Direct and indirect defences induced by piercing-sucking and chewing herbivores in Medicago truncatula.

Direct and indirect defences against feeding induced by chewing (Spodoptera littoralis) and piercing-sucking (Tetranychus urticae) herbivores, as well as components of signal transduction, were investigated in the model legume Medicago truncatula. Emitted volatiles, representing a mechanism of indirect defence, were measured and identified by gas chromatography/mass spectrometry (GC-MS). As elements of direct defence, the accumulation of phenolic compounds and of reactive oxygen species (ROS) was assessed using microscopic techniques. Jasmonic acid (JA) and salicylic acid (SA) concentrations w

Colostrum from healthy Brazilian women inhibits adhesion and contains IgA antibodies reactive with Shiga toxin-producing Escherichia coli.

UNLABELLED: Although Shiga toxin-producing Escherichia coli (STEC) has been isolated in Brazil, severe manifestations of the infection, such as haemorrhagic colitis and haemolytic-uraemic syndrome, are extremely rare in our population. Enteropathogenic Escherichia coli (EPEC) is the main aetiological agent of acute infantile diarrhoea in Brazil. There are many similarities between STEC and EPEC, such as the ability to produce attaching and effacing (A/E) lesions and some virulence-associated factors. Our aim was to investigate the presence of anti-STEC antibodies in healthy people living in an

The cyclopentenone prostaglandin 15-deoxy-Delta(12,14)-prostaglandin J(2) attenuates the development of acute and chronic inflammation.

Peroxisome proliferator-activated receptors (PPARs) are members of the nuclear hormone receptor superfamily of ligand-activated transcription factors that are related to retinoid, steroid, and thyroid hormone receptors. The PPAR-gamma receptor subtype seems to play a pivotal role in the regulation of cellular proliferation and inflammation. Recent evidence also suggests that the cyclopentenone prostaglandin (PG) 15-deoxyDelta(12,14)-PGJ(2) (15d-PGJ(2)), which is a metabolite of prostaglandin D(2), functions as an endogenous ligand for PPAR-gamma. We postulated that 15d-PGJ(2) would attenuate i

Antibiotic activity of an isocyanide metabolite of Trichoderma hamatum against rumen bacteria.

A metabolite of Trichoderma hamatum, 3-(3-isocyanocyclopent-2-enylidene)propionic acid, was tested for its effects on growth of and carbohydrate metabolism in 11 strains of functionally important rumen bacteria. To standardize the biological activity of this unstable metabolite, a rapid, aerobic disc diffusion assay was developed using Escherichia coli ATCC 11775. In an anaerobic broth dilution assay using a medium lacking rumen fluid and containing a soluble carbohydrate, the minimum inhibitory concentration of the metabolite which completely inhibited growth of the rumen bacteria for 18 h at

Neddylation-dependent CUL3-KLHL12 E3 ligase drives microglial oxidative stress and neuroinflammation in traumatic brain injury by targeting GCLM for degradation.

BACKGROUND: Microglia-mediated neuroinflammation and oxidative stress are pivotal drivers of secondary injury following traumatic brain injury (TBI). While neddylation governs essential cellular functions, its specific contribution to microglial activation and TBI pathology remains poorly understood. METHODS: We integrated bulk microglial RNA sequencing profiles with single-cell RNA sequencing (scRNA-seq) datasets from TBI mouse brains. To assess therapeutic potential, we employed a controlled cortical impact mouse model and treated animals with the neddylation inhibitor MLN4924. The role of

Functional analysis of NtWRKY70 regulating tobacco defense against Rhizoctonia solani AG3-TB.

Rhizoctonia solani AG3-TB is a destructive necrotrophic pathogen causing tobacco target spot disease. WRKY70 acts as a key regulator integrating salicylic acid (SA) and jasmonic acid (JA) signals in plant immunity. Our previous study found that NtWRKY70 in Nicotiana tabacum L. is significantly induced by R. solani AG3-TB infection; however, its functional mechanism remains elusive. In this study, bioinformatics analysis showed that NtWRKY70 is a nuclear localized protein with conserved domains and transcriptional autoactivation activity. Further investigations using virus-induced gene silencin

Development of a Brain-Penetrant Nurr1 Agonist Tool.

The ligand-sensing transcription factor nuclear receptor-related 1 (Nurr1), is thought to mediate neuroprotective activity and is implicated in various neurodegenerative diseases. Nurr1 ligands have been developed as tools to capture the receptor's potential in vitro and in vivo, but a dedicated brain-targeting agonist has been lacking. Here, we employed the scaffold of the dihydroorotate dehydrogenase (DHODH) inhibitor and strong Nurr1 activator vidofludimus to develop a descendant with a high brain-to-blood ratio. Scaffold-hopping from cyclopentene to thiophene, enhanced fluorination, and re

Evaluation of Carob and Pomegranate Extracts as Inducers of Resistance Against Verticillium Wilt in Olive Tree.

Pomegranate and carob extracts have been characterized as potential bioprotectors against Verticillium wilt of olive caused by Verticillium dahliae, suggesting they may act as host resistance inducers. This study aimed to elucidate whether these extracts activate metabolic pathways involved in olive defense against V. dahliae infection through marker gene expression analysis. The relative expression of seven resistance-related marker genes was evaluated by RT-qPCR in noninoculated and treated olive plants. Subsequently, the effect of the extracts as inducers of resistance against Verticillium

ABA Signaling De-Repression Coordinates With Cytokinin to Drive Large Petal Formation in Brassica napus.

Petal size is a critical agronomic trait influencing pollination efficiency and yield in field crops. However, the cellular basis and upstream hormonal interplay governing petal size in Brassica napus remain unclear. In this study, utilizing a natural variation population of "Zhongshuang 11", we investigated biological mechanisms underlying petal development through integrated phenotypic screening, paraffin section-based cytological analysis, targeted hormone metabolomics, and transcriptome. The results revealed that the single-flower area of large petals (BEP; 13.0 mm2) was significantly larg

A herbivore-inducible CYP82 catalyzes DMNT formation in Idesia polycarpa leaves.

BACKGROUND: Herbivore-induced volatile organic compounds (HIPVs) are important components of plant defense, but the molecular basis of their biosynthesis in woody plants remains poorly understood. Among HIPVs, the homoterpenes (3E)-4,8-dimethyl-1,3,7-nonatriene (DMNT) and (E,E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene (TMTT) are well-characterized herbivore-induced volatiles in herbaceous plants, whereas their biosynthesis in woody species remains largely unexplored. Here, we characterized herbivore-induced volatile emissions from Japanese orange cherry (Idesia polycarpa) leaves following fee

Crosstalk Between Inflammation and Cellular Senescence in Osteoarthritis: Modulatory Actions of Olive Oil and Its Bioactive Compounds.

Osteoarthritis is a degenerative joint disorder characterized by progressive cartilage degradation, subchondral bone alterations, and persistent low-grade inflammation. Recent findings have identified a strong interplay between inflammation and cellular senescence in osteoarthritis. Chronic inflammatory signals promote the accumulation of senescent cells, while senescent cells amplify inflammatory pathways through their senescence-associated secretory phenotype (SASP). This bidirectional loop accelerates tissue damage by perpetuating oxidative stress, matrix degradation, and the release of pro

Occurrence of influenza antivirals and resistance development in influenza A viruses in aquatic environments: A risk assessment.

Influenza antivirals (IAs) have been detected in aquatic environments inhabited by dabbling ducks, the natural reservoir of influenza A virus (IAV), raising concerns about the development of antiviral resistance. Because novel human IAV strains often contain genetic material of avian origin, this may contribute to resistance in viruses with pandemic potential. This study aimed to assess the environmental risk posed by four IAs-oseltamivir carboxylate (OC), zanamivir (ZA), peramivir (PE), and amantadine (AM)-based on their potential for environmental release, environmental stability, and induct

Methyl jasmonate promotes host defense by activating autophagy during Mycobacterium tuberculosis infection.

BACKGROUND: Jasmonates are a class of plant hormones that play multiple roles in plant growth, development, stress responses, and antimicrobial activity. However, their role in antimicrobial responses in mammalian cells and in vivo, particularly against Mycobacterium tuberculosis (Mtb), remains unclear. Tuberculosis remains a global challenge in human health, particularly with the rise of multidrug-resistant strains. Thus, exploring novel therapeutic agents is urgently warranted. In this study, we investigated the antimicrobial effects of methyl jasmonate (MeJA) against Mtb to evaluate its pot

Synergistic effects of pre-harvest drought and shade on flavor and aroma enhancement in fermented tea: Insights into JA/ABA signaling and metabolic regulation.

Drought stress during summer significantly impairs tea quality. This study is the first to systematically investigate the combined effects of pre-harvest drought and shade treatments on flavor and aroma compounds' spatial and temporal regulation during black and white tea processing. Through physiological and biochemical analyses, HPLC, electronic tongue analysis, and HS-SPME-GC-MS, we found that drought+shade (D + S) treatment significantly increases free amino acids and theaflavin levels, thereby enhancing umami, aftertaste, and richness. Using the OAV method, we identified 18 key volatiles,

Genome-wide insights into the nomenclature, evolution and expression of tobacco TIFY/JAZ genes.

A systematic nomenclature for tobacco TIFY/JAZ proteins was established via genome-wide analysis, and the gene transcription patterns and potential functions of these proteins were analyzed as well. Intensive studies focused on the plant-specific JAZ regulators of jasmonate (JA) signaling in tobacco due to their critical roles in regulating JA-mediated development, secondary metabolism, and stress responses. JAZs comprise a subfamily of the TIFY proteins, yet the reported TIFY/JAZ regulators of tobacco spp. are tangled in naming confusion, which resulted in nomenclature chaos. Here, we identif

Revealing the molecular mechanism of biosynthesis and transcriptional regulation of PAs, caffeine and linalool globally under simulative stress in coffee plants.

Coffee has become one of the most popular beverages worldwide due to the variety of bioactive compounds, which also play crucial roles against biotic and abiotic stresses. However, little is known about how these defensive compounds are produced in coffee. Here, we found that the whole biosynthetic pathways and the production of caffeine and proanthocyanidins (PAs) were promoted under Methyl Jasmonate (MeJA) treatment. Co-expression data showed that some transcription factors were shared by caffeine and PA regulation, and further several candidate caffeine regulators were identified. The biosy

Induction of systemic resistance through calcium signaling in Arabidopsis exposed to air plasma-generated dinitrogen pentoxide.

Plasma technology, which can instantaneously transform air molecules into reactive species stimulating plants, potentially contributes to developing a sustainable agricultural system with high productivity and low environmental impact. In fact, plant immunity activation by exposure to a reactive gas mainly consisting of dinitrogen pentoxide (N2O5) was recently discovered, while physiological responses to N2O5 are rarely known. Here, we demonstrate early (within 10 min) physiological responses to N2O5 gas in Arabidopsis. Exposure to N2O5 gas induced an increase in cytosolic Ca2+ concentration w

CDP-DAG synthases regulate plant growth and broad-spectrum disease resistance.

Phosphatidic acid (PA) functions as a cell membrane component and signaling molecule in plants. PA metabolism has multiple routes, in one of which PA is converted into cytidine diphosphate diacylglycerol (CDP-DAG) by CDP-DAG synthases (CDSs). CDS genes are highly conserved in plants. Here, we found that knock-down of the CDS gene enhanced the resistance of Arabidopsis thaliana to multiple pathogens, with a growth penalty. When Arabidopsis leaves were treated with chitin or flg22, reactive oxygen species (ROS) production in cds mutants was significantly higher than that in the wild-type (WT). S
