Diazepam (3 nM-3 microM) enhanced GABA binding to well-washed synaptosomal membranes in a concentration-dependent manner. Half maximal enhancement occurred at 20 nM diazepam. Kinetic analysis by non-linear Scatchard analysis revealed that the primary action of diazepam was to increase the affinity of GABA binding to a low affinity site. These results support electrophysiological and other biochemical observations of benzodiazepine-GABA interactions.

Using dissociated spiral ganglion cell cultures obtained from 3-day-old rat cochlea, we investigated the response of auditory neurones to gamma-aminobutyric acid (GABA) using patch-clamp techniques. OBJECTIVE: Catatonia is a neuropsychiatric disorder that can respond rapidly to treatment with benzodiazepines. Historically, lorazepam is the most used agent for "the lorazepam challenge test" (LCT), but comparative evidence for alternative benzodiazepines remains limited. A temporary intravenous (IV) lorazepam shortage within our health system created a natural experiment by restricting access to lorazepam. AIMS: Many patients with glioblastoma (GBM) suffer from comorbid neurological/psychiatric disorders and, therefore, are treated with psychopharmacological agents. Diazepam (DIA) is widely adopted to treat status epilepticus, alleviate anxiety, and inhibit chemotherapy-associated delayed emesis in GBM patients.
