Axillary lymph node involvement remains one of the strongest prognostic factors in early breast cancer and continues to guide both surgical and adjuvant treatment decisions. Historically, axillary lymph node dissection (ALND) represented the standard staging approach, but it is associated with substantial morbidity, including a significant risk of lymphedema. Over recent decades, robust evidence from multiple surgical de-escalation trials has demonstrated that less invasive approaches-most notably sentinel lymph node biopsy (SLNB) and omission of ALND in selected scenarios-have significantly r

BACKGROUND/AIM: The tumor microenvironment is an indispensable component of tumor development. Although fibroblasts and immune cells have been widely studied in tumorigenesis, the role of neurons in cancer has received limited investigation. This study aimed to evaluate the impact of neural signaling on tumorigenesis across cancer types and identify potential therapeutics for neuron-associated tumor development.
MATERIALS AND METHODS: A systematic analysis was performed using the Neuro_Cancer database (https://bio-bigdata.hrbmu.edu.cn/Neuro_Cancer/) and its integrated tools, including neural

Breast cancer remains the most frequently diagnosed malignancy in women, and resistance to cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitors limits long-term treatment efficacy. This study aimed to identify long non-coding RNAs (lncRNAs) associated with predicted sensitivity to CDK4/6 inhibitors and to investigate their biological functions in breast cancer. Transcriptomic data from The Cancer Genome Atlas (TCGA) and drug sensitivity data from the Genomics of Drug Sensitivity in Cancer 2 (GDSC2) database were integrated, and drug sensitivity was predicted using the oncoPredict algorithm. BACKGROUND: Cyclin B2 (CCNB2), a member of the cyclin family, plays a crucial role in glycolysis and the progression of pancreatic ductal adenocarcinoma (PDAC). Lactate-dependent histone modifications, particularly histone lactylation, have emerged as novel markers associated with glycolysis. However, the specific mechanisms by which CCNB2 influences histone lactylation in PDAC remain unclear.
METHODS: We investigated the involvement of CCNB2 in PDAC growth and progression using glycolysis inhibitors in vivo and in vitro. The expression levels of CCNB2 in PDAC were assessed through bioinforma

'Superenhanced' transcription of oncogenes by aberrant looping of upstream enhancer elements to transcriptional regulatory regions is a mechanism of oncogene overexpression. Non-selective cyclin-dependent kinase inhibitors (CDKi) that target transcriptionally regulatory CDKs, including CDK7, 9, 12 and 13, reduce mRNA levels of super-enhanced oncogenes and have activity against thyroid cancer cells. We hypothesized that more specific inhibitors of CDKs would have differential activities in thyroid cancer cells and may be suitable for further studies. We selected thyroid cancer cell lines with a

INTRODUCTION: Although morphologic assessment carries utmost importance in differentiating malignant mesotheliomas (MM) from reactive mesothelial proliferations (RMP), sometimes it may fail to clearly differentiate between these two entities. The aim of this study is to evaluate the potential role of cyclin D1 immunohistochemistry in this differentiation.
MATERIAL AND METHODS: Eighty cases (40 MM, 40 RMP) were examined. Nuclear staining of the mesothelial cells was evaluated after cyclin D1 immunostaining. Immunostaining was scored according to the percentage of immunopositive cells (0 %, 1-2

BACKGROUND/AIM: The traditional melanoma staging system is not ideal for predicting patients' individual risk of disease recurrence and death. Subsequently, suboptimal adjuvant treatment may be offered. Hence, identification of biomarkers to optimize risk stratification is warranted. Cyclins and cyclin-dependent kinase inhibitors, key players in the cell cycle regulation, are candidate prognostic biomarkers. Herein, their expression and prognostic value in melanoma are studied.
PATIENTS AND METHODS: The expression of Cyclin D1, Cyclin E, p16, p21, and p27 were assessed using immunohistochemis…

BACKGROUND: Multiple CDK4/6 inhibitors have been approved for the treatment of HR + /HER2- advanced breast cancer. Nevertheless, there is currently a scarcity of safety reports on CDK4/6 inhibitors within large sample cohorts.
METHODS: We employed a disproportionality analysis of the FAERS database to detect safety signals for the three marketed CDK4/6 inhibitors (palbociclib, abemaciclib, and ribociclib). We retrieved pertinent reports from 2004 Q1 to 2023 Q3. Four asymmetric analyses were utilized to assess signals.
RESULTS: A total of 459 positive signals were obtained at the preferred te

BACKGROUND: Neutrophilic inflammation, characterized by dysregulated neutrophil activation, triggers a variety of inflammatory responses such as chemotactic infiltration, oxidative bursts, degranulation, neutrophil extracellular traps (NETs) formation, and delayed turnover. This type of inflammation is pivotal in the pathogenesis of acute respiratory distress syndrome (ARDS) and psoriasis. Despite current treatments, managing neutrophil-associated inflammatory symptoms remains a significant challenge.
AIM OF REVIEW: This review emphasizes the role of cyclin-dependent kinases (CDKs) in neutrop

Splenic marginal zone lymphoma (SMZL) is one of the most common B-cell lymphomas that affect the spleen. We report a case with splenomegaly and lymphocytosis that showed a clonal B-cell population lacking CD5 and CD10 expression. Notably, the atypical lymphoid cells showed prolymphocytoid morphology and expressed cyclin D1. Fluorescence in-situ hybridization was negative for CCND1/IgH rearrangement. The prolymphocytoid morphology and cyclin D1 expression present a diagnostic pitfall. The clinical presentation, morphology, immunophenotype, and molecular genetic findings are most consistent with

Mutations in CSA and CSB proteins cause Cockayne syndrome, a rare genetic neurodevelopment disorder. Alongside their demonstrated roles in DNA repair and transcription, these two proteins have recently been discovered to regulate cytokinesis, the final stage of the cell division. This last finding allowed, for the first time, to highlight an extranuclear localization of CS proteins, beyond the one already known at mitochondria. In this study, we demonstrated an additional role for CSA protein being recruited at centrosomes in a strictly determined step of mitosis, which ranges from pro-metapha

Cyclin D1 is a regulatory subunit of -Cyclin Dependent Kinases 4 and 6 (CDK4/6) and regulates progression from G1 to S phase of the cell cycle. Dysregulated cyclin D1-CDK4/6 contributes to abnormal cell proliferation and tumor development. Phosphorylation of threonine 286 of cyclin D1 is necessary for ubiquitin-dependent degradation. Non-phosphorylatable cyclin D1 mutants are stabilized and concentrated in the nucleus, contributing to genomic instability and tumor development. Studies investigating the tumor-promoting functions of cyclin D1 mutants have focused on the use of artificial promote

BACKGROUND: Tearing of inner layer of aorta causes aortic dissection (AD), a severe disease with high morbidity and mortality. The pathological development of AD partially results from apoptotic death of aortic endothelial cells (AECs), the trigger and the molecular regulation of which remain largely unknown. Cyclin-dependent kinase 5 (CDK5) was initially detected in the brain as a proline-directed serine/threonine protein kinase regulating neuronal cell cycle re-entry and arrest. The abnormal expression of CDK5 leads to apoptotic cell death following abortive cell cycle re-entry in some neuro

The INK4 family is an important family of cyclin-dependent kinase inhibitors (CDKIs) and consists of CDKN2A, CDKN2B, CDKN2, and CDKN2D. Abnormal expression of CDKN2A has been reported in hepatocellular carcinoma (HCC) and is associated with the prognosis of patients and infiltration of immune cells. However, there is a lack of systematic research on the roles of the other INK4 family members in the diagnosis, prognosis, and immune regulation of HCC. Using online public databases and clinical samples, we comprehensively analyzed the INK4 family in HCC. All four INK4 proteins were overexpressed

In the adult brain, new neurons are constitutively derived from postnatal neural stem cells/progenitors located in two neurogenic regions: the subventricular zone (SVZ) of the lateral ventricles (migrating and differentiating into different subtypes of the inhibitory interneurons of the olfactory bulbs), and the subgranular layer of the hippocampal dentate gyrus. Cyclin D2 knockout (cD2-KO) mice exhibit reduced numbers of new hippocampal neurons; however, the proliferation deficiency and the dysregulation of adult neurogenesis in the SVZ required further investigation. In this report, we chara

Adult mammals have limited potential for cardiac regeneration after injury. In contrast, neonatal mouse heart, up to 7 days post birth, can completely regenerate after injury. Therefore, identifying the key factors promoting the proliferation of endogenous cardiomyocytes (CMs) is a critical step in the development of cardiac regeneration therapies. In our previous study, we predicted that mitogen-activated protein kinase (MAPK) interacting serine/threonine-protein kinase 2 (MNK2) has the potential of promoting regeneration by using phosphoproteomics and iGPS algorithm. Here, we aimed to clarif

Acute megakaryoblastic leukaemia (AMKL) is characterized by expansion of megakaryoblasts, which are hyper-proliferative cells that fail to undergo differentiation. Insight to the cell-cycle regulation revealed important events in early or late megakaryocytes (MKs) maturation; the cyclin-dependent kinases 4 and 6 (CDK4/6) have been reported to participate in the development of progenitor megakaryocytes, mainly by promoting cell cycle progression and DNA polyploidization. However, it remains unclear whether the continuous proliferation, but not differentiation, of megakaryoblasts is related to a
