This review summarizes the time that has passed from the initial registration of the cells that turned out to be platelets up to today's advanced methodologies in platelet investigation. The first reports of "granular masses" appeared in the 1840s, but these "granular masses" remained an unsolved mystery until the 1870s. The breakthrough came in the 1873-1882 period. The cells that later turned out to be platelets were further identified by the German Professor Max Schultze, and later by Osler, who described their disk-like structure. These initial descriptions of platelets were expanded by im

Heatstroke-induced coagulopathy (HSIC) is a life-threatening complication of heatstroke that often progresses to disseminated intravascular coagulation. Despite its prognostic significance, standardized treatment strategies have not yet been established. This scoping review aimed to systematically map existing treatment approaches and outcome measures for HSIC in both clinical and experimental research. We conducted a scoping review in accordance with PRISMA-ScR and the Joanna Briggs Institute methodology. Literature searches were conducted in MEDLINE, Web of Science, and Scopus on June 2, 202

Disseminated intravascular coagulation can occur due to different causes but commonly following sepsis. Trauma-induced coagulopathy (TIC) occurs on hospital arrival in approximately 25% of seriously injured patients who initially presents with impaired hemostasis and a bleeding phenotype that can later progress to a prothrombotic phase. Following traumatic injury, ineffective hemostasis is driven by massive blood loss, tissue damage, and hyperfibrinolysis. This initial impaired hemostasis continues until surgical or other management strategies not only to stop the causes of hemorrhage but also

Disorders of coagulation in children often prove challenging to the medical care team. The aims of this study were to assess the spectrum and prevalence of coagulation disorders among children attending Mansoura University Children Hospital (MUCH), Mansoura, Egypt. A total of 105 pediatric patients were referred to MUCH. They were divided into two groups: congenital coagulation disorders (75 cases, age 45.36 +/- 48.59 months), and acquired coagulation disorders (30 cases, age 56.13 +/- 61.61 months). All patients were subjected to thorough history taking including the nature of bleeding, famil

Regulatory mechanisms responsible for limiting blood clot formation are critical for maintaining normal haemostasis. Dysregulation can lead to bleeding (e.g. haemophilia) or thrombosis. New findings showing that tissue factor pathway inhibitor-alpha (TFPIα) binds coagulation factor V(a) and inhibits prothrombinase assembly highlights that our understanding of the initiation of coagulation is evolving. Work over the past decade on the biochemistry of FV activation has laid the groundwork for deciphering the mechanistic bases that may underpin how TFPIα mediates these anticoagulant effects. Coll

The authors give an account on the relations between disseminated intravascular coagulation, sepsis and multiorgan failure. Disseminated intravascular coagulation is defined as an acquired disorder of blood clotting with an increased turnover of thrombocytes, fibrinogen and coagulation factors. The authors discuss aetiopathogenetic aspects, possibilities of laboratory diagnosis, anticoagulation and substitution therapy and prophylaxis of disseminated intravascular coagulation in septicaemia. They emphasize comprehensive treatment of septicaemias where haematological monitoring and therapy must

INTRODUCTION: There are significant differences in the activated partial thromboplastin time (APTT) critical values reported in different studies, most of which does not make recommendations for any specific clear detection systems. The International Council for Standardization in Hematology (ICSH) recommends that APTT critical values be established based on the reagent type, coagulation factor sensitivity and heparin response. The objective of this study was to establish APTT critical values by using different reagents and based on single coagulation factor deficiencies.
METHODS: The APTT va

Perioperative and periprocedural bleeding complications substantially increase the morbidity and mortality. The main causes are intervention-related or trauma-related factors, patient-related factors, such as congenital or acquired hemostatic disorders as well as pre-existing medicinal anticoagulation as a result of the underlying disease. Despite the variety of currently available test procedures, the constellation of a clinically relevant bleeding situation without tangible pathological laboratory findings or causes often occurs. This constellation, called bleeding disorder of unknown cause

Dysregulated innate immunity participates in the pathomechanisms of disseminated intravascular coagulation (DIC) in trauma-induced coagulopathy. Accidental and regulated cell deaths and neutrophil extracellular traps release damage-associated molecular patterns (DAMPs), such as histones, nuclear and mitochondrial DNA, and high-mobility group box 1, into circulation immediately after trauma. DAMP-induced inflammation activation releases tissue factor-bearing procoagulant extracellular vesicles through gasdermin D-mediated pore formation and plasma membrane rupture by regulated cell death. DAMPs

Disseminated intravascular coagulation (DIC) is a life-threatening complication in sepsis and other critical conditions. The International Society on Thrombosis and Haemostasis (ISTH) released the diagnostic criteria for overt DIC in 2001. Since then, ISTH overt DIC has been used as the global standard criterion for a decompensated stage of DIC. Because detecting an earlier stage of DIC would be useful for therapeutic considerations, the scientific standardization committees of the ISTH introduced the sepsis-induced coagulopathy (SIC) scoring system in 2019. The SIC scoring system is specifica

One of the significant complications of severe COVID-19 infections is a coagulopathy that seems to be related to the occurrence of venous and arterial thromboembolic disease. The coagulation changes mimic but are not identical to disseminated intravascular coagulation (DIC). The vast majority of patients with COVID-19 do not meet the criteria for usual forms of DIC. In addition, there seem to be features of a strong local pulmonary thrombotic microangiopathy and direct endothelial cell infection and injury by the virus that affect the coagulopathic response to severe COVID-19. It seems COVID-1

Hemostasia is a defense mechanism that protects vascular integrity, avoids blood loss, and maintains blood fluidity throughout the circulatory system. The biochemical processes leading to blood clot formation are complex, and alterations can appear at any point within the chain of events. While a range of alterations can affect the coagulation factors, some are more common than others in the general population, including congenital (hemophilia A and B, Von Willebrand's disease) and acquired disorders (anticoagulant drugs). Such diseases require special consideration in the context of dental tr

Platelets are anucleated cells produced by megakaryocytes, from which they inherit all the components necessary to carry their functions. They circulate in blood vessels where they play essential roles in coagulation, wound repair or inflammation, and have been implicated in various pathological conditions such as thrombosis, viral infection or cancer progression. The importance of these cells has been established over a century ago, and effective anti-platelet medications with different mechanisms of action have since been developed. However, these therapies are not always effective and can i

Seminars in Thrombosis and Hemostasis (STH) celebrates 50 years of publishing in 2024. To celebrate this landmark event, STH is republishing some archival material. This manuscript represents the second most highly cited paper ever published in STH. The manuscript published without an abstract, and essentially represented a State of the Art Review on the bleeding time, a relatively invasive procedure that required an incision on the skin or earlobe of a patient, and timing how long it took for the incision to stop bleeding. The bleeding time test was first described in 1901 by the French physi

The most common clinical and biological expressions of constitutional abnormalities of haemostasis on which rests the diagnostic approach are influenced by the level where deficiency occurs as well as by the degree of this deficiency and its mode of transmission. Four situations encountered in practice are described: a severe form with loud and early clinical expression, a moderate form with subdued clinical expression but potential risk of haemorrhage, a genetic enquiry and the discovery of a biological abnormality. Concerning the first two situations, the stress is placed on their clinical s
