Synaptic transmission from splanchnic nerves to the adrenal medulla of guinea-pigs.

1. Membrane potentials were recorded with conventional intracellular microelectrodes from chromaffin cells in isolated, bisected adrenal glands from guinea-pigs. 2. All cells were electrically excitable and responded to depolarizing current with all-or-nothing action potentials that were blocked by tetrodotoxin. 3. Input resistance was 180 +/- 14 M omega and this was lower than that reported for isolated chromaffin cells using patch electrodes. 4. All cells responded to transmural stimulation with action potentials that arose from excitatory synaptic potentials in response to the excitation of

Dual presynaptic effects of 5-hydroxytryptamine on peripheral noradrenergic synapses.

The aim of the present experiments was to study the effects of 5-hydroxytryptamine (5-HT) on the responses to postganglionic stimulation of two models of the peripheral sympathetic nervous system: the isolated nictitating membrane of the cat and the guinea-pig isolated atria. In the nictitating membrane of the cat, 5-HT (0.1 microM) shifted to the left the frequency-response curve to nerve stimulation. This potentiating effect was prevented by 5-HT receptor antagonists (0.1 microM methysergide, 0.1 microM pizotifen and 0.1 microM morphine) and also by the beta-adrenoreceptor blocker propranolo

Sensitization and habituation of the plantar cushion reflex in cats.

The plantar cushion reflex in cats was examined as a model system in a mammal for the study of the effects of repeated stimulation on neural transmission. Effects of various frequencies and intensities of stimulation were similar to those seen in other reflex systems. For instance, for a fixed number of stimuli, habituation of the plantar cushion reflex was more marked at 10 Hz than at 2.0 Hz, and with 1.0 X threshold stimulation than with 5.0 X threshold stimulation. Sensitization occurred at intermediate intensities and frequencies of stimulation. Dorsal root potentials were studied; changes

Disuse in the lateral geniculate nucleus of the cat.

1. An attempt has been made to produce disuse in the lateral geniculate nucleus (LGN) of the cat, in the synapses between the optic nerve fibres and the principal cells of the nucleus. Evidence is produced that destruction of the visual receptor cells by iodoacetate or 1,5-di(p-aminophenoxy) pentane dihydrochloride effectively silences the optic nerve discharge and so achieves this result.2. Disuse of the LGN synapses does not cause any decrease in synaptic efficiency. The LGN response to a single stimulus to the optic nerve was not appreciably altered, and the depression which normally follow

Impaired motor cortex plasticity in patients with Noonan syndrome.

OBJECTIVE: Noonan syndrome (NS; OMIM 163950) is a developmental disorder caused by activating mutations in various components of the RAS-MAPK pathway. Recent in vitro studies demonstrated impairment of synaptic plasticity caused by RAS-MAPK pathway hyperactivity. Induction of synaptic plasticity critically depends on the level of attention. We therefore studied the induction of synaptic plasticity in patients with NS and healthy volunteers under different conditions of attention using transcranial magnetic stimulation. METHODS: We investigated 10 patients with NS and healthy controls (HC) usi

Preexposure to drug administration context blocks the development of tolerance to sedative effects of diazepam.

The development of tolerance to benzodiazepine (BDZ) and other psychoactive agents such as morphine, alcohol, and barbiturates is thought to be a contingent or learning phenomenon. In a previous report, we demonstrated a positive correlation between the development of tolerance to the sedative effects of diazepam (DZ) and hippocampal synaptic plasticity. The results of the present work show that the development of tolerance to the hypolocomotor action of DZ (5 mg/kg) for 4 days and the associated increase in synaptic plasticity are context specific. Because animal preexposure to the drug admin

Spatiotemporal distribution of intracellular calcium transients during epileptiform activity in guinea pig hippocampal slices.

Calcium ions are known to play an important role in epileptogenesis. Although there is clear evidence for increased neuronal calcium influx during epileptiform potentials, direct measurements of the corresponding intracellular calcium transients are rare and the origin of calcium influx is not known. Therefore the spatial and temporal distribution of intracellular calcium transients during epileptiform activity in guinea pig hippocampal slices was monitored with the use of the indicator Calcium-Green and a fast optical recording method. Two models of epilepsy (bicuculline and low Mg2+) were co

pH transients evoked by excitatory synaptic transmission are increased by inhibition of extracellular carbonic anhydrase.

Excitatory synaptic transmission has been associated with a rapid alkalinization of the brain extracellular space. These pH shifts are markedly increased by acetazolamide, an inhibitor of carbonic anhydrase. Although this effect can be readily explained by inhibition of extracellular carbonic anhydrase, this enzyme has been considered strictly intracellular in the central nervous system. To determine whether these alkaline shifts are regulated by extracellular carbonic anhydrase, we studied the effects of a membrane impermeant, dextran-bound inhibitor of this enzyme. Extracellular alkaline tra

An electrophysiological study on the common antitussive drug dextromethorphan in hippocampal slices in the rat.

Dextromethorphan, a common antitussive drug derived from the opioid class of morphinans, has been reported to have antiepileptic properties in some experimental tests. The effects of dextromethorphan were tested on the synaptic responses of the interconnections between Schaffer collaterals and CA1 pyramidal cells in hippocampal slices of the rat. Dextromethorphan, up to 100-200 microM, did not affect the basal field potentials resulting from electrical stimulation of the radiatum (0.1 Hz, 100-200 microA, 75 mu sec) of CA1 pyramidal cells. Larger concentrations of the drug (300-400 microM) depr

Muscle reinnervation--I. Restoration of transmitter release mechanisms.

Following sciatic nerve crush the restoration of neuromuscular transmission in the extensor digitorum longus muscle of rat proceeds in a well defined manner: (a) as soon as the nerve-muscle contact is reformed, a subthreshold end-plate potential is recorded; no 'non-transmitting stage' is observed; (b) 24 hours later muscle action potentials are induced by nerve stimulation; (c) miniature end-plate potentials are absent or very rare at the newly reinnervated end-plates; their frequency returns to normal in about 4 weeks; (d) the frequency is also very much reduced in 30 mM K+ and hypertonic so

Postnatal maturation of forelimb motor cortex and its impact on behavior in rats.

The motor cortex expands postnatally in several species; however, the timeline of body representation development and its impact on hand movements remain unclear. We investigated the relationship between forelimb motor cortex maturation and forelimb behavior in 50 male Wistar rats aged 4 to 8 weeks. Intracortical microstimulation (ICMS) was used to examine motor representations in the motor cortex, and motor-evoked potentials (MEPs) were recorded from the wrist dorsiflexors. Grip strength and kinesiological features of pellet-reaching tasks were assessed weekly, and wrist dorsiflexor morpholog

The metabolite p-cresol impairs dendritic development, synaptogenesis, and synapse function in hippocampal neurons: Implications for autism spectrum disorder.

Autism spectrum disorder (ASD) is a heterogeneous neurodevelopment disorder resulting from different etiological factors, both genetic and/or environmental. These factors can lead to abnormal neuronal development on dendrite and synaptic function at the central nervous system. Recent studies have shown that a subset of ASD patients display increased circulation levels of the tyrosine metabolite, p-cresol, related to chronic intestinal disorders because of dysbiosis of the intestinal microbiota. In particular, abnormal presence of intestinal Clostridium sp. has been linked to high levels of p-c

Electrically Reconfigurable Floating Gate Optoelectronic Synaptic Pixels for In-sensor Convolutional Image Feature Extraction with Built-in Contrast Enhancement.

Image feature extraction and enhancement are fundamental operations in real-time object detection using convolutional neural networks (CNNs). In conventional architectures, continuous data transfer between sensors, memory, and processing units leads to high energy consumption and latency. In-pixel computing using optoelectronic synaptic (OS) devices offers a promising solution by enabling sensing and computation within the same hardware. However, most OS studies remain primarily device-centric and lack circuit-level considerations necessary for scalable system integration. Here, we present a t

High-order dynamics in an ultra-adaptive neuromorphic vision device.

Neuromorphic hardware for artificial general vision intelligence holds the potential to match and surpass biological visual systems by processing complex visual dynamics with high adaptability and efficiency. However, current implementations rely on multiple complementary metal-oxide-semiconductor or neuromorphic elements, leading to significant area and power inefficiencies and system complexity. This is owing to a key challenge that no single electronic device, to our knowledge, has yet been demonstrated that can integrate retina-like and cortex-like spiking and graded neuronal dynamics oper

Direct current stimulation-induced synaptic plasticity in the sensorimotor cortex: structure follows function.

BACKGROUND: Non-invasive direct current stimulation (DCS) of the brain induces functional plasticity in vitro and facilitates motor learning across species. The effect of DCS on structural synaptic plasticity is currently unknown. OBJECTIVE: This study addresses the effects and the underlying mechanisms of anodal DCS on structural plasticity and morphology of dendritic spines in the sensorimotor cortex (M1/S1). METHODS: A DCS electrode setup was combined with a chronic cranial window over M1/S1 in transgenic Thy1-GFP mice, to allow for in vivo 2-photon microscopy and simultaneous DCS. Contra

Direct current stimulation boosts hebbian plasticity in vitro.

BACKGROUND: There is evidence that transcranial direct current stimulation (tDCS) can improve learning performance. Arguably, this effect is related to long term potentiation (LTP), but the precise biophysical mechanisms remain unknown. HYPOTHESIS: We propose that direct current stimulation (DCS) causes small changes in postsynaptic membrane potential during ongoing endogenous synaptic activity. The altered voltage dynamics in the postsynaptic neuron then modify synaptic strength via the machinery of endogenous voltage-dependent Hebbian plasticity. This hypothesis predicts that DCS should exh

Blocking of Dendrodendritic Inhibition Unleashes Widely Spread Lateral Propagation of Odor-evoked Activity in the Mouse Olfactory Bulb.

The olfactory circuitry in mice involves a well-characterized, vertical receptor type-specific organization, but the localized inhibitory effect from granule cells on action potentials that propagate laterally in secondary dendrites of mitral cell remains open to debate. To understand the functional dynamics of the lateral (horizontal) circuits, we analyzed odor-induced signaling using transgenic mice expressing a genetically encoded Ca2+ indicator specifically in mitral/tufted and some juxtaglomerular cells. Optical imaging of the dorsal olfactory bulb (dOB) revealed specific patterns of glom
