Two modifications of macrophage migration inhibition test, one of George and Vaughan and the other one of Svejcar, were performed on a total of 78 cases of untreated syphilis at various stages. Barth Syndrome (BTHS) is an X-linked mitochondrial cardioskeletal myopathy caused by defects in TAFAZZIN, a gene responsible for cardiolipin remodeling. The first total synthesis of cyclic depsipeptide antibiotic LL-A0341β1 (LL) is described. The configuration of the β-methyltryptophan (β-MeTrp) residue was established by preparing all four stereoisomers of Fmoc-β-MeTrp which were used for the synthesis of LL via Fmoc solid phase peptide synthesis. The most active of the four peptides was the one containing (2R,3R)-β-MeTrp. The activity of LL was strongly inhibited by cardiolipin (CL), but not other common phospholipids found in bacterial cell membranes, suggesting that LL interacts with CL in the lipid membrane.

Monozygotic twins, 27 years old, with primary anti-phospholipid syndrome have been described. The first of the twins had obstetric morbidities (one miscarriage at 12 weeks and premature birth of a growth-restricted fetus at 27 weeks secondary to pre-eclampsia) and the second had venous thrombosis. Lupus anticoagulant was present in both and anti-cardiolipin antibodies were present in high titres when re-tested at appropriate intervals. Development of clinical features of APS in monozygotic twins points to an underlying genetic basis in the pathogenesis of anti-phospholipid syndrome.

BACKGROUND AND PURPOSE: Carvedilol is a third-generation β-adrenoceptor antagonist, which also stimulates β-arrestins. β-arrestins initiate intracellular signalling and are involved in insulin release and sensitivity. Carvedilol is superior in effectiveness to other drugs that are used for similar indications and does not cause insulin resistance or diabetes, which can occur with other β-antagonists. We have shown that carvedilol increased glucose usage in C2C12 cells. We investigate the biased agonist efficacy of carvedilol on β-arrestins.

BACKGROUND: Barth syndrome (BTHS) is an X-linked recessive disorder characterized by cardiomyopathy, skeletal myopathy and cyclic neutropenia in male patients. It is caused by mutations in the TAZ gene coding for the tafazzin, a protein involved in the remodeling of cardiolipin. Loss of cardiolipin in the inner mitochondrial membrane results in respiratory chain dysfunction. No specific symptom has been identified in female carriers.
