PROBLEM: Adenomyosis (AM) is associated with immune response and inflammation. However, the role of T cell subsets in AM development has not been thoroughly understood.
METHOD OF STUDY: Patients with focal or diffuse AM were recruited. Serum cytokines were quantified by enzyme-linked immunosorbent assay (ELISA). Different T cell subsets in the blood and ectopic endometrium were determined by flow cytometry.
RESULTS: Serum interleukin-6 (IL-6) and macrophage-colony-stimulating factor (GM-CSF) were increased in patients with focal or diffuse AM before focused ultrasound ablation surgery (FUAS)

COVID-19 is known to impair red blood cell (RBC) function, which may affect oxygen transport and disease progression. However, the metabolic consequences of SARS-CoV-2 infection on RBCs and how these changes relate to disease severity and sex differences, have not been systematically explored. Here, we compared RBC metabolomic profiles from healthy controls and individuals with COVID-19 using nuclear magnetic resonance (NMR) to gain insight into disease mechanisms and potential biomarkers. Multivariate and univariate analyses were performed within the men and women cohorts, and clinical factor

Precise control of micro-/nano-scale objects in vivo is crucial for biomedical applications. Here, we demonstrate a biohybrid optical manipulation strategy that integrates a long-distance manipulation fiber probe (LDMFP) and natural red blood cells (RBCs) to construct endogenous biotweezers for optical micromanipulation in vivo. By using the engineered LDMFP, an extended focal field was generated, which could capture flowing RBCs within the living blood vessel. Benefiting from their smooth disk-shape and homogenous refractive index distribution, these trapped RBCs can function as natural biotw…

Red blood cells (RBCs) are the most abundant human cell type, and interactions with the RBC membrane are at the heart of many processes relevant for human health, such as immune system modulation, as well as binding by foreign pathogens and pharmacological drugs. To better study such membrane interface interactions, it would be useful to employ surface-attached model lipid membranes derived from RBCs to enable surface-sensitive biophysical and biochemical measurements. Here, we present approaches to prepare two such types of RBC-derived model lipid membranes─tethered RBC liposomes and supporte…

Gaucher disease (GD), the most common lysosomal storage disorder, alters red blood cell (RBC) mechanics and circulation, contributing to vascular occlusions, bone infarcts, and splenomegaly. However, the individual roles of GD-RBC biophysical properties in these processes remain unclear. Here, we present a combined computational-experimental investigation to quantitatively characterize GD-RBC biophysical properties and determine how specific mechanical parameters drive abnormal RBC behavior. Informed by experimental data, we independently quantify key RBC properties, including shear modulus (

SARM1 (sterile α and TIR motif-containing protein-1) is an NADase enzyme that serves as the central executioner of Wallerian axon degeneration. Given this, SARM1 is of high interest as a candidate therapeutic target, and SARM1 inhibitors are currently in clinical trials for treatment of neurodegeneration. Beyond neuroscience, emerging studies reveal that SARM1 may also drive aspects of bone fragility, liver pathology, adipose expansion, and insulin resistance in metabolic disease. However, we lack methods to quantify SARM1 activation in humans to better define patients at high risk of SARM1-me

SIGNIFICANCE: Standard methods to characterize peripheral blood mononuclear cells (PBMCs) are often destructive, lack metabolic information, or do not provide single-cell resolution. Label-free tools that nondestructively measure single-cell metabolism within PBMCs can provide new layers of information to characterize disease state and cell therapy potential.
AIM: We aim to determine whether nondestructive fluorescence lifetime imaging microscopy (FLIM) of the endogenous metabolic cofactors nicotinamide adenine dinucleotide (phosphate) (reduced form) and flavin adenine dinucleotide (oxidized

The placental intervillous space is a unique immunological niche where circulating maternal immune cells come into direct contact with the fetal syncytiotrophoblast. While adaptations in immune cell composition are known to occur in the maternal decidua throughout pregnancy, it remains unclear whether similar changes take place in the intervillous space. Here, we demonstrate that the intervillous immune cell composition undergoes dynamic changes during pregnancy, with a decreased proportion of NK cells and an increased proportion of T cells from second trimester to term pregnancy. Interestingl

Red blood cell (RBC) deformability is a critical biophysical property that enables effective passage of RBCs through microvasculature and ensures proper oxygen delivery. Impairment of this property is associated with various pathological conditions, including type 2 diabetes mellitus (T2DM). In this study, we developed an automated microfluidic platform for high-throughput and real-time assessment of RBC deformability under controlled flow conditions. The device features a structured microchannel design and integrated imaging to quantify individual cell deformation responses. Comparative analy

The potential role of selenium (Se) in modulating cadmium (Cd) cytotoxicity in human peripheral blood mononuclear cells (PBMCs) was investigated by assessing pro- and anti-inflammatory as well as redox biomarkers. We also examined the potential synergistic effects of SARS-CoV-2 N-protein with Cd in PBMCs. PBMCs were isolated from adults and treated for 48 h with sodium selenite (Na₂SeO₃; 0.04, 0.4, and 4 µM) and cadmium chloride (CdCl₂; 0.1 and 1 µM), in the presence or absence of SARS-CoV-2 N-protein. We measured total Se and Cd levels, cell viability, cytotoxicity, intracellular reactive oxy

Red blood cells (RBCs) have emerged as promising carriers for therapeutic and diagnostic agents due to their long circulation time, biocompatibility, and immune-evasive properties. Hypotonic encapsulation is the most widely employed technique; however, the correlation between nanoparticle sizes and the hypotonic osmolarities required for efficient encapsulation remains unclear. In this study, the size-dependent osmolarity requirements governing nanoparticles into RBCs are investigated. Using monodisperse gold nanoparticles as model systems, the optimal osmolarity correlated with nanoparticle d…

Umbilical cord blood (CB)-derived hematopoietic stem and progenitor cells (HSPCs) hold immense potential for regenerative medicine, particularly in hematologic malignancies and immune disorders. CB offers several advantages, including easy collection and reduced risk of graft-versus-host disease compared to other sources, like bone marrow. However, the clinical application of CB is often limited due to the relatively small number of HSPCs present in CB grafts, which can be insufficient for adult patients. This limitation has prompted researchers to explore various methods to expand HSPCs ex vi

INTRODUCTION: Illicit drug use, particularly the synthetic opioid fentanyl, presents a significant global health challenge. Previous studies have shown that fentanyl enhances viral replication; yet, the mechanisms by which it affects HIV pathogenesis remain unclear. This study investigated the impact of fentanyl on HIV replication in CD4+ T lymphocytes.
METHODS: CD4+ T lymphocytes from HIV-negative donors were activated, infected with HIVNL4-3, and treated with fentanyl. HIV proviral DNA and p24 antigen expression were quantified using real-time PCR and ELISA, respectively. Single-cell RNA li

Neurodegenerative diseases (ND) are characterized by progressive loss of neuronal function. Mechanisms of ND pathogenesis are incompletely understood, hampering the development of effective therapies. Langerhans cell histiocytosis (LCH) is an inflammatory neoplastic disorder caused by hematopoietic progenitors expressing MAPK activating mutations that differentiate into senescent myeloid cells that drive lesion formation. Some patients with LCH subsequently develop progressive and incurable neurodegeneration (LCH-ND). Here, we show that LCH-ND is caused by myeloid cells that are clonal with pe

Stroke is the leading cause of severe long-term disability. Cell therapy has recently emerged as an approach to facilitate functional recovery in stroke. Although administration of peripheral blood mononuclear cells preconditioned by oxygen-glucose deprivation (OGD-PBMCs) has been shown to be a therapeutic strategy for ischemic stroke, the recovery mechanisms remain largely unknown. We hypothesised that cell-cell communications within PBMCs and between PBMCs and resident cells are necessary for a polarising protective phenotype. Here, we investigated the therapeutic mechanisms underlying the e

Conventional and regulatory T cells (Treg) are dynamic mediators of maternal immune tolerance to the developing feto-placental unit. Functional evaluation of T cells at the maternal-fetal interface is crucial to elucidate the immunologic basis of obstetric complications. Our objective was to define the T cell phenotype and function of uterine intervillous blood (IVB) in pregnancy with and without preeclampsia. We hypothesize that preeclampsia is associated with impaired immune tolerance and a pro-inflammatory uterine T cell microenvironment. In this cross-sectional study, maternal peripheral b
