Cyclin-dependent kinase 13 (CDK13) has emerged as a critical regulator of oncogenic metabolism, but its role in rewiring lipid metabolism in clear cell renal cell carcinoma (ccRCC) remains undefined. Here we identify CDK13 as a master orchestrator of lipid dysregulation in ccRCC, demonstrating that it drives de novo lipogenesis through a phosphorylation-dependent RNA N6-methyladenosine (m6A) modification axis. Clinically, CDK13 overexpression correlates with advanced tumor stage, poor prognosis and aberrant lipid accumulation in patient-derived ccRCC tissues. Mechanistically, CDK13 directly ph

Resistance to tyrosine kinase inhibitors (TKIs), particularly pazopanib, is the primary driver of treatment failure in patients with advanced clear cell renal cell carcinoma (ccRCC). However, the epitranscriptomic mechanisms underlying TKI resistance, especially the regulatory role of N6-methyladenosine (m6A) modification, remain poorly understood. We integrated multi-omics analyses with in vitro and in vivo functional assays, multi-cohort clinical validation, and development of targeted small-molecule inhibitors to dissect the role of methyltransferase-like 16 (METTL16) in ccRCC resistance to

SnoRNAs are regulatory RNAs that play indispensable roles in ribosomal RNA processing and translation. Their distinct structural conformations determine specific protein-binding partners, thereby mediating diverse epigenetic modifications. Most snoRNAs are transcribed from introns of snoRNA host genes (SNHGs). Processed snoRNAs can further yield piwi-interacting RNAs (piRNAs) and snoRNA-derived fragments (sdRNAs). These small RNA products are frequently dysregulated in tumors and exert significant oncogenic functions. In cancer, snoRNA dysregulation stems from DNA-level alterations such as chr

Drought poses a major threat to global food security, making it critical to understand the molecular mechanisms underlying plant responses to water scarcity. Epigenetic modifications, including DNA methylation and histone alterations, play central roles in regulating genes and hormonal pathways essential for drought adaptation. MicroRNAs, while primarily functioning as post-transcriptional regulators, can also influence epigenetic pathways and contribute to chromatin remodelling, suggesting a role in modulating epigenetic memory. Investigating these interactions is essential for understanding

Post-transcriptional modifications of RNA, particularly m6A methylation, are emerging as critical epigenetic regulators of neurodevelopment and behavior. Although the RNA demethylase, fat mass and obesity-associated (FTO) protein has been implicated in autism spectrum disorder (ASD), the precise mRNA targets and downstream molecular pathways through which it governs behavior remain unclear. Here, we identify a significant upregulation of FTO in the medial prefrontal cortex (mPFC) of BTBR mice, a well-established ASD model. Targeted knockdown of Fto in the mPFC successfully ameliorates core ASD

Rheumatoid arthritis (RA) is a systemic autoimmune disease that predominantly affects synovial joints, especially those of the hands, elbows, wrists, knees, and shoulders. RA frequently co-occurs with major depressive disorder (MDD), amplifying disease burden and complicating clinical outcomes. This study employed a multi-step integrative bioinformatics and structural biology framework to identify candidate molecular biomarkers for RA and MDD. Differential gene expression analysis and weighted gene co-expression network analysis (WGCNA) were performed on the epitranscriptomic dataset. These an

The RNA N6-methyladenosine (m6A) modification, catalyzed by methyltransferase-like 3 (METTL3), is a key epigenetic regulator of oral health and disease. This narrative review positions METTL3 as a dual-function master switch in oral biology. It promotes stem cell-driven regeneration but also drives disease progression. Physiologically, METTL3 enhances the odontogenic/osteogenic differentiation of dental pulp stem cells (DPSCs). It stabilizes key transcripts (e.g., lncSNHG7, GDF6, STC1) via m6A modification, activating Wnt/β-catenin signaling to foster dentinogenesis and pulp vitality-key goals

N6-methyladenosine (m6A) is a critical epitranscriptomic regulator of neuronal function. Cerebral ischemia induces m6A hypermethylation due to decreased expression of m6A demethylase fat mass and obesity-associated (FTO) protein. Previously, we showed that cerebral overexpression of FTO with an adeno-associated virus (AAV) 9 protects the post-stroke brain. We presently evaluated the mechanistic basis for FTO-dependent m6A demethylation in post-ischemic neuroprotection using the mice transient middle cerebral artery occlusion model of experimental stroke. Based on the bioinformatic predictions

SARS-CoV-2, the causative agent of COVID-19, manipulates host gene expression through multiple mechanisms, including disruption of RNA processing. Here, we identify a novel function of the viral nonstructural protein 14 (NSP14) in inducing N 7-methylguanosine (m 7 G) modification in the internal sequences of host mRNA. We demonstrate that NSP14 catalyzes the conversion of guanosine triphosphate (GTP) to m 7 GTP, which is subsequently incorporated into mRNA by RNA polymerase II, resulting in widespread internal m 7 G modification. This activity is dependent on NSP14's N 7-methyltransferase ( N

N6-methyladenosine (m6A) is the most abundant and reversible epitranscriptomic modification in eukaryotes, playing a pivotal role in regulating various RNA metabolic processes, including splicing, nuclear export, translation and degradation. Emerging evidence indicates that m6A modification is indispensable in biological processes of bone cells such as proliferation, differentiation and apoptosis. Given its pivotal influence on osteoblastogenesis and osteoclastogenesis, m6A modification, particularly via METTL3, has attracted considerable attention in osteoporosis (OP). In this review, we prob

tRNA modifications are central to bacterial translational control. Here, we integrated genetics, mass spectrometry, epitranscriptomics, and comparative genomics to map the tRNA modification genes of the Gram-positive pathogens Streptococcus mutans and Streptococcus pneumoniae. Both species show a marked loss of modifications dependent on Fe-S enzymes, consistent with a broader trend of Fe-S enzyme reduction in Streptococcus central metabolism. In addition, the D, m1A, m7G, t6A, and i6A modifications were mapped in S. pneumoniae tRNAs, and we confirmed that a unique DusB1 enzyme is responsible

Dysregulated RNA modifications, stemming from the aberrant expression and/or malfunction of RNA modification regulators operating through various pathways, play pivotal roles in driving the progression of haematological malignancies. Among RNA modifications, N6-methyladenosine (m6A) RNA modification, the most abundant internal mRNA modification, stands out as the most extensively studied modification. This prominence underscores the crucial role of the layer of epitranscriptomic regulation in controlling haematopoietic cell fate and therefore the development of haematological malignancies. Add

T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematological malignancy with a poor prognosis, urging for novel therapeutic targets and treatment strategies. N6-methyladenosine (m6A) is a crucial methylation modification that affects the pathogenesis of leukemia by regulating the mRNA of key genes. Interferon regulatory factor 8 (IRF8) is a crucial transcription factor for hematological lineage commitment, but its role in T-ALL is unclear. Here, IRF8 is shown to suppress T-ALL. The expression of IRF8 is abnormally silenced in patients with T-ALL. Knockout of Irf8 significantly ha

N6-methyladenosine (m6A) methylation is the most common form of RNA modification in eukaryotes and has been shown to act as an important epitranscriptomic marker, which can regulate gene expression, messenger RNA stability, alternative splicing and translation efficiency. Recent progress on determining insect m6A function has been limited to the dipteran Drosophila melanogaster, in which m6A is involved in neuronal functions and sex determination. Methylation and function of m6A in other insects, however, remain unknown. Here, we investigated a transcriptome-wide profile of m6A in the lepidopt
