A call for neurovascular monitoring in an era of longer missions and broader spaceflight participation. Commentary.

Spaceflight-Associated Neuro-ocular Syndrome (SANS) has emerged as a critical neuro-ophthalmic risk for human space exploration, particularly as mission duration increases and access to space expands. Current spaceflight ocular surveillance and research protocols have prioritized structural imaging and selected neuroimaging/physiological assessments. However, accumulating evidence suggests that SANS is not confined to the posterior pole as a purely structural optic nerve head phenomenon but may also involve vascular and hemodynamic alterations. At the same time, structural changes at the optic

A novel mineral-organic bone adhesive promotes calvarial bone regeneration in microgravity conditions.

Microgravity significantly impacts astronaut physiology, causing accelerated bone loss and impairing bone regeneration. As human space exploration expands, effective and minimally invasive bone repair treatments are crucial. In this study, we investigated the regenerative potential of a novel mineral-organic bone adhesive, Tetranite® (TN), compared to the osteoinductive rhBMP2 (Infuse®). We used a critical-size calvarial bone defect model in mice, with half launched on a 60-day mission to the International Space Station and the other half serving as ground controls. Histological and quantitati

An eye on long-duration spaceflight: Controversies, countermeasures and challenges.

Space flight-associated neuroocular syndrome (SANS) is a consequence of long-duration space flight and is detected in two-thirds of astronauts. In-flight, this can cause a change in the refraction of the eyes, requiring graded hypermetropic 'superfocus adjustable' glasses, optic nerve head oedema and choroidal folds. While the optic disc oedema resolves on returning to gravitational force, the choroidal folds and axial length shortening remain. Controversy remains over the role of intracranial pressure in the development of these changes. A recent case report has re-energised the debate as to …

Biological effects of space radiation on human cells: history, advances and outcomes.

Exposure to radiation is one of the main concerns for space exploration by humans. By focusing deliberately on the works performed on human cells, we endeavored to review, decade by decade, the technological developments and conceptual advances of space radiation biology. Despite considerable efforts, the cancer and the toxicity risks remain to be quantified: 1) the nature and the frequency of secondary heavy ions need to be better characterized in order to estimate their contribution to the dose and to the final biological response; 2) the diversity of radiation history of each astronaut and

Hormonal responses of metoclopramide-treated subjects experiencing nausea or emesis during parabolic flight.

Metoclopramide was orally administered (10 or 20 mg) to 22 subjects, 75 min before parabolic flight. Serum levels of ACTH, EPI, NE, and vasopressin (AVP) were unaltered by metoclopramide. AVP and ACTH (1.2 and 36 pg.ml-1) were elevated 77 and 3.8-fold (92.3 and 135 pg.ml-1) following emesis, after 40 parabolas (68.7 and 140 pg.ml-1) and landing (8.7 and 79 pg.ml-1). Seven subjects displaying no nausea and no emesis demonstrated smaller elevations (8.2 and 2.2-fold). Of 15 vomiting subjects, 7 reported no nausea and had lower elevations of AVP with faster recoveries. These findings are consiste

[These vestibular problems in the absence of gravity...].

For a few years, more and more astronauts complain to endure space motion sickness during the two or three first days of their mission. This is due to the repetition of shifting and sudden head movements, which becomes possible by the increasing of volume of the new space stations. To avoid that payload specialists onboard be obliged to renounce to conduct planned experiments, it has been necessary to find effective solutions to detect by ground based tests the candidates sensitive to space motion sickness and perfect therapeutic means able to avoid unexpected arrival of these symptoms, and ev…

Motion sickness: part II--a clinical study based on surgery of cerebral hemisphere lesions.

Man has always been intrigued with the localization of function within the brain but has paid insufficient attention to the long and the sho … s. The symptoms described above were reminiscent of those experienced by some of the cosmonauts and astronauts and it called the authors' attention to this "motion sickness in space." The areas from which such responses may be elicited are the temporoparieto-occipital regions, which are nourished by the posterior cerebral artery and its branches. Vascular in …

Food intake and nutritional status during EXEMSI. Experimental Campaign for the European Manned Space Infrastructure.

Modifications of food intake by astronauts during long-duration spaceflights have been observed. Various psychological stress factors, such as isolation, confinement, constrained community and boredom, are thought to play a role in this phenomenon. For this reason it was decided to include a nutritional investigation in the EXEMSI simulation study, in which four crew members (1 female and 3 males) were isolated and confined for 60 days in a space station-like environment. The Food and Nutritional Management System, developed for this experiment, provided on-line analysis of all available foods…

Imaging in spaceflight associated neuro-ocular syndrome (SANS): Current technology and future directions in modalities.

With plans for future long-duration crewed exploration, NASA has identified several high priority potential health risks to astronauts in space. One such risk is a collection of neurologic and ophthalmic findings termed spaceflight associated neuro-ocular syndrome (SANS). The findings of SANS include optic disc edema, globe flattening, retinal nerve fiber layer thickening, chorioretinal folds, hyperopic shifts, and cotton-wool spots. The cause of SANS was initially thought to be a cephalad fluid shift in microgravity leading to increased intracranial pressure, venous stasis and impaired CSF ou…

Cardiovascular response to lower body negative pressure stimulation before, during, and after space flight.

BACKGROUND: It is well known that space travel cause post-flight orthostatic hypotension and it was assumed that autonomic cardiovascular control deteriorates in space. Lower body negative pressure (LBNP) was used to assess autonomic function of the cardiovascular system. METHODS: LBNP tests were performed on six crew-members before and on the first days post-flight in a series of three space missions. Additionally, two of the subjects performed LBNP tests in-flight. LBNP mimics fluid distribution of upright posture in a gravity independent way. It causes an artificial sequestration of blood,

Rendering knowledge graphs from aerospace dentistry processes for clinical decision support systems.

NASA and commercial space flight companies expect to make long-term missions into deep space with the Artemis project and Mars mission. Health care in deep space is a priority as the missions require limited crew and equipment to accommodate astronauts' needs. Analyzing the probable clinical procedures and workflows for this setting is needed to ensure quality care and safety. This paper introduces Business Process Modeling and Notation (BPMN) and OWL2-based modeling to represent aerospace dental workflows for low-earth orbit and potential deep space missions. We developed BPMN process models …

Artificial Gravity Attenuates the Transcriptomic Response to Spaceflight in the Optic Nerve and Retina.

The development of eye pathology is a serious concern for astronauts who spend time in deep space. Microgravity is a major component of the spaceflight environment which could have adverse effects on ocular health. The use of centrifugation to exert forces that partially or fully mimic Earth-level gravity in space is a possible countermeasure to mitigate the effects of microgravity on the eye. Therefore, we subjected mice on the International Space Station (ISS) to microgravity (0 G) or artificial gravity by centrifugation at 0.33 G, 0.67 G, and 1 G, and then performed RNA sequencing (RNA-seq)…

Monitoring the Impact of Spaceflight on the Human Brain.

Extended exposure to radiation, microgravity, and isolation during space exploration has significant physiological, structural, and psychosocial effects on astronauts, and particularly their central nervous system. To date, the use of brain monitoring techniques adopted on Earth in pre/post-spaceflight experimental protocols has proven to be valuable for investigating the effects of space travel on the brain. However, future (longer) deep space travel would require some brain function monitoring equipment to be also available for evaluating and monitoring brain health during spaceflight. Here,…

The lack of mass transfer in bone lacunar-canalicular system may be the decisive factor of osteoporosis under microgravity.

During spaceflight, astronauts experience 1-1.5% bone loss per month, especially in the lumbar spine, pelvis and lower limbs. The bone loss leads to osteoporosis and increased the risk of fracture. Current researches focus on anti-osteoporosis under microgravity mainly by inhibiting bone resorption of osteoclasts and / or increasing bone formation of osteoblasts. However, studies on the effects of mass transfer in the bone lacunar-canalicular system (LCS) on osteoporosis are lacking. Osteocytes reside in the lacunae and communicate with other osteocytes, osteoblasts and osteoclasts through the…
