Effects of Three Kinds of Carbohydrate Pharmaceutical Excipients-Fructose, Lactose and Arabic Gum on Intestinal Absorption of Gastrodin through Glucose Transport Pathway in Rats.

BACKGROUND: Some glucoside drugs can be transported via intestinal glucose transporters (IGTs), and the presence of carbohydrate excipients in pharmaceutical formulations may influence the absorption of them. This study, using gastrodin as probe drug, aimed to explore the effects of fructose, lactose, and arabic gum on intestinal drug absorption mediated by the glucose transport pathway. METHODS: The influence of fructose, lactose, and arabic gum on gastrodin absorption was assessed via pharmacokinetic experiments and single-pass intestinal perfusion. The expression of sodium-dependent glucos…

Material compatibility and processing challenges in droplet deposition modelling additive manufacturing: A study on pharmaceutical excipients Polyvinylpyrrolidone/vinyl acetate (PVP/VA) and Polycaprolactone (PCL).

Additive manufacturing (AM) enables the production of complex, lightweight, and customized components with superior quality. Selecting the r … brication. We investigated the printability and part quality in DDM using two common pharmaceutical excipients: Polyvinylpyrrolidone/vinyl acetate 6:4 (PVP/VA), which is highly brittle, and Polycaprolactone (PCL), known for its low solubility and role in controlled drug release. Different ratios of PVP/VA and PCL were compounded via hot melt extrusion (HME) …

Anisotropic excipient-free buccal wafer engineered by directional freezing for structure-guided neuro-immune therapy against chemotherapy-induced emesis.

The management of chemotherapy-induced nausea and vomiting (CINV) in dysphagic patients is limited by the absence of mucosal delivery platforms that combine ultrafast adhesion, sustained release, and excipient-free composition. To address this, we engineered an excipient-free, anisotropic buccal wafer (IJ-ODW) via directional freeze-drying. The material features an engineered bilayered architecture, a dense top stratum integrated with a lamellar porous substrate, which resolves the mucosal "speed-stability paradox" by enabling ultrafast disintegration (15 s) together with strong, hydrogen-bond

From Infection to Oncology: AI-Assisted Drug Repurposing Uncovers Anti-Infective and Excipient-Class Compounds as PPAR-α-Linked Anticancer Agents.

Drug repurposing offers an efficient strategy to identify novel anticancer agents from marketed compounds. We screened a library of ~1400 approved drugs using a PPARE-PPAR-α luciferase reporter assay to uncover modulators of lipid metabolism that induce cancer cell death without direct binding to PPARα. This primary screen identified 224 compounds inhibiting reporter activity by >50%, of which 26 demonstrated ≥50% cytotoxicity in KAIMRC1 breast cancer and HCT116 colorectal cancer cells at 10 µM after 48 h incubation. AI-based DrTarget analysis classified these hits, with Group 1 comprising fou

3D printed sex-specific medicines: Excipient-mediated modulation boosts systemic drug exposure by more than three-fold in male rats.

Excipients, historically regarded as inert, are now being recognised for their ability to actively modulate biological targets, including intestinal efflux transporters such as P-glycoprotein (P-gp). We have previously shown that polyethylene glycol (PEG) excipients can selectively enhance the oral bioavailability of P-gp substrate drugs, particularly in males. This study examined how 3D printed formulations containing PEG 2000 influence the pharmacokinetics of silodosin, a P-gp substrate drug used for the treatment of benign prostatic hyperplasia in ageing men, using male and female Wistar ra…

Hidden Drug-Excipient Interaction by HPLC-MS/MS Analysis: Polysorbate 80 Enhances Nimodipine Systemic Exposure Through Competitive Metabolic Inhibition.

Pharmaceutical excipients have traditionally been regarded as pharmacologically inert carriers, serving merely as vehicles for active pharmaceutical ingredients. However, this conventional paradigm is increasingly challenged by emerging evidence revealing clinically significant drug-excipient interactions that can profoundly influence therapeutic outcomes. This study investigates a previously unrecognized pharmacokinetic interaction between nimodipine, a dihydropyridine calcium channel blocker, and polysorbate 80, a commonly used solubilizing excipient in nimodipine formulations. A novel HPLC-…

Concoctive principles of detoxification and retention of the main toxicity of Tripterygium wilfordii and its anti-inflammatory efficacy by concocting with the medicinal excipient Spatholobi Caulis juice.

Tripterygium wilfordii (TW), which has severe hepatotoxicity, is commonly used as anti-rheumatism. Using the juice of auxiliary herbs in concocting poisonous herbs is a conventional method for toxicity reduction or efficacy enhancement. Traditional Chinese Pharmacy textbooks record that Spatholobi Caulis (SC) can alleviate the side effects caused by TW and also possesses excellent hepatoprotective effect. However, it is still unclear how the concoctive principles of hepatotoxicity reduction and anti-inflammatory efficacy retention of TW after being concocted with the medicinal excipient SC jui

DSC, FT-IR and NIR with Chemometric Assessment Using PCA and HCA for Estimation of the Chemical Stability of Oral Antidiabetic Drug Linagliptin in the Presence of Pharmaceutical Excipients.

Pharmaceutical excipients should not interact with active substances, however, in practice, they sometimes do it, affecting the efficacy, stability and safety of drugs. Thus, interactions between active substances and excipients are not desirable. For this reason, two component mixtures of oral antidiabetic drug linagliptin (LINA) with four excipients of different reactivity, i.e., lactose (LAC), mannitol (MAN), magnesium stearate (MGS) and polyvinylpyrrolidone (PVP), were prepared in a solid state. A high temperature and a high humidity of 60 °C and 70% RH, respectively, were applied as stres…

An animal's sex influences the effects of the excipient PEG 400 on the intestinal P-gp protein and mRNA levels, which has implications for oral drug absorption.

There is a growing body of evidence which suggests that formerly regarded "inert" pharmaceutical excipients have the potential to influence oral drug bioavailability. The solubilizing agent polyethylene glycol 400 (PEG 400), for instance, has a sex-specific effect on P-glycoprotein (P-gp)-mediated drug bioavailability. We hypothesized that such an effect could be via PEG-induced alteration of P-gp activity and/or expression to different extents in males and females. To test this hypothesis in vivo, we investigated the influence of orally administered PEG 400 on the protein content and mRNA exp…

Cholesterol-butyl-biguanide conjugate as a novel excipient enhances the efficacy of doxorubicin liposomes against triple-negative breast cancer by potentiating immunogenic cell death.

Triple-negative breast cancer (TNBC) is an aggressive malignancy with limited therapeutic options and poor prognosis. Although doxorubicin (DOX)-loaded liposomes are clinically used, their tumor-targeting efficiency and overall therapeutic efficacy remain suboptimal. To overcome these limitations, we designed and synthesized a novel cationic lipid excipient, cholesterol-butyl-biguanide (CBB), and formulated DOX-loaded CBB liposomes (Lip CBB@DOX) via active loading. Physicochemical characterization confirmed well-defined vesicular morphology, a positive surface charge, and pH-responsive drug re

Controlled Solvent Removal from Antiviral Drugs and Excipients in Solution Enables the Formation of Novel Combination Multi-Drug-Motifs in Pharmaceutical Powders Composed of Lopinavir, Ritonavir and Tenofovir.

Diverging physicochemical properties of HIV drug combinations are challenging to formulate as a single dosage form. We have found that 2-to-4 hydrophilic and hydrophobic HIV drugs in combination can be stabilized with lipid excipients under a controlled solvent removal process to form a novel pharmaceutical powder distinct from typical amorphous material. This discovery has enabled production of a drug combination nanoparticle (DcNP) powder composed of 3 HIV drugs-water-insoluble lopinavir (LogP = 4.7) and ritonavir (LogP = 5.6) and water-soluble tenofovir (LogP = -1.6).
