from eight subjects with both peanut and walnut specific IgE were investigated in an inhibition test. After the sera were incubated with either peanut or walnut protein extracts, the quantity of Ig

BACKGROUND: Peanut allergy is one of the most common forms of food allergy encountered in clinical practice. In most cases, it does not spontaneously resolve; furthermore, it is frequently implicated in acute life-threatening reactions. The current management of peanut allergy centres on meticulous avoidance of peanuts and peanut-containing foods. Allergen-specific oral immunotherapy (OIT) for peanut allergy aims to induce desensitisation and then tolerance to peanut, and has the potential to revolutionise the management of peanut allergy. However, at present there is still considerable uncer

Cutoffs (decision points) of the peanut skin prick test (SPT) and specific IgE level for predicting peanut allergy have been proposed. It is not known whether decision points indicating a significant risk of severe reactions on challenge differ from those indicating probable allergy. We aimed at determining the usefulness of allergy tests for predicting the risk of anaphylaxis on challenge following the ingestion of up to 12 g of peanut in peanut-sensitized children. Children attending the Allergy Clinic who had a positive peanut SPT and completed open-label in-hospital peanut challenges were

BACKGROUND: Oral immunotherapy (OIT) offers a promising therapeutic option for peanut allergy. Given that during OIT an allergic patient ingests an allergen that could potentially cause a serious reaction, the safety of OIT is of particular concern.
OBJECTIVE: The purpose of this study was to examine safety during the initial escalation day, buildup phase, and home dosing phase in subjects enrolled in a peanut OIT study.
METHODS: Skin, upper respiratory tract, chest, and abdominal symptoms were recorded with initial escalation day and buildup phase dosings. Subjects also maintained daily dia

BACKGROUND: Dietary exposure to high levels of the fungal toxin, aflatoxin, occurs in West Africa, where long-term crop storage facilitates fungal growth.
METHODS: We conducted a cross-sectional study in Benin and Togo to investigate aflatoxin exposure in children around the time of weaning and correlated these data with food consumption, socioeconomic status, agro-ecological zone of residence, and anthropometric measures. Blood samples from 479 children (age 9 months to 5 years) from 16 villages in four agro-ecological zones were assayed for aflatoxin-albumin adducts (AF-alb) as a measure of

Acute reaction to food allergens is a fairly common problem that is often seen in the allergist's office, its incidence being specially high in childhood. Milk and eggs are the most common sensitizing foods, but usually the type of food allergens responsible for these reactions varies according to food habits in different countries. Legumes occupy an important role in the Spanish diet, being responsible for a large number of allergic reactions. It has been shown that legumes occupy the fourth place in importance among the food allergens, inducing hypersensitivity reactions in Spanish children.

The diagnosis of food allergy is often difficult to make by conventional means. Histories are frequently ambiguous, and skin testing is of dubious reliability because of the number of false-positive and false-negative reactions. We have evaluated the radioallergosorbent test (RAST) for the in vitro measurement of the specific IgE antibodies to nuts, including Brazil nut, almond, walnut, pecan, cashew, and the legume, peanut. Serums were obtained from 18 patients with a history of nut allergy and IgE level and specific IgE antibodies were measured. Thirteen of the 18 patients had significantly

BACKGROUND: Peanut (PN) allergy is an IgE-mediated hypersensitivity with a deviated adaptive immune response dominated by IgE-producing B cells and type 2 T cells (Th2). The in vivo immune response upon antigen encounter, for example, after oral food challenge (OFC), has not been investigated over prolonged timeframes.
METHODS: Here, we investigated the kinetics of allergen-specific adaptive immune responses in PN-allergic and non-allergic adults before OFC (d0) and thereafter at d7, d14, d21, d28 by determining PN-specific CD40L+4-1BB+ conventional (Tconv) and FoxP3+ Helios+ CD40L-4-1BB+ reg

Homeodomain-leucine zipper (HD-Zip) transcription factors play important roles in plant growth, development, and abiotic stress responses. However, bioinformatic analyses and functional studies of HD-Zip family in peanut are scarce. In this study, 128 AhHDZ genes were identified and classified into four subfamilies in the phylogenetic analysis. Transcriptomic data and RT-qPCR analysis indicated the expression levels of AhHDZ4 and AhHDZ15 were significantly elevated in response to 12 h of salt stress, while AhHDZ4/15/60/69/126 all showed a progressive increase over time in response to drought s

Peanut scab, caused by Elsinoë arachidis, is a major disease in peanut-growing regions of China. To clarify the phenotypic characteristics and toxigenic divergence of the pathogen, 70 strains from major peanut-growing regions were subjected to morphological characterization, phylogenetic analysis, elsinochrome (ESC) quantification, and phytotoxicity assessment. Based on colony color, the strains were divided into five morphological groups (Group A-E), with the dark red pigmented Group D being dominant (50%). Growth rates varied among strains, with the coefficient of variation within each morph

Peanut peptides (PP) are promising plant-derived functional ingredients, but bitterness and gastrointestinal instability limit oral applications. This study integrated complex enzymolysis, LC-MS/MS-assisted in silico screening, molecular dynamics (MD), and W1/O/W2 double-emulsion microencapsulation to establish a laboratory-scale discovery-to-delivery workflow for antioxidant PP. Under optimized alkaline protease/flavor protease hydrolysis (5:5, w/w; 10% enzyme; 59 °C; 9 h), LC-MS/MS identified 763 peanut-derived peptides. Stepwise screening using PeptideRanker (>0.5), ToxinPred3.0 (<0.5), and
