Ultrafast deactivation of bilirubin: dark intermediates and two-photon isomerization.

Bilirubin is a neurotoxic product responsible for neonatal jaundice, which is generally treated by phototherapy. The photoreaction involves ultrafast internal conversion via an elusive intermediate and Z-E isomerization with minor yield (less than 3% in solution). The structure of the intermediate remains unclear. Here, the combination of UV-vis and mid-IR ultrafast transient absorption spectroscopy reports a comprehensive picture of the mechanism and provides essential structural information about the intermediate species. Thus, spectral dynamics during the earliest ps unveils a wavepacket tr

The Dark Side of the Force - Constraints and Complications of Cell Therapies for Stroke.

Cell therapies are increasingly recognized as a promising option to augment the limited therapeutic arsenal available to fight ischemic stroke. During the last two decades, cumulating preclinical evidence has indicated a substantial efficacy for most cell treatment paradigms and first clinical trials are currently underway to assess safety and feasibility in patients. However, the strong and still unmet demand for novel stroke treatment options and exciting findings reported from experimental studies may have drawn our attention away from potential side effects related to cell therapies and th

The bipolarity of light and dark: A review on Bipolar Disorder and circadian cycles.

BACKGROUND: Bipolar Disorder is characterized by episodes running the full mood spectrum, from mania to depression. Between mood episodes, residual symptoms remain, as sleep alterations, circadian cycle disturbances, emotional deregulation, cognitive impairment and increased risk for comorbidities. The present review intends to reflect about the most recent and relevant information concerning the biunivocal relation between bipolar disorder and circadian cycles. METHODS: It was conducted a literature search on PubMed database using the search terms "bipolar", "circadian", "melatonin", "cortis

Entrainment of the human circadian clock to the natural light-dark cycle.

The electric light is one of the most important human inventions. Sleep and other daily rhythms in physiology and behavior, however, evolved in the natural light-dark cycle [1], and electrical lighting is thought to have disrupted these rhythms. Yet how much the age of electrical lighting has altered the human circadian clock is unknown. Here we show that electrical lighting and the constructed environment is associated with reduced exposure to sunlight during the day, increased light exposure after sunset, and a delayed timing of the circadian clock as compared to a summer natural 14 hr 40 mi

Near-infrared spectroscopy: exposing the dark (venous) side of the circulation.

The safety of anesthesia has improved greatly in the past three decades. Standard perioperative monitoring, including pulse oximetry, has practically eliminated unrecognized arterial hypoxia as a cause for perioperative injury. However, most anesthesia-related cardiac arrests in children are now cardiovascular in origin, and standard monitoring is unable to detect many circulatory abnormalities. Near-infrared spectroscopy provides noninvasive continuous access to the venous side of regional circulations that can approximate organ-specific and global measures to facilitate the detection of circ

Photodynamic therapy for granuloma annulare: more than a shot in the dark.

BACKGROUND: Granuloma annulare (GA) is a benign granulomatous and inflammatory skin disorder. The pathogenesis remains enigmatic and convincingly effective treatment options are not available. Inspired by a report showing photodynamic therapy (PDT) to be effective in a single patient with GA, we sought to evaluate this benefit in a series of patients with GA. OBSERVATIONS: PDT was performed in 7 consecutive patients with histologically confirmed GA located at the extremities. First, 20% ALA gel was applied under an occlusive dressing for 5 h, followed by illumination with 100 J/cm(2) by a sta

Dark Winter and the spring of 1972: deflecting the social lessons of smallpox.

This article examines how the master status of bioterrorism has distracted professional and political attention from the social lessons of smallpox. I illustrate this by comparing an influential bioterrorism simulation known as Dark Winter with the social history surrounding the Yugoslavian smallpox epidemic of 1972. Dark Winter's epidemiological premises were largely based upon what was learned from the Yugoslavian outbreak. Yet, although this epidemic was non-deliberate, the exercise did not attend to the social conditions within which it developed. Most notably, it did not consider that thi

The application of chromatic dark-adapted kinetic perimetry to retinal diseases.

PURPOSE: To demonstrate the value of a 2-color perimetric procedure for determining cone and rod system contributions to the dark-adapted kinetic visual field (VF). DESIGN: Prospective evaluation of perimetric testing procedure. PARTICIPANTS: Five patients with retinal diseases and 6 visually normal individuals. METHODS: Long- and short-wavelength stimuli were presented under dark-adapted conditions in a Goldmann perimeter. Visual fields were measured for the II and V test target sizes with a long-wavelength filter (cut-on at 600 nm) and a short-wavelength filter (cutoff at 510 nm). Light i

A dark, firm dry-like condition in turkeys condemned for cyanosis.

A case-control study (n = 130) was conducted on toms condemned for cyanosis. Color (CIE L*a*b*), pH, and physical characteristics were measured on the Pectoralis major at slaughter and after 24 h. Meat from carcasses condemned for cyanosis had dark, firm, dry-like traits. It was darker and redder and showed higher water-holding capacity, lower cooking loss, and higher gel strength than did controls. Perimortem pH was negatively correlated with the lightness (L*) of meat at the time of slaughter (r = -0.58) and at 24 h postmortem (r = -0.64), positively correlated with water-holding capacity (r

Central leptin stimulates corticosterone secretion at the onset of the dark phase.

Leptin, a hormone secreted by adipose tissue in proportion to body adiposity, is proposed to be involved in the central nervous regulation of food intake and body weight. In addition, evidence is emerging that leptin regulates neuroendocrine and metabolic functions as well, presumably via its action in the central nervous system (CNS). To investigate this regulatory effect of leptin, we infused 3.5 microg of human leptin directly into the third cerebral ventricle (i3vt) of lean male Long-Evans rats, 90 min before the onset of their dark phase. Before and after infusion, blood samples were with

The adjustment of the melatonin rhythm of rats by 90-min dark pulses.

The pineal melatonin patterns were measured in rats kept 1, 2, 3, 4, 6, 8, 16, or 28 days under the lighting conditions with a dark period of 90 min only (from 18:00 to 19:30 h). The samples were collected at 30-min intervals, and melatonin was measured by radioimmunoassay. During the first and second dark pulse the melatonin levels were low, but thereafter they were significantly higher than the usual daytime levels. The increase of the synthesis started within 15 min after the light offset. The results suggest a stable adjustment of the melatonin rhythm by the dark pulses.

Bonded porcelain veneer masking of dark tetracycline dentinal stains.

Resin-bonded porcelain veneer restorations can attractively mask dark dentinal tooth discoloration caused by tetracycline staining. When placing these restorations, special attention must be given to tooth preparation, handling of the porcelain, and veneer thickness. When treating adolescent patients whose occlusal development is not as yet completed, there are additional special considerations. This case report describes the placement of 6 resin-bonded porcelain veneer restorations in an adolescent patient to conceal dark tetracycline staining. Emphasis is placed on proper tooth preparation a

Intramuscular streptomycin effect on dark cells of utricle in guinea pigs.

Streptomycin has been used in the treatment of Meniere's disease for almost half a century. Clinical studies showed that streptomycin may eliminate vertigo attacks and stabilize or improve hearing in the majority of patients. Animal experiments have demonstrated severe damage to the vestibular hair cells after streptomycin treatment. This study is to observe the effect of intramuscular streptomycin on dark cells of utricle. Guinea pigs that had received intramuscular injections of streptomycin sulfate 400 mg/kg per day for 1 and 2 weeks were studied. The dark cells of utricle were observed und

Abnormal dark adaptation and rhodopsin kinetics in Sorsby's fundus dystrophy.

Scotopic visual thresholds and time courses for dark adaptation were determined in eight patients with Sorsby's fundus dystrophy. Rhodopsin regeneration also was recorded in two. All patients had poor night vision and a visible yellow deposit at the level of Bruch's membrane that was confluent in the posterior pole. In retinal regions with the yellow deposit, scotopic thresholds were elevated, the rod-cone break was delayed or indistinct, the time courses for the rod portion of the dark adaptation curve was prolonged, and rhodopsin regeneration was slow in the one patient in whom measurements

Cone dystrophy and supernormal dark-adapted b-waves in the electroretinogram.

A male patient suffering from cone dystrophy was followed over 9 years. In addition to the typical clinical and electrophysiologic signs, supernormal b-waves were found in the dark-adapted electroretinogram. Our case is compared with 12 similar patients described in the literature. Our patient differed from the other patients in the following aspects: he was male and had a congenital stationary disease with a small pigment epithelial scar in the left eye only and no other fundus changes up to the age of 22 years. He did not complain of night blindness. The dark-adapted electroretinogram of our
