Cyclofenil is a modulator of estrogen receptor and is used clinically as an ovulation-inducing drug with antiviral activity. However, the mechanism by which cyclofenil works without estrogen receptor remains unknown. In this study, we screened for a small-molecule drug targeting the NLRP3 inflammasome and found that cyclofenil inhibited the secretion of IL-1β in a dose-dependent manner. Intriguingly, cyclofenil impedes activation of the NLRP3 inflammasome, independent of estrogen receptor, without interfering with the priming stage. Additionally, cyclofenil blocks ASC oligomerization and pyrop

Adjuvant hormonal therapy is one of the most important treatments of hormone-receptor-positive breast cancer and includes selective estrogen receptor modulators, aromatase inhibitors, and luteinizing hormone-releasing hormone analogs. In patients receiving these drugs, a progressive recession of frontal-temporal hairlines is often observed, such as a certain grade of hair miniaturization in the same areas and the central scalp area, producing a pseudo-female androgenic alopecia, which has to be considered oncotherapy-induced alopecia. The aim of this work, is to describe the clinical aspects a…

The female genital and lower urinary tracts share a common embryological origin, arising from the urogenital sinus and both are sensitive to the effects of the female sex steroid hormones throughout life. Estrogen is known to have an important role in the function of the lower urinary tract and estrogen and progesterone receptors have been demonstrated in the vagina, urethra, bladder and pelvic floor musculature. In addition estrogen deficiency occurring following the menopause is known to cause atrophic change and may be associated with lower urinary tract symptoms such as frequency, urgency,

The endocrine pancreas has emerged as a target for estrogens. The functions of pancreatic α-, β- and δ-cells are modulated by the endogenous hormone, 17β-estradiol (E2). Low physiological concentrations (100pM-1nM) of E2 rapidly decrease the activity of the ATP-sensitive potassium channel (K(ATP)) and enhance glucose-induced insulin release in β-cells in an estrogen receptor β (ERβ)-dependent manner. In addition to the insulinotropic action of ERβ, the newly described estrogen receptor, GPR30, is involved in the insulinotropic effects of high doses of E2 (100nM-5μM). The specific GPR30 agonist

Neuroinflammation plays a dual role in the central nervous system, offering protection in acute phases but contributing to chronic damage in neurodegenerative diseases. Estrogen, traditionally recognized for its reproductive functions, exerts extensive neuroprotective effects by modulating neuroinflammatory processes across multiple levels. This review explores the actions of estrogen through its receptors in astrocytes, microglia, and neurons, emphasizing its regulation of signaling pathways such as PI3K/Akt, NF-κB, and WNT/β-catenin. Estrogen also enhances mitochondrial function, promotes DN

The authors present a review referring to cellular and biochemical control systems of bone remodeling as well as the effect of estrogens on modulation of bone metabolism. Systemic and local control factors such as PTH, calcitonin, vitamin D, prostaglandins, insulin-like growth factor-1, transforming growth factor-beta are analyzed as possible indirect targets for the estrogen induced inhibition of bone resorption. A direct receptor-linked action of estrogens on the osteoblast-lining cell unit, the most important modulator or bone metabolism, is furtherwise examined. The precise mechanism of th

The compounds that bind steroid hormone receptors including estrogen receptors (ERs), progesterone receptor (PR) or androgen receptor (AR), and induce or modulate a steroid hormone receptor-mediated response could be defined as endocrine disruptors (EDs). Currently, there are no standard methods to determine whether a chemical is an endocrine disruptor or not. Most results of in vitro and in vivo data are derived from assays that measure estrogenic activity, thus fewer data are available from assays that measure androgenic and progestogenic activities. In this review, we introduce a novel in v

Proteolysis by the 26S proteasome is an important regulatory mechanism that governs the protein stability of several steroid/nuclear receptors and that has been implicated in the control of receptor transcriptional activation function. Herein, we report that thyroid hormone can prevent estrogen-induced proteolysis of estrogen receptor-alpha (ERalpha) protein in lactotrope cells of the pituitary. The stabilization of ERalpha protein by thyroid hormone represents a selective blockade against estradiol-stimulated degradation, because thyroid hormone (but not glucocorticoid) can protect estrogen-a

The progesterone receptor (PR) occurs in two major forms, the full-length PRB and the amino-truncated PRA, which lacks 164 amino-terminal residues. PRB functions as a strong transcriptional activator of progesterone-responsive genes, whereas PRA is inactive in several cell types where it may even act as a trans-dominant repressor of PRB and other steroid receptors, like the glucocorticoid receptor or, reportedly, the estrogen receptor. We initially observed that a PR deleted of its entire amino domain (PR538-C) is incapable of trans-repressing PRB or glucocorticoid receptor, suggesting that a

INTRODUCTION: Since the phenotype and clinical need of the hypogonadal individuals changes dramatically over time, versatile therapies are needed.
AREAS COVERED: The authors review the available evidence on possible therapies.
EXPERT OPINION: In the case of primary hypogonadism starting early in life, substitution with testosterone (T) is the only choice. For secondary congenital hypogonadism, we recommend starting with gonadotrophins to allow the testes to reach pubertal size. Thereafter, T replacement therapy can be administered until fertility is desired. At that time, gonadotrophins shou

The aim of this study was to elucidate the possible direct regulatory role of the endocannabinoids in the modulation of LH secretion in rabbits, a reflex ovulator species. The cannabinoid receptor type 1 (CB1) was characterized by RT-PCR techniques in the anterior pituitary of intact and ovariectomized does treated with GnRH and primed with estrogen and CB1 antagonist, rimonabant. Cannabinoid receptor type 1 immune reaction was evidenced by immunohistochemistry in the cytoplasm of approximately 10% of the pituitary cells with a density of 8.5 ± 1.9 (per 0.01 mm(2)), both periodic acid-Schiff p

Breast cancer is the leading malignancy among women worldwide, with endocrine therapy (e.g., selective estrogen receptor modulators, aromatase inhibitors) serving as a cornerstone of treatment; although these therapies are effective against hormone-sensitive breast cancer, they alter patients' systemic hormone profiles, which may disrupt the balance of the ocular surface microbiota that maintains ocular homeostasis. This review systematically examines the mechanisms of mainstream breast cancer endocrine therapies, the dynamics of the ocular surface microbiota, the association between hormonal …

A small subset of lactotroph adenomas is resistant to dopamine agonists (DA) and can also demonstrate aggressive or even malignant behavior. The implicated mechanisms are not clearly defined. Management can be challenging and requires a multidisciplinary approach. In DA resistant prolactinomas, switching to another DA could be the first option to consider. Further strategies include surgery and radiotherapy used alone or in combination. In cases of aggressive or malignant prolactinomas, temozolomide could be offered. Immune checkpoint inhibitors have been also recently proposed as an alternati

PURPOSE OF REVIEW: This article reviews fibroids management in the perimenopausal period, and addresses future directions in care.
RECENT FINDINGS: Aromatase inhibitors, selective estrogen receptor modulators and antiprogestogens for medical management and minimally surgical techniques are promising treatments.
SUMMARY: The disease and the symptoms may persist in the peri and postmenopausal periods. The assumption that they will resolve with the onset of the menopause is too simplistic and not always valid. The number of perimenopausal women who wish to retain their uterus for reasons other …

Osteoporosis is a skeletal disorder characterized by compromised bone strength that predisposes a person to increased fracture risk. Fractures are often associated with increased morbidity, higher mortality, loss of function and even psychological consequences. Pharmacotherapeutic interventions (e.g., bisphosphonates, selective estrogen receptor modulators, calcitonin, and teriparatide) in women with postmenopausal osteoporosis provide substantial reduction in fracture risk over and above risk reduction with calcium and vitamin D supplementation alone. The importance of nutritional support alo…

Epidemiologic studies revealed that the prevalence of Parkinson disease is higher in males than in females and that the progression of the disease might be rapid in males compared with females. The reason for the gender difference is unknown; however, estrogens may be involved. Many studies have revealed that estrogens provide neuroprotective effects and that the protective mechanisms include antioxidant property and upregulation of Bcl-2, brain-derived neurotrophic factor, and glial cell-derived neurotrophic factor (GDNF). Upregulation of Bcl-2 or GDNF is mediated by nonnuclear estrogen recep

Recently, we demonstrated that melatonin secretion was increased in male patients with GnRH deficiency and decreased to normal levels during testosterone treatment. These data suggested that gonadal steroids modulate melatonin secretion, probably by activating specific receptors in the pineal gland. We used immunohistochemistry to localize gonadotropin (LH and FSH) and gonadal steroid (androgens and estrogens) receptors in human pineal glands. Tissues were obtained at autopsy from 25 males, aged 19-87 yr, and five prepubertal children, aged 0.2-10 yr. Positive staining for all four types of re

Maturation of sperm within the epididymis is a pre-requisite for fertilization in mammals. Epididymal function is controlled by a complex array of hormones and growth factors. While testosterone is the primary stimulus for epididymal development and sperm maturation, the importance of estrogen effects on efferent ductules has been increasingly recognized, and points to a need to clarify the role of estrogen receptor-mediated action in the epididymis. Estrogens modulate the expression of genes involved in fluid absorption in the efferent ductules and the epididymis. The present review highlight

The decline in estrogen levels in postmenopausal women is a major risk factor for the high prevalence of osteoporosis and vascular calcification in this population. Estrogen deficiency leads to an imbalance in the RANKL/OPG ratio, abnormalities in the Wnt/β-catenin pathway, and disruptions in bone morphogenetic protein (BMP) signaling, thereby inducing osteoporosis and vascular calcification. Moreover, estrogen deficiency affects calcium and phosphorus metabolism and induces the release of extracellular vesicles from the aging bone matrix, which mediate interactions between bone and vascular t
